Abstract: Advances in the techniques and methodologies for the evaluation of protein quality are presented for agricultural researchers. An overall view of the strengths and weaknesses of the various evaluation methods is provided along with in-depth descriptions of several new and established techniques. The discussions cover the protein requirements of humans and their relevance to protein quality evaluation, the analytical methods for determining the amino acids profiles of particular foods, chemical and microbiological assays of protein quality, use of animal experimentation and clinical methods, and choice of procedure. The specific methodologies described include the Kjeldahl procedure for total nitrogen determination, sulphur amino acid analysis by performic acid pre-oxidation, the four-enzyme in vitro assay for protein digestibility, and procedures for the determination of net protein utilization and relative protein value in rats.

Consistent with the need to provide safe food for young children, particularly during the complementary feeding period between 12 and 36 months and the period of rapid development to age 59 months, the Food and Agriculture Organization of the United Nations (FAO) convened an Expert Working Group the FAO Headquarters, Rome, Italy, from 6 to 9 November 2017. The meeting addressed questions related to protein quality evaluation in two distinct products used to feed children in different conditions: Ready to Use Therapeutic Food (RUTF) and Follow up Formula for Young Children (FUF-YC). Specific meeting objectives were: •To determine the appropriate comparative protein or amino acid reference pattern to define protein quality for use in FUF-YC and RUTF. •To provide guidance on the preferred protein quality assessment methodology that should be stipulated with the standards for FUF-YC and RUTF. •To provide guidance on the measurement of protein and amino acid digestibility. •To provide the appropriate reference amino acid profiles and the amino acid composition of common ingredients used for FUF-YC and RUTF. •To provide cost implications for countries to use PDCAAS in FUF-YC and RUTF. This report provides future research recommendations including the need to generate data on the true ileal digestibility for different protein sources so that Digestible Indispensable Amino Acid Score (DIAAS) values can be used in the future.

Abstract: The use of a target protein against which the quality of other dietary proteins could be compared has been examined in rats. Milk, the natural food during the early life of mammals, was proposed as the target standard. The milk standard diet was compared with proteins of different origins, amino acid pattern, or lacking an essential amino acid. A chemical score based on rat's milk (CSM-R) was developed to evaluate the quality of the protein diets; it combined determination of a protein's most limiting amino acid with a capacity for ranking proteins by nutritional values. Diets were evaluated for protein efficiency ration, body weight gain, levels of amino acids and protein complementation. Results indicated that rat milk contains a proportion of amino acids which provides maximum protein utilization in growing rats, and may also protect the lactating mother. Other diets were usually inferior. The CSM-R score appeared to be a useful analytic tool in evaluating the quality of dietary proteins.

Variations in feeding frequency may lead to difference in utilization of dietary nutrients. The effects of feeding frequency have been tested in growing pigs and finishing pigs using respiratory quotient and postprandial kinetics of plasma metabolites. A reduced feeding frequency resulted in an increased lipogenesis and proteolysis shortly after the meal both in growing and finishing pigs. Nevertheless, growing and finishing pigs exhibit different metabolic pathways when feeding frequency is reduced.

Protein quality evaluations currently require that an animal bioassay be performed to determine true protein digestibility (TPD). This measure is used to calculate the protein digestibility corrected amino acid score (PDCAAS) prior to qualifying for a protein content claim. Several in vitro assays evaluating protein digestibility that currently reside in literature have not been used to assess protein quality, hence this thesis was undertaken to evaluate the pH-drop (PHD) and two-step digestion (TSD) method to determine PDCAAS in vitro on 65 plant-based proteins composed of different species, variety and heat treatment. Strong significant correlations were indicated by both in vitro protein quality calculations for the PHD (R = 0.91; P ≥ 0.0001) and the TSD (R = 0.92; P ≥ 0.0001) with the PDCAAS. This relationship suggests that either method to evaluate protein digestibility may be reliable to calculate PDCAAS in vitro, thus reducing the time, expense and ethical quandaries that currently apply to evaluating protein quality content claims.
