A key determinant of synaptic efficacy is the localization and density of neurotransmitter receptors in post-synaptic membrane. During the development of the neuromuscular junction (NMJ) the nicotinic acetylcholine receptor (AChR) becomes localized in high-density clusters via the motor neuron-derived protein agrin, its signaling partner MuSK, and the intracellular scaffolding protein rapsyn; however, many of the mechanisms and interactions involved in AChR localization have remained elusive. In this thesis I investigate how rapsyn interacts with the AChR, how this is regulated by agrin-induced phosphorylation of the AChR, and how this contributes to receptor localization and stability at the NMJ. First, using chimeric proteins containing an exogenous transmembrane domain and the cytoplasmic loop of the AChR subunits we demonstrate that rapsyn binds constitutively to the MA helix in the C-terminal region of the [alpha], [beta] and [varepsilon] cytoplasmic loops. Second, we determine how rapsyn also mediates agrin/MuSK-induced phosphorylation of the AChR. We show that MuSK/rapsyn expression induces parallel but independent phosphorylation of AChR [beta]Y390 and [delta]Y393, and that this is mediated by rapsyn's c-terminal domains, which localize and/or activate cytoplasmic tyrosine kinases that phosphorylate the receptor. Third, we used CD4 subunit loop chimeric proteins to screen for additional interactions that localize the AChR. Intriguingly, we find that a 20 amino acid region of the AChR-[beta] loop centered on Y390 is sufficient for postsynaptic localization, and that phosphorylation of βY390 is required for clustering. Interestingly, phosphorylation of [beta]Y390 regulates binding of the 20 amino acid motif to rapsyn and we show that this increases the stoichiometry of rapsyn to AChR. Thus, rapsyn appears to interact with the AChR via two distinct sites: constitutive binding to the MA-helix of one subunit cytoplasmic loop and regulated binding to the Y390 region of [beta] subunit loop. Fourth, we investigated whether agrin signaling - and enhanced rapsyn binding - not only clusters the AChR but also increases its metabolic stability. Indeed, we find that phosphorylation of AChR-[beta]Y390 increases receptor half-life by ~1.8 fold, which likely helps maintain functional levels of receptor at both developing and mature NMJs. Together, these findings help define the molecular interactions and regulatory mechanisms that localize nicotinic acetylcholine receptors at the NMJ, ensuring reliable neuromuscular transmission.
