The development of synchrotron radiation (SR) as a research tool was driven largely by the needs of materials scientists and solid-state physi cists. However, the availability of SR has extended significantly the capa bility of scientists who study biological structure with radiation. This volume contains some of the results reported at a symposium held at Brookhaven National Laboratory in May 1988 to discuss the application of synchrotron radiation to structural biology. The book addresses the most recent developments in structural and functional proteomics underlying the recent contributions given in these areas by our laboratory to the instrumentations, the methods and the procedures as mutuated from the nanoscale sciences and technologies. These developments introduced in the last few years make now possible protein massive identification (mass spectrometry and biomolecular arrays down to nanoamounts) and protein structural characterization in solution and in crystals down to the atomic scale to an extent and to a degree so far unmatched. Emphasis is placed in the growth by nanobiofilm template of protein crystals of any type and size from millimeter to micron, leading in combination with microfocus synchrotron technology and atomic force microscopy to the definition of a new field called nanocrystallography. The few useful examples being shown, concerning yet structurally unsolved proteins, point this very promising approach nanotechnology-based in structural proteomics using highly focused X-rays. This has not to be confused with the important study of nanocrystals, both organic and inorganic, and novel diamond like nanocomposite materials and devices having 3D protein crystals as matrices to be equilibrated with nanoparticles/gold/silver to be utilized in the most diversified electronic applications here also summarized. vii Acknowledgments We are particularly grateful to Giuseppe Zanotti at the University of Padova for his fundamental collaboration during all the crystallographic studies.

This book the second volume in the "Springer Series in Biophysics" col lects together contributions to the conference on "Biophysics and Syn chrotron Radiation" held in July 86 at Frascati. This meeting addressed the advances on the structure of biological molecules obtained by using synchrotron radiation. In fact it was most timely to review the results of the research in biophysics which is rapidly developing at synchrotron radiation facilities. Moreover, there was interest to discuss the new perspectives opened up by the future high brilliance synchrotron radia tion sources. With the use of synchrotron radiation, x-ray spectroscopy of biological molecules is firmly established in the techniques of EXAFS and XANES. Contributions to the detailed knowledge of local structure of active sites of metalloproteins by this approach are presented in this volume, together with a number of studies of -the interaction of metal ions with other important biological macromolecular systems. Structural determination of very large biological systems at high reso lution, including a protein and its substrate, are reported. The experi mental advances in protein crystallography presented here reduce the time for solving protein structures, thus satisfying a major require ment of the rapidly-expanding field of protein engineering.

The individual papers that comprise this monograph are derived from two American Chemical Society (ACS) Fall National Meetings that focused on the current uses of synchrotron radiation (SR) research techniques. The first Symposium was held in Washington, DC, in August 1994, and the second convened in Chicago, IL, in August 1995. The intent of these symposia was to present a broad overview of several current topics in industrial, chemical, and materials-based SR research to a chemically inclined audience. The SR techniques covered were divided roughly into the three general fields of industrial, chemical, and materials science for this purpose. Included within these four categories are environmental, geologic, atomic/molecular, analytical, solid state physics, surface science, and biological applications of SR. There is little doubt that structural biology and environmental science are the largest growth areas in SR research as this monograph goes to press. The spirit of these symposia was to bring together the expert synchrotron radiation user with new and potential users of SR techniques. There are now a preponderance of particle storage rings, located throughout the world, devoted exclusively to the production of SR. There have been great improvements in the particle accelerators and storage rings from which SR emanates. These newest third generation SR sources are the result of the successful collaboration between SR users and accelerator physicists which has made a reality out of experiments never before possible.
