T and B Lymphocytes: Recognition and Function is the 16th volume of the 1979 ICN-UCLA Symposia on Molecular and Cellular Biology. This book is organized into seven sections, encompassing 74 chapters that summarize molecular level progress in the field of immunobiology. It emphasizes cell membrane, techniques developed for its analysis, and varied products of the major histocompatibility complex, as well as other receptors on cells of the immune system. The book starts by describing two approaches for molecular association evaluation. First is the use of artificial membranes or lipid vesicles for the incorporation of antigens for recognition by T-lymphocytes both at the afferent and efferent levels. Second is the chemical crosslinking of cell surface components with a variety of different crosslinking reagents. This is followed by a discussion on the molecular nature of a T-cell receptor that provides insights of the target(s) recognized by T-lymphocytes. Several chapters discuss data dealing with the issue of the apparent differential recognition by T- and B-cells while both use the same VH genes. Antiidiotypic sera, together with the activity of ""regulator"" T-cells, are also discussed, with regard to their role in networks of regulation of the immune response. This book presents works demonstrating that human T-lymphocytes can also be cloned and both noncytotoxic proliferating T-lymphocytes and Tc can be so obtained. This is along with the continuing studies regarding heterogenity of B-lymphocytes as well as macrophages. Part V focuses on the interaction between lymphocyte and virus and molecular modifications of viral-infected cells. The subsequent section deals with recognition, reactivities, and function of T- and B-lymphocytes. Considerable chapters in this section discuss T-cell cytotoxicity, dichotomy of MHC control over anti H-Y cytotoxic T-cell responses, mouse alloantibodies, and mixed lymphocyte reactions. The concluding part describes immunological tools, such as synthetic membranes and cloned T-cells with biological function.

How do you discriminate yourself from other people? This question must sound odd to you since you easily recognize others at a glance and, without any effort, would not mistake them for yourself. However, it is not always easy for some people to discriminate themselves from others. For example, patients with schi- phrenia often talk with “others” living inside themselves. Thus it is likely that n- mally your brain actively recognizes and remembers the information belonging to yourself and discriminates it from the information provided by others, although you are not conscious of it. This brain function must have been particularly important for most animals to protect their lives from enemies and for species to survive through evolution. Similarly, higher organisms have also acquired their immune system through evolution that discriminates nonself pathogens and self-body to protect their lives from pathogens such as bacteria or viruses. The brain system may distinguish integrated images of self and nonself created from many inputs, such as vision, sound, smell, and others. The immune system recognizes and distinguishes a variety of structural features of self and nonself components. The latter actually include almost everything but self: for example, bacteria, viruses, toxins, pollens, chemicals, transplanted organs, and even tumor cells derived from self-tissue. To this end the immune system recruits different kinds of immune cells, such as B and T lymphocytes, natural killer (NK) cells, dendritic cells, and macrophages.

The importance of thymus-dependent cells, or T cells, in the generation of a successful immune response was first realized in the early sixties. In the follow ing two decades, a succession of elegant experiments established the antigen specificity of T cells and their ability to perform both as regulatory and effector cells. T cells were shown to be essential in most immune reactions, playing a crucial role in augmenting the activity of effector T and B cells against 'foreign' antigen, as well as in the suppression of effector activity against self antigens. The means by which T cells differentiate 'foreign' from 'self' antigens is based on their recognition of antigen almost exclusively in the context of self major histocompatibility complex products, unlike B cells, which recognize an tigen alone. It is this recognition, mediated by the T-cell receptor, that sets into motion the diverse cell-cell interactions, which control the differentiation and regulation of the immune response. Although its importance was well established, the molecular nature of the T-cell receptor remained elusive for two decades. Many hypotheses as to its structure and precise function were put forward, using immunoglobulin as a basis for conjecture, but "the Holy Grail of Immunology" remained ephemeral until three years ago. In the ensuing years, both immunologists and molecular biologists have contributed to an explosion of data unsurpassed by any previous period in the field.

T-cells are an essential component of the immune system that provide protection against pathogen infections and cancer and are involved in the aetiology of numerous autoimmune and autoinflammatory pathologies. Their importance in disease, the relative ease to isolate, expand and manipulate them ex vivo have put T-cells at the forefront of basic and translational research in immunology. Decades of study have shed some light on the unique way T-cells integrate extrinsic environmental cues influencing an activation program triggered by interactions between peptide-MHC complexes and the antigen-recognition machinery constituted of clonally distributed T-cell receptors and their co-receptor CD4 or CD8. This symposium was established in 1976 for the purpose of bringing to gether once every two or three years, active investigators in the fore front of contemporary immunology, to present their findings and to discuss their significance in the light of current concepts and to identify important new directions of investigation. The founding of the symposium was stimulated by the achievement of major breakthroughs in the under standing of the immune recognition of proteins and peptides. We believed that these breakthroughs will lead to the creation of a new generation of peptides which should have enormous potential in biological, therapeutic and basic applications. This anticipated explosion has finally occurred and many applications of these peptides are now being realized. The main symposia topics of the fourth symposium were: T-cell recognition of proteins, structure and function of the T-cell receptor, presentation of protein antigens, recycling and activation of membrane receptor molecules, Ir-gene control of T-cell responses and methods of cell separation. The molecular features recognized by antibodies on proteins were the first immune recognition sites to be local ized and confirmed by synthetic peptides. The complete antigenic structures of several proteins have been defined, and individual antigenic sites have been described on many more proteins. More recently, major breakthroughs have been reported in the immune recognition of proteins by T cells.
