This thesis examined the contribution of dopaminergic neurons of the ventral tegmental area (VTA), a crucial component of the mesolimbic dopamine system, to the process of thermoregulation. -- The control of body temperature, as a physiological regulator in response to environmental factors or emotional triggers, is vital to the survival of all animals. However, very little is understood in regard to the neurotransmitters, brain areas and neural pathways that are involved in its regulation. As such, many autonomic conditions, such as Parkinson's disease and psychogenic fever, which exhibit thermoregulatory symptoms are poorly understood and consequently undertreated. -- Dopamine is a neurotransmitter known to be involved in the regulation of central bodily cognitive and motor functions. Recent findings have indicated that it also plays an important role in body temperature. However, its central mechanisms of action are unknown. One of the major nuclei in the brain that contains dopamine-producing neurons is the VTA, which is approximately 30-70% dopaminergic. Thus, the present study aimed to elucidate the specific role of VTA dopamine neurons in the control of thermoregulatory outputs. -- Within the past decade new techniques, such as chemogenetics, have emerged which allow for the specific study of neurotransmitter function in defined brain regions. Hence, this study utilized, the designer receptors exclusively activated by designer drugs (DREADDs) system, in conjunction with specifically modified transgenic animals and specially designed ligands to investigate the effect of temporary neuronal population activation on the common thermoregulatory response, known as emotional hyperthermia. Targeted toxins were also applied to determine the effect of dopamine signalling reduction on various thermoregulatory parameters. All biological parameters were assessed in conscious, freely moving animals through the chronic implantation of thermistor probes, locomotor behaviour tracking and food consumption monitoring. -- The results presented in this thesis conclusively demonstrate the vital contribution of dopaminergic VTA neurons to the process of thermoregulation; as well as their impact on physiological processes - such as feeding and locomotion. VTA dopaminergic enhancement was found to substantially increase the baseline temperature of animals. This was simultaneously accompanied by changed feeding patterns and locomotor hyperactivity. Specific dopamine receptor influence and feeding behaviour alterations were assessed as a potential contributing factor to the observed temperature elevation. This facilitated the validation of D1 dopamine receptor involvement in the process of body temperature control. Lastly, the influence of the mesolimbic dopamine system, originating in the VTA and projecting to the nucleus accumbens, was assessed. Although this pathway has been implicated in locomotion, feeding and emotional processing behaviours - making it a potential candidate for the control for thermoregulation, it was determined that it is likely not the primary pathway responsible for thermoregulation. -- Taken together, the evidence provided in this investigation adds fundamental biological information to the field of integrative neuroscience, which helps the understanding of connections between brain regions and physiological responses. Furthermore, the research conducted validates the use of modern genetic techniques in conscious animal physiological research. Importantly, the identification of a thermoregulatory brain region, as well as the receptor type involved in its function, may assist in the development of new, more effective, and targeted treatments for those suffering with thermoregulatory diseases.

Intermittent social defeat stress induces cross-sensitization to psychostimulants and escalation of drug self-administration. These behaviors could result from the stress-induced neuroadaptation in the mesocorticolimbic dopamine circuit. Brain-derived neurotrophic factor (BDNF) in the ventral tegmental area (VTA) is persistently elevated after social defeat stress, and may contribute to the stress-induced neuroadaptation in the mesocorticolimbic dopamine circuit. BDNF modulates synaptic plasticity, and facilitates stress- and drug-induced neuroadaptations in the mesocorticolimbic system. The present research examined the role of mesolimbic BDNF signaling in social defeat stress-induced cross-sensitization to psychostimulants and the escalation of cocaine self-administration in rats. We measured drug taking behavior with the acquisition, progressive ratio, and binge paradigms during self-administration. With BDNF overexpression in the ventral tegmental area (VTA), single social defeat stress-induced cross-sensitization to amphetamine (AMPH) was significantly potentiated. VTA-BDNF overexpression also facilitates acquisition of cocaine self-administration, and a positive correlation between the level of VTA BDNF and drug intake during 12 hour binge was observed. We also found significant increase of DeltaFosB expression in the nucleus accumbens (NAc), the projection area of the VTA, in rats received intra-VTA BDNF overexpression. We therefore examined whether BDNF signaling in the NAc is important for social defeat stress-induced cross-sensitization by knockdown of the receptor of BDNF (neurotrophin tyrosine kinase receptor type 2, TrkB) there. NAc TrkB knockdown prevented social defeat stress-induced cross-sensitization to psychostimulant. Also social defeat stress-induced increase of DeltaFosB in the NAc was prevented by TrkB knockdown. Several other factors up-regulated by stress, such as the GluA1 subunit of Alpha-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) receptor and BDNF in the VTA were also prevented. We conclude that BDNF signaling in the VTA increases social defeat stress-induced vulnerability to psychostimulants, manifested as potentiated cross-sensitization/sensitization to AMPH and escalation of cocaine self-administration. Also BDNF signaling in the NAc is necessary for the stress-induced neuroadaptation and behavioral sensitization to psychostimulants. Therefore, TrkB in the NAc could be a therapeutic target to prevent stress-induced vulnerability to drugs of abuse in the future. DeltaFosB in the NAc shell could be a neural substrate underlying persistent cross-sensitization and augmented cocaine self-administration induced by social defeat stress.

Lors de mon travail de thèse, j'ai voulu explorer en détails les mécanismes cellulaires à l'origine de l'inversion de la plasticité due à la cocaïne par les mGluR. J'ai pu mettre en évidence que la sous-unité GluR2 des récepteurs AMPA joue un rôle crucial dans ce phénomène. De plus, il s'est avéré que le rôle joué par les mGluR in vitro est tout aussi critique in vivo. Cette capacité des mGluR a permis de révéler une autre caractéristique de la plasticité due à la cocaïne, à savoir que si les altérations au niveau de la VTA sont maintenues assez longtemps, elles entrainent alors une adaptation synaptique dans les régions ciblées, comme le noyau accumbens. Ces résultats contribuent ensemble à une meilleure compréhension des mécanismes cellulaires apparaissant dès les premières prises de drogues, et pouvant finalement mener à leur usage compulsif.
