Potassium ions K+ ions are vital in physiological processes, particularly with regard to the cardiovascular system. Modulators influence the channels through which these ions flow and the research into modulator drugs of these channels is the subject of rapidly-advancing research. This work is intended to be of use as a tool for those working in the pharmaceutical industry. Early chapters concentrate on the synthesis and structure-activity relationships of potassium channel modulators. These are followed by discussion on the application of these drugs to different target organs, such as the cardiovascular system and the airways. The book concludes with a broad overview of clinical experience and future prospects.; The authors show how potassium channel activators and potassium channel blockers each have a role to play as therapeutic agents for many disorders, including asthma and respiratory tract disorders, diseases of the central nervous system such as epilepsy, and vascular diseases such as angina, peripheral vascular disease and high blood pressure. This book should therefore also be of interest to those researching into this wide range of diseases.

Modulation of potassium channels plays a very important part in nerve cell function in the autonomic nervous system. Compounds are now in development which can open or close potassium channels and will have numerous therapeutic applications. The major areas of clinical relevance will be hypertension, asthma and irritable bladder syndrome. The book describes the pharmacology of the channels and strategies for the design of drugs which exploit them. It consists of 17 chapters by international experts in the field which are written at a high level and will be of interest to pharmacologists and those in related clinical research disciplines

Being the crucial components of living cells, ion channels are important targets of therapeutic agents. Historically, it has been challenging to develop drugs on this target class. A major issue with target based ion channel drug development is the identification of effective small chemical leads for medicinal chemistry optimization to the clinical candidate status. Thus enough attention has been paid to the study of structure and functions of ion channels and their potential inhibitors. The present book compiles important chapters authored by eminent workers in the field to cover important recent advances in the studies of the structure and functions of ion channels and their inhibitors, such as sodium ion, potassium ion, chloride ion, calcium ion channel inhibitors. The book may be of great use to the students and scientists working in the area of molecular biology, biochemistry, physiology, neurobiology, and medicinal chemistry.

Cellular neurobiology has been transformed in the past decade by new technologies and fundamental discoveries. One result is an enormous increase in our understanding of how ion channels function in nerve and muscle cells and a widening perspective on the role of ion channels in non-neuronal cell physiology and development. Patch clamp techniques now permit direct observation of the transitions between functional confor mations of individual ion channels in their native membrane. Recombinant DNA techniques are being used to determine the primary structure of ion channel proteins and to test hypotheses about channel conformations, sites of grating and modulation, and the basis of ion selectivity. At the same time, biochemical techniques have revealed intricate signalling systems in side cells, involving second messengers such as calcium, phospholipids and cyclic nucleotides, which interface with the external milieu through GTP binding proteins and regulate cell metabolism by altering protein phos phorylation. This panorama of second messenger systems has greatly increas ed our application for their potential role in regulating ion channel function. We now recognize that ion channels are much more complicated than we once thought, and more interesting. They are not simply isolated macro molecules in the membrane, gated directly by depolarization or trans mitter binding to open briefly at a fixed conductance and then close or inactivate. Instead, individual channels now appear to have many open and closed states that are regulated independently by voltage and transmitters.

The Textbook of Ion Channels is a set of three volumes that provides a wide-ranging reference source on ion channels for students, instructors and researchers. Ion channels are mem- brane proteins that control the electrical properties of neurons and cardiac cells; medi- ate the detection and response to sensory stimuli like light, sound, odor, and taste; and regulate the response to physical stimuli like temperature and pressure. In non-excitable tissues, ion channels are instrumental for the regulation of basic salt balance that is critical for homeostasis. Ion channels are located at the surface membrane of cells, giving them the unique ability to communicate with the environment, as well as the membrane of intracellular organelles, allowing them to regulate internal homeostasis. Ion channels are fundamentally important for human health and diseases, and are important targets for pharmaceuticals in mental illness, heart disease, anesthesia, pain and other clinical appli- cations. The modern methods used in their study are powerful and diverse, ranging from single ion-channel measurement techniques to models of ion channel diseases in animals, and human clinical trials for ion channel drugs. Volume III includes coverage of key ion channel regulators and their mechanisms, the role of ion channels working in concert in selected physiological systems, and examples of ion channel mutations and dysfunction in a selection of diseases. Chapters on ion channel regulation include splice variants, calcium–calmodulin regulation, regulation by G pro- teins, and lipids. A selection of ion channels in physiological systems includes ion chan- nels of the heart, ion channels in immune cells and their role in pancreatic beta cells and regulation of insulin secretion, and the role of channels in sperm and eggs. While disease mechanisms are integrated into the chapters of Volume II, Volume III offers special consid- eration of ion channels in epilepsy, cystic fibrosis and pain syndromes. All three volumes give the reader an introduction to fundamental concepts needed to understand the mechanism of ion channels, a guide to the technical aspects of ion channel research, offer a modern guide to the properties of major ion channel families, and includecoverage of key examples of regulatory, physiological, and disease roles for ion channels.

Ion channels are crucial components of living cells. Situated in the cell's membranes. they allow particular ions to pass from one side of the membrane to the other. In recent years the patch clamp technique has allowed the activity of individual channels to be measured, and recombinant DNA technology has led to fascinating detail on their structure. Together, these technical advances have produced a great flowering of knowledge and understanding about the subject, itself leading to further breakthroughs in science and medicine. Ion Channels provides an introduction to this scientific endeavour. It emphasises the molecular structure of channels as determined by gene cloning technology. This knowledge illuminates discussions of the permeability and selectivity of channels, their gating and modulation, their responses to drugs and toxins and the human diseases caused when they do not function properly.
