The STRENDA Commission (Standards for Reporting Enzyme Data), founded in 2003, is concerned with the improvement of the quality of reporting functional enzyme data to support, inter alia, enzyme kinetics for application in the in silico investigation of biological systems. The Commission has developed a set of guidelines for the reporting of data in publications. This 4th ESCEC symposium, organized by the Beilstein-Institut together with the STRENDA Commission, provided a platform to discuss the checklists, to consider further suggestions and to improve the existing recommendations. The STRENDA group took also the opportunity to discuss about the presented STRENDA electronic data capturing tool with members from diverse standardizations initiatives and systems biology groups such as MIBBI, YSBN, EFB and SYSMO, editorial board members from journals and all participating experimentalists and theoreticians. The focus of this presentation was to find answers on the questions such as how to organize and store these massive data sets in standard and easily accessible forms, which new experimental tools have to be developed to gather and configure such data into interactive models, which parameters should be measured, what kind of data constitute the minimum required information, and which experimental conditions should be recommended. The Beilstein Symposia address contemporary issues in the chemical and related sciences by employing an interdisciplinary approach. Scientists from a wide range of areas are invited to present aspects of their work for discussion, with the aim of not only advancing science, but also enhancing interdisciplinary communication. Traditionally, the Beilstein Symposia are kept small with up to 40 participants to provide a convivial atmosphere for the both lectures and intense discussions and the ready exchange of thoughts and ideas.

The multi-disciplinary approaches which are now typical of research in the natural and life sciences use a combination of modern experimental techniques which have led to an increased accuracy in the measurements of enzyme structures and activities. Modern analysis methods often result in the generation of huge amounts of data and large data sets, which are subsequently published in electronic data repositories and in written papers. However, data in both the literature and in databases suffer from the fact that they are often non-comparable due to incomplete and imprecise descriptions of materials and methods. Furthermore, if the experimental conditions are not fully and accurately stated, the values of the functional data of enzyme activities are of little use for, in particular, systems biology applications.Further problems occur even when the data are well reported; they will have often been collected under quite disparate conditions so that researchers are faced with the problem of the range of method-specific enzyme data. This is often an issue when data move between researchers whose data are supplied by laboratories that use different methods, and can, in the worst case, lead to misinterpretation of laboratory findings. The topics of the Proceedings of the 5th Symposium on ESCEC range from describing how the modification of enzyme structures affects the kinetics of enzyme reactions to discussing new results, approaches and methodologies for establishing physiological ties between sequence, structure and kinetics and modeled networks of collaborative enzymes. The overall topic - standard representation of enzyme data - is still considered and discussed in precise detail.

The kinetic mechanisms by which enzymes interact with inhibitors and activators, collectively called modifiers, are scrutinized and ranked taxonomically into autonomous species in a way similar to that used in the biological classification of plants and animals. The systematization of the mechanisms is based on two fundamental characters: the allosteric linkage between substrate and modifier and the factor by which a modifier affects the catalytic constant of the enzyme. Combinations of the physically significant states of these two characters in an ancestor-descendant-like fashion reveal the existence of seventeen modes of interaction that cover the needs of total, partial and fine-tuning modulation of enzyme activity. These interactions comprise five linear and five hyperbolic inhibition mechanisms, five nonessential activation mechanisms and two hybrid species that manifest either hyperbolic inhibition or nonessential activation characteristics depending on substrate concentration. Five essential activation mechanisms, which are taxonomically independent of the mentioned basic species, complete the inventory of enzyme modifiers. Often masked under conventional umbrella terms or treated as anomalous cases, all seventeen basic inhibition and nonessential activation mechanisms are represented in the biochemical and pharmacological literature of this and the past century, either in the form of rapid or slow-onset reversible interactions, or as irreversible modification processes. The full potential of enzyme inhibitors and activators can only be appreciated after elucidating the details of their kinetic mechanisms of action exploring the entire range of physiologically significant reactant concentrations. This book highlights the wide spectrum of allosteric enzyme modification in physiological occurrences as well as in pharmacological and biotechnological applications that embrace simple and multiple enzyme-modifier interactions. The reader is guided in the journey through this still partly uncharted territory with the aid of mechanistically-oriented criteria aimed at showing the logical way towards the identification of a particular mechanism.
