In Recognition of the Forgotten Generation D. L. MULCAHyl Pollen was long believed to serve primarily a single function, that of delivering male gametes to the egge A secondary and generally overlooked value of pollen is that it serves to block the transmission of many defective alleles and gene combinations into the next generation. This latter function comes about simply because pollen tubes carrying defective haploid genotypes frequently fail to complete growth through the entire length of the style. However, the beneficial consequences of this pollen selection are diluted by the fact that the same deleterious genotypes are often transmitted through the egg at strictly mendelian frequencies (Khush, 1973). Gene expression in the pollen might thus at least appear to be a phenomenon of trivial consequence. Indeed, Heslop-Harrison (1979) rightly termed the gametophytic portion of the angiosperm life cycle, the "forgotten generation." This neglect, however, came about despite subtle but constant indications that pollen is the site of intense gene activity and selection. For example, Mok and Peloquin (1975) demonstrated that relatively heterozygous diploid pollen shows heterotic characteristics whereas relatively homozygous diploid pOllen does not. This was proof positive that genes are expressed (that is, transcribed and translated) in the pollen. 1 Department of Botany, University of Massachusetts Amherst, MA 01003, USA viii However, the implications for pollen biology of even this recent and well known study were not widely recognized.

Pollen, the plant structure most widely used by humans, is a key structure in plant reproduction giving rise to fruits and seeds. Moreover, the biotechnological use of pollen is of great importance for plant breeders since it allows to obtain varieties with better utilization and yield. In the first part, the successive steps of pollen development in the anther from floral induction to pollen germination and fertilization are thoroughly examined; the second part is devoted to pollen behaviour in vitro.

The last decade has seen tremendous progress in our knowledge of the pollen development and gene expression on one hand and the characterization of pollen specific proteins on the other. In compiling the chapters for this volume, we have pragmatically categorized these basic developments in pollen molecular biology and biotechnology into two sections based on their applications in agricul ture and implications in medicine. Pollen developmental biology and gene expression: applications in agricul ture. Pollen development is an extremely complex process encompassing a series of biochemical, physiological and genetic events. At the basic level, sporophyt ically expressed genes may expound our knowledge of unique processes of cellular differentiation which ultimately give rise to a full-fledged organism. At the applied level, the studies on the pollen and male sporophyte-specific gene expression, and of promoters and transcription factors of relevant genes have the potential to manipulate the fertility in certain cash crops leading to agricultural biotechnology.

Our knowledge of pollen, the gold dust that carries the male germ line of flowering plants and is vital for sexual reproduction and seed formation, has 'come of age' with the publication of this book. Here, for the first time in a single volume, are all the ideas and techniques developed in the last two decades concerning the manipulation of pollen and pollen tubes in plant breeding and biotechnology. Pollen has never been an easy topic to come to grips with, with its variable and often inexplicable terminology that has made it a more difficult field in which to work. This book will remedy that, with its overview of pollen biology and pollen-pistil interactions that explains terms and concepts of the male function of pollen in a way that is readily understandable to the student and professional plant scientist and plant breeder.

Pollen grains are everywhere - in the air, in the water, in soil and in the food we eat. Pollen has direct relevance in agriculture, horticulture, plant breeding, crop improvement and biotechnology. They are further of use for montioring cytotoxic effects - by herbicides, pesticides and pollutants - testing for allergic reactions, and for basic studies on gene expression, research on differentiation and polarity. Detailed instructions of the standard techniques, which have all been tested and improvised by the authors, are given, such as collection and storage of pollen, pollen culture, germination, tests for viability, incompatibility and isolation of protoplasts. Introduced by an explanation of the principles involved, the step-by step protocols are complemented by personal notes and precautions, specifying the reagents used and various appendices on basic and specific requirements for laboratory exercises on pollen.

The subject of this volume is the reproductive biology of plants. A steadily growing interest in this field is the result of at least two factors, as pointed out with great foresight by one of the driving forces in the field, H.E Linskens (Linskens 1964): most of the food consumed by humans takes the form of plant reproductive parts, and molecular biology now provides pow erful tools for investigating and manipulating plant reproductive systems. Molecular biology and the allied discipline of biotechnology are solidly represented in the papers in this book. The editors of Angiosperm Pollen and Ovules believe that the chapters herein contain some of the most excit ing findings of contemporary biology, and hope that the readers of this book will share their enthusiasm. The editors express sincere and grateful thanks for help from Carla Frova, Enrico Pe, and especially to Giorgio Binelli, all of the University of Milan. Without these three tireless and enthusiastic individuals it would not have been possible to maintain the apparently effortless proceeding of this congress. We thank also the organizing committee and the organizations that generously provided financial support (both listed below). Finally, we extend thanks to M. Cresti, D. Charlesworth, D. Hess, E Hoekstra, R. Bruce Knox, J.P. Mascarenhas, M.E. Nasrallah, P.L. Pfahler, A. Snow, and M.T.M. Willemse for chairing sessions.
