In mammals, the postnatal brain contains regions where proliferation is maintained, such as the subventricular zone (SVZ) or sub-granular zone (SGZ) which are capable of producing new neurons of the olfactory bulb (OB) or dentate gyrus (DG). In addition, the DG is mostly generated postnatally. My project was to investigate the role played by Slit and Robo molecules in the postnatal and adult forebrain, particularly in neurogenesis. Using a genetic approach in vivo, we generated mutant mouse lines for Slits and Robos (deficient and conditional) that we crossed with mice expressing Cre recombinase in the SVZ, the OB and the DG/SGZ. In these animals, the absence of Slits/Robos causes severe developmental abnormalities of the forebrain commissures similar to those described in full knockouts. The absence of certain Slit/Robo molecules in the dorsal telencephalon is linked with defects in neuronal migration of SVZ-derived cells, but barely any effect on their proliferation. Also, in some mice deficient for Robos, we observed a decrease in the number of granule neurons generated in the DG and in the OB. In addition, the results obtained from the line Foxg1::Cre; Slit2lox/lox show a decrease in the number of new neurons from the SVZ and reduced hippocampal neurogenesis. The hippocampus of mice Foxg1::Cre;Slit2lox/lox is of normal size at birth, but very small in adults. In conclusion, I have performed one of the first in vivo characterizations of the role of Slits and Robos proteins in the postnatal forebrain.

Developing the second edition of Neural Development and Stem Cells was neces- tated by the rapid increase in our knowledge of the development of the nervous system. It has become increasingly clear that stem cells are a heterogeneous population that changes extensively during development. Perhaps the most important advance in our understanding of stem cell behavior has been the realization that regionalization of stem cells occurs early in development and this bias toward differentiation in phe- types of neurons or cells characteristic of a particular part of the brain appears to persist even after prolonged culture. We have therefore included additional chapters on olf- tory epithelial stem cells and retinal stem cells, both of which differ in their properties from ventricular zone and subventricular zone–derived neural stem cells. It is also now clear from an analysis of mutants and transgenics where the death or self-renewal pa- way is altered that cell death regulates stem cell number. As a consequence, this second edition includes a separate chapter on cell death that summarizes the important changes in the death pathway that occur as stem cells mature. The existing chapters in the book have also been extensively revised and updated by experts who have generously c- tributed their time and expertise. Theneurotransmitter dopamine has just celebrated its 50thbirthday. The discovery of dopamine as a neuronal entity in the late 1950s and the notion that it serves in neurotransmission has been a milestone in the field of neuroscience research. This milestone marked the beginning of an era that explored the brain as an integrated collection of neuronal systems that one could distinguish on basis of neurotransm- ter identities, and importantly, in which one started to be able to pinpoint the seat of brain disease. The mesodiencephalic dopaminergic (mdDA) system, previously designated as midbraindopaminergic system, has received much attention since its discovery. The initial identification of dopamine as a neurotransmitter in the central nervous system (CNS) and its relevance to psychiatric and neurological disorders have stimulated a plethora of neurochemical, pharmacological and genetic studies into the function of dopamine neurons and theirprojections. In the last decade, studies on gene expression and development have further increased the knowledge of this neuronal population and have unmasked a new level of complexity. The start of the molecular dissection of the mdDA system has been marked by the cloning and characterization ofNurrl and Pitx3. These transcription factors were shown to have a critical function during mdDA development. These initial studies have been followed by the identification of many other proteins, which have a crucial function in the creation of a dopamine neuron permissive region, induction of precursors, induction of terminaldifferent- tion and finally maintenance of the mdDA neuronal pool.
