This book has had a three-fold origin, corresponding to the discoveries made by the three authors and their collaborators during the last few years - mostly since 1962. A most fruitful symposium on the cerebellum was held in Tokyo at the time of the International Physiological Congress in September 1965, and there was then formulated the project of writing this book so as to organize all this new knowledge and make it readily available, and to give opportunity for the con ceptual developments that may be seen in Chapters XI, XII and XV in particular. The present account of the physiological properties of the cerebellar cortex is based to a large extent on systematic investigations that were concerned with discovering the mode of operation of the constituent neuronal elements of the cerebellar cortex. This work was carried out in the Physiology Department of the Australian National University from 1963 to 1966 in collaboration with several visiting scientists - initially Drs. ANDERSEN, OscARssaN and VooRHOEVE and later Drs. LuNAs, SAsAKI and STRATA - to all of whom grateful thanks are extended for a great many of the figures, and even more significantly for the original and critical contributions that they made to so many aspects of this exploration into the mode of operation of the neural machinery of the cerebellar cortex.

Regeneration, the homeostatic ability to maintain tissue structure in the face of normal cell turnover or loss of tissue damaged by trauma or disease, is an essential developmental process that continues throughout life. As recently as a decade ago, any serious discussion of the possibility of regeneration becoming a practical medical tool in the near future had the air of science fiction or over-optimistic speculation. The term “regenerative medicine” was certainly on many lips but few actually expected to soon see it applied in a clinical setting. A tidal wave of discovery has changed that and investigating the cellular mechanisms of natural regeneration has become one of the hottest topics in developmental biology and biomedicine in general. Many researchers entering the field find that the regeneration literature is still quite diffuse perhaps owing to the disparate biological systems that have been the object of study including hydra, planaria, newts, axolotls and more recently several mouse strains. Early in the 80’s date the first observations on the existence of hormonal steroids that may be synthesized and act in the nervous system. In order to refer to these endogenous steroids, proved important to control both central and peripheral nervous system, it was proposed the term “neurosteroids” (NSs). Over the years, their importance in regulating the physiological functions of neuronal and glial cells increased progressively. These steroids can be involved in several pathophysiological conditions such as depression, anxiety, premenstrual syndrome (PMS), schizophrenia and Alzheimer disease. Among the different classes of NSs, the progestagens revealed particularly important. The progesterone metabolite 5a-pregnan-3a-ol-20-one, also named tetrahydroprogesterone or allopregnanolone (ALLO) was one of the first most important steroid that was originally shown to act as neurosteroid. ALLO is synthesized through the action of the 5aR-3a-HSD, which converts P into DHP and subsequently, via a bidirectional reaction, into ALLO. NSs exert complex effects in the nervous system through ‘classic’, genomic, and ‘non-classic’, non-genomic actions. ALLO displays a rapid ‘non-genomic’ effect, which mainly involves the potent modulation of the GABA type A (GABA-A) receptor function. Recently a membrane receptor has been identified as target for ALLO effects, i.e. the membrane progesterone receptors (mPRs) that are able to activate a signalling cascade through G protein dependent mechanisms. By these ways, ALLO is able to modulate several cell functions, acting as neurogenic molecule on neural progenitor cells, as well as by activating proliferation and differentiation of glial cells in the central and peripheral nervous system. In this topic, we review the most recent acquisitions in the field of neurosteroids, focusing our attention on ALLO because its effects on the physiology of neurons and glial cells of the central and peripheral nervous system are intriguing and could potentially lead to the development of new strategies for neuroprotection and/or regeneration of injured nervous tissues and for the treatment of neuropsychiatric disorders.

This book is organized into three parts that correspond with the main groups of chapters delivered during the Cajal Centenary Meeting on The Neutron Doctrine. These chapters represent important aspects of the morphology, development, and function of the cerebellum and related structures. Clearly an exhaustive analysis of all aspects of the cerebellar system, as they relate to the legacy of Ramon y Cajal, would be impossible to contain in just one volume, given its far-reaching impact. Instead, we deliberately steered away from the traditional handbook approach that some of us have taken in the past and selected those aspects of cerebellar research currently under vigorous study that would also represent the widest scope of interest for neuroscientists in general and for cerebellar specialists in particular. In particular, we felt that as the discrete anatomy of the cerebellum is quite well known, only certain aspects of the structure should be discussed here. For example, the organization of the pontocerebellar pathways, we felt, would be particularly interesting given the enormity of the system in higher vertebrates. Also of interest is the distribution and development of the synaptology and neurotransmitter properties in this cortex. Indeed, from the point of view of cerebellar development, this may represent one of the clearest paradigms in the understanding of rules for neurogenesis for the central nervous system.
