Early embryo development in mammals begins with the recognition and fusion of gametes of both sexes, followed by a series of mitotic cell divisions. The gamete fusion results in the formation of totipotent cells, which are capable of developing into three basic cell lineages in mammals. These include the trophoblast, surrounding the embryo and facilitating communication with the mother, the epiblast which develops into the fetus, and the primitive endoderm which forms the yolk sac. Until implantation, the embryos undergo remarkable changes, including histone modifications, chromatin remodeling, epigenetic modifications of chromatin, the onset of transcription from the embryonic genome, and the first differentiation to the formation of the trilaminar disc, containing all primary cell lineages required for the formation of fetal tissues and organs. Remarkably, some of the most complex events of early development, such as fertilization, are achieved during complete transcriptional silencing. Another unique feature is how fast the development proceeds. In humans, reaching the stage of early gastrulation takes approximately two weeks.

As the age of childbearing increases, reduced female gamete (egg) quality has emerged as the primaery cause of infertility in women. This book addresses molecular mechanisms that regulate oocyte quality as well as intrinsic and extrinsic factors that compromise these mechanisms. Eggs are produced through a unique cell division called meiosis. Oocyte meiosis is initiated early in fetal life. Shortly before birth, the oocyte undergoes a prolonged arrest at prophase I until the age of puberty, at which selected oocytes resume meiosis I. Following meiotic resumption and during oocyte maturation, tight regulation of nuclear and DNA-related mechanisms ensures proper homologous chromosome segregation prior to another arrest at metaphase II. Any errors prior to completion of meiosis I can lead to chromosome segregation errors and aneuploidy, the most common genetic cause of miscarriage and congenital disorders such as Down syndrome. In addition to nuclear maturation, several cytoplasmic processes are orchestrated to support cell division and to generate developmentally competent eggs capable of supporting fertilization and early embryonic development. The interaction between these nuclear and cytoplasmic processes is critical for producing good-quality oocytes. "Fertilization and embryonic development depend on the acquisition of developmental competence by the oocyte, which is defined by the achievement of growth and meiotic maturation. Therefore, learning the fundamental biology of these two crucial events under the physiological conditions is essential for understanding oocyte developmental abnormalities which directly manifest as infertility. In this thesis, I addressed both events of oocyte growth and meiotic maturation. Manuscript I is focused on the possible role of Yes-associated protein (YAP) __ the key effector of the Hippo signalling pathway which acts as a transcriptionl co-activator and is well known for being a powerful promoter of organ size and development __ in the process of oocyte growth. We report that YAP is expressed in the oocyte throughout the entire window of oocyte development but is constitutively excluded from the nucleus and thus inactive as a transcriptional co-factor. We also show that this nuclear exclusion, which causes YAP not to play a significant transcription-based physiological role in promoting oocyte growth, is induced by multiple phosphorylation-dependent and -independent mechanisms acting cooperatively within the oocyte. In manuscript II, we move on from the growth phase and focus on the next stage of oocyte development, meiotic maturation. It is known that during oocyte growth in mammals, the surrounding granulosa cells elaborate many specialized actin-rich filopodia, termed transzonal projections (TZPs), that contact the oocyte plasma membrane and enable transmission of essential signals and metabolites from the granulosa cells towards the oocyte. During meiotic maturation, the TZPs are lost, freeing the germ cell from these granulosa-cell regulatory influences prior to fertilization. Here we define the signalling pathway that regulates TZPs maintenance and loss. We show that prior to fully grown oocyte maturation, high levels of cyclic GMP (cGMP) in the neighbouring granulosa cells (cumulus cells) maintain TZPs. However, during maturation, activation of epidermal growth factor-like receptor (EGFR) signalling within the cumulus cells decreases the cGMP level. Lack of cGMP in turn enables Rho-associated kinase (ROCK), a key cytoskeleton organizer, to trigger retraction of the TZPs likely via actomyosin contractility. Our results reveal a new role for both EGFR and cGMP in regulating cumulus cell-oocyte communication and identify the molecular mechanism underpinning TZP loss during maturation. The findings in this thesis provide new insights into molecular basis of oocyte normal and healthy development which could be applied in the clinical methods and therapeutic innovations aiming to preserve fertility"--
