This manual is designed to provide a detailed and practical guide for students, researchers, and practitioners involved in the study of biochemistry, molecular biology, and plant tissue culture. The topics covered herein are fundamental to the understanding and application of laboratory techniques and processes used in a variety of biological and biochemical studies. The manual starts with the preparation of solutions, pH adjustment, and the use of buffers essential skills in any biological laboratory. It then progresses through qualitative tests for carbohydrates and amino acids, quantitative estimations of glucose and proteins, and titration methods for amino acids and lipids, providing a comprehensive overview of common biochemical assays. These methods are critical for gaining insights into the molecular composition and behavior of biological samples. Special focus is placed on enzyme kinetics and how factors such as pH, temperature, and substrate concentration influence enzyme activity concepts that are vital in both research and applied biochemistry. Additionally, techniques like paper chromatography and thin-layer chromatography (TLC) for separating amino acids and monosaccharides are explored, giving readers practical skills for analyzing and identifying complex biomolecules. The manual also addresses the increasingly important field of plant biotechnology, introducing sterilization techniques, tissue culture media composition, and the preparation of stock solutions for Murashige and Skoog (MS) nutrient medium. It covers callus induction, micro-propagation, and the processes of hardening and acclimatization, which are essential for producing genetically uniform plantlets in vitro. Moreover, the manual provides demonstrations on advanced molecular techniques such as DNA isolation, gel electrophoresis, and DNA fingerprinting, tools that are indispensable for genetic studies and molecular diagnostics. By compiling these diverse yet interrelated techniques, this manual aims to equip readers with a solid foundation in both traditional and cutting-edge laboratory practices. Whether used in educational settings or research laboratories, this manual serves as an invaluable resource for mastering the essential techniques of modern biological science.

This manual is a comprehensive guide to the study of plant cells and various physiological and biochemical processes that govern plant growth and function. Aimed at students, researchers, and educators in the fields of botany and plant biology, it covers a broad spectrum of topics including the structure and distribution of stomata, imbibition, osmosis, plasmolysis, transpiration, photosynthesis, respiration, and nutrient analysis. As plant cells form the foundational building blocks of plant life, understanding their structure and functions provides crucial insights into broader plant processes that are essential for agricultural productivity, ecological balance, and the understanding of environmental responses. The topics covered in this manual provide both theoretical and practical knowledge, allowing readers to grasp the complexities of plant physiology through an integrated approach. The experimental methods detailed, such as the measurement of root pressure, the rate of transpiration, separation of photosynthetic pigments using paper chromatography, and the estimation of relative water content, offer hands-on techniques that aid in the application of knowledge in real-world scientific investigations. With advancements in technology, the manual also delves into cutting-edge techniques, such as the use of Infra-Red Gas Analyzers (IRGA) to measure photosynthetic CO2 assimilation. This work is designed to serve as a valuable resource for those wishing to explore the fascinating world of plant physiology and biochemistry. It is my hope that this practical manual will inspire a deeper understanding and appreciation of plant life and contribute to further research and discovery in this ever-evolving field.

The aim of this manual is to encompass a broad range of the latest plant molecular biology techniques. However, it is acknowledged that any manual will be read (and hopefully) used by a wide range of people with different levels of experience. Hence the remit of the manual was widened to include a full range of basic molecular tech niques, so that novices do not have to consult several texts to enable the execution of each major experiment. The manual is divided into three main parts: Part I: Basic Molecular Techniques The raison d'etre behind this part is to provide a background knowledge of molecular techniques, but also to reduce duplication in later chapters (this is particularly true of the methods contained in Chap. 1). All authors provided very detailed methods and often forgot that so me of these would be covered earlier. A particular favourite was DNA extraction methods, wherc everyone managed to provide a slightly different variant! My view was that it is far less confusing for the reader to be presented with one standard protocol and accom panying troubleshooting tips, than to read a different version in each chapter. In this way the basic techniques are addressed more in depth (and my apologies to all authors for judicious use of the delete key!). RNA methodology is covered in Chapter 3. This proceeds from the fundamentals of extraction, northern blotting etc. , to cDNA libraries.

The discipline of plant breeding has undergone transformation due to the assimilation of the rapid developments in molecular biology. The existing books on plant breeding deal mainly with the classical approaches, while specialized books on molecular approaches usually lack discussion of the classical methods. The book Plant Breeding for 21st Century attempts to present the complete picture of plant breeding ranging from the classical to the molecular approaches applied to crop improvement. The book is divided into four sections: Classical Plant Breeding, Transgenic technology, Molecular Markers, and Miscellaneous. The first section deals with the classical plant breeding and is divided into eight chapters. The second section has four chapters and describes transgenic technology. The third section discusses various aspects of molecular markers and is spread over three chapters. The final section has a single chapter dealing with variety release, seed multiplication and intellectual property rights. This book is designed primarily for graduate students, viz., B.Sc. agriculture and B.Sc. science students with botany as one of the subjects, who would get their first exposure to plant breeding. It would also be useful for the post-graduate students, especially in botany, and to teachers of the subject. The book is written in simple and easy to understand language. Illustrations and photographs have been provided wherever they were expected to facilitate comprehension of the subject under discussion.
