The rapid advances made in the study of the synthesis, structure and function of biological macromolecules in the last fifteen years have enabled scientists concerned with antimicrobial agents to achieve a considerable measure of understanding of how these substances inhibit cell growth and division. The use of antimicrobial agents as highly specific inhibitors has in turn substantially assisted the investigation of complex biochemical processes. The literature in this field is so extensive however, that we considered an attempt should be made to draw together in an introductory book the more significant studies of recent years. This book, which is in fact based on lecture courses given by us to undergraduates at Liverpool and Manchester Universities, is therefore intended as an introduction to the biochemistry of antimicrobial action for advanced students in many disciplines. We hope that it may also be useful to established scientists who are new to this area of research. The book is concerned with a discussion of medically important antimicrobial compounds and also a number of agents that, although having no medical uses, have proved invaluable as research tools in biochemistry. Our aim has been to present the available information in a simple and readable way, emphasizing the established facts rather than more controversial material. Whenever possible, however, we have indicated the gaps in the present knowledge of the subject where further information is required.

The central theme of this book is that systems of cell-cell signalling via nerves, hormones, local mediators and growth factors are not distinct phenomena, but branches of one general mechanism. These topics therefore can and should be discussed in an integrated manner, and the division of cell signalling studies into separate pigeonholes such as neuroscience, endocrinology or cancer biology is unnecessary, if not counterproductive. I also believe it to be unfortunate that there is not a collective term to describe neurotransmitters, hormones, local mediators and growth factors, other than clumsy phrases such as "extracellular signal molecule". The lack of a short and distinctive word for these entities genuinely hampers people from thinking about them in an integrated way. Having decided that it was presumptuous to invent a new term, I have chosen in this book the term first messenger to cover all types of extracellular signal molecule, because of the widespread acceptance of the term second messenger to represent the intracellular signal molecules that are produced in response to many of them. I have given the book the title "biochemical messengers", which is a global term to cover both first and second messengers. The impetus for writing the book came, as must often be the case, when I had to put together a course on cell-cell signalling for biochemistry students at the University of Dundee.

Enzyme kinetics has undergone very rapid growth and development during the past fifteen years and has been well received by the biochemical community. A cursory glance at the current biochem ical literature reveals the increasing popularity of enzyme ki netics1 yet, there are very few books available to guide the enzymologist who wishes to conduct kinetic experiments. This monograph was undertaken to provide the fledgling kineticist with an outline of contemporary initial rate enzyme kinetics. A large portion of the material contained in this book is presented in a second-year, graduate-level course in biochemistry at Iowa State University. I have found that the presentation in this course has enabled students without a strong background in math ematics to undertake initial rate studies at the research bench. The monograph obviously is more comprehensive than any course could be, and should permit similar accomplishment. As the title implies, the major emphasis of this monograph is on initial rate enzyme kinetics. I considered at length the advis ability of including chapters on integrated rate equations and on the theory and application of rapid reaction kinetics, such as rapid-mixing stopped-flow, and temperature-jump kinetics. These, however, are topics that would require a good deal of space to develop if they were to be helpful to the beginner.

The development of powerful new techniques and refmements of tech niques in molecular genetics in recent years, and the surge in interest in biotechnology based on genetic methods, have heralded a new golden age in molecular genetics, and stimulated in diverse disciplines much interest in the technologies themselves and their potential uses in basic and applied biomedical sciences. Although some excellent specialist laboratory manuals (especially the Cold Spring Harbor Laboratory manuals by I. H. Miller; R. W. Davies et al. ; and T. Maniatis et al. ) on certain chapters of molecular genetics exist, no general text that covers a broad spectrum of the sub ject has thus far been published. The purpose of this manual is to pre sent most, though of necessity not all of the important methods of molecular genetics, in a series of simple experiments, many of which can be readily accomplished by the microbiologist, biochemist or biotechnologist that has had only limited exposure to genetics. The remainder of the experiments require either greater familiarity with the subject, or guidance by someone with such experience. The book should, therefore, not only enable individuals to acquire new proce dures for ongoing projects, but also serve as a basis for the teaching of molecular genetic techniques in formal predoctoral and postdoctoral laboratory courses.
