Receptors and Hormone Action, Volume III, is part of a multivolume series that summarizes advances in the field of hormone action. The articles contained in these books are oriented toward a description of basic methodologies and model systems used in the exploration of the molecular bases of hormone action, and are aimed at a broad spectrum of readers including those who have not yet worked in the field as well as those who have considerable expertise in one or another aspect of hormone action. This book opens with a chapter on the physiological properties of the thyroid hormone receptors in the intact animal. This is followed by separate chapters on ß-adrenergic receptors; the study of hormone-receptor interaction by measuring the biological responses induced by the actions of gonadotropins on Leydig cells; chemical and immunochemical properties of hCG and PMSG treated with glycosidases; and binding of follitropin (FSH) to rat testes. Subsequent chapters deal with the control of changes of gonadotropin responsiveness of the granulosa cell during follicular maturation; regulation of prolactin receptors by steroid hormones; and the role of membrane protein phosphorylation in the effects of neurotransmitters.

This is followed by separate chapters on conformational forms of the estrogen receptor; the relationship of early responses of the cell to estrogen to DNA synthesis; the role of receptors in the anabolic action of androgens; and biology of progesterone receptors. Subsequent chapters deal with the molecular structure and analysis of progesterone receptors; the regulation of gene expression by glucocorticoid hormones; studies of the aldosterone receptor in the adrenalectomized rat kidney; and existence of gonadal steroid receptors in brain and pituitary tissue.

The purpose of this book is to focus attention on recent developments in steroid and sterol hormone action. Many authors have generously contributed to the book. As a result, there is a great diversity of opinion! A majority of the chapters deal with steroid or sterol hormone receptors. This is not meant to imply that receptor-mediated mechanisms are the sole or even the most important mechanisms by which steroid hormones act in the cell. There is wealth of evidence showing that other, non-receptor events, are important also. Steroid hormone recep tor research and the study of nuclear events mediated by steroids are presently the most intensely studied aspects of sterol hormone action and our selection of topics reflects this trend. We have also included chapters on vitamin 0 sterols and thyroid hormone in the book, as there is pood evidence that these hormones act in a manner similar to other classical steroids. 1 IMMUNOCHARACTERIZATION OF THE NUCLEAR ACCEPTOR SITES FOR THE AVIAN OVIDUCT PROGESTERONE RECEPTOR A. GOLDBERGER, M. HORTON, T. C. SPELSBERG Department of Biochemistry and Molecular Biology, Mayo Clinic and Mayo Graduate School of Medicine, Rochester, MN 55905 INTRODUCTION It is well known that steroid hormones, certain vitamins and sterols, enter target cells and bind to specific protein receptors in the cyto plasm or nucleus (1-4). This binding is saturable, high affinity, and steroid specific.

Mechanisms of Hormone Action: A NATO Advanced Study Institute focuses on the action mechanisms of hormones, including regulation of proteins, hormone actions, and biosynthesis. The selection first offers information on hormone action at the cell membrane and a new approach to the structure of polypeptides and proteins in biological systems, such as the membranes of cells. Discussions focus on the cell membrane as a possible locus for the hormone receptor; gaps in understanding of the molecular organization of the cell membrane; and a possible model of hormone action at the membrane level. The text also ponders on insulin and regulation of protein biosynthesis, including insulin and protein biosynthesis, insulin and nucleic acid metabolism, and proposal as to the mode of action of insulin in stimulating protein synthesis. The publication elaborates on the action of a neurohypophysial hormone in an elasmobranch fish; the effect of ecdysone on gene activity patterns in giant chromosomes; and action of ecdysone on RNA and protein metabolism in the blowfly, Calliphora erythrocephala. Topics include nature of the enzyme induction, ecdysone and RNA metabolism, and nature of the epidermis nuclear RNA fractions isolated by the Georgiev method. The selection is a valuable reference for readers interested in the mechanisms of hormone action.

The Eleventh Midwest Conference on Endocrinology and Metabol ism in September 1975 brought together a number of leading invest igators in the areas of steroid, peptide, acetylcholine, and cate cholamine hormone receptor studies. This book is based upon the reports of investigations into hormone receptor biochemistry and physiology presented at the Conference as well as on the ensuing discussions. However, many of the manuscripts were written after the Conference. Because of this, the reader will find that some literature references and results of i~vestigations are more up to-date than the Conference date would suggest. Perusal of this and previous volumes will attest to the high quality of this annual conference, due primarily to the efforts of the Planning Committee. We also acknowledge the superb efforts of the staff of Conferences and Short Courses for their assistance in organizing the Conference, and Mrs. Linda Bennett for expert sec retarial assistance in the typing of the book. Last, but certain ly not least, we express our appreciation to the various organiza tions that provided the necessary financial assistance. Leonard R. Forte John M. Franz David M.
