The progress in protein and nucleic acid chemistry together with improvements of the previously employed tissue culture techniques have led to the solution of problems such as that of the generation of antibody diversity or of the molecular structure of T and B cell membrane receptor for antigen which had challenged the past generations of immunologists. Thanks to this progress an impressive amount of knowledge has been accumulated on certain cell types that were relatively "mysterious" until recently. The B lymphocyte represents a typical example of such a cell. With these considerations in mind, we have started to organize a NATO summer school on "The molecular basis of B cell differentation and function" that had the specific aim of bringing up to date a selected number of young investigators. During the course, held in Santa Margheri ta Ligure, October 1-11, 1985 it became apparent that rather than a formal school we had organized an informal series of Workshops where both the Faculty members and the participants were discussing their own data in addition to reviewing the general progress in the field. The deep commi ttment of everyone to his research subj ect and the enthusiasm had caused this very successful change in the shape of the course. At the end we asked the members of the Faculty and the participants to write a summary of their data and of their point of view on selected subj ect s.

Signaling through antigen receptor initiates a complex series of events resulting in the activation of genes that regulate the development, proliferation and differentiation of lymphocytes. During the past few years, rapid progress has been made in understanding the molecular basis of signaling pathways mediated by antigen and cytokine receptors. These pathways involve protein tyrosine kinases which are coupled to downstream regulatory molecules, including small guanine nucleotide binding proteins (e. g. p21'OS), serine threonine kinases (e. g. , members of the ERK family), and a large group of transcription factors. More recently, there have been breakthroughs in elucidating the genetic defects underlying three X-linked primary immunodeficiency diseases in humans. This volume surveys aspects of these rapidly developing areas of research. The book is divided into 5 different sections. Section I deals with signaling pathways in B lymphocytes. It includes a contemporary assessment of B cell antigen receptor structures, and discussion of the role of Ig-a/lg-B polypeptides in linking the antigen receptor to intracellular signal transduction pathways. The role of accessory molecules in the regulation of signaling by the B cell antigen receptor is also considered. Section II adopts a similar approach to the analysis of the antigen receptor on T lymphocytes. The importance of specialized signaling motifs in the CD3 polypeptides, mechanisms whereby these motifs may interact with the lymphocyte-specific protein tyrosine kinases, and the downstream consequences of these interactions are reviewed. In addition, the role of antigen-induced apoptosis in the generation of immunological tolerance is discussed.

Diabetes mellitus is rapidly increasing in prevalence throughout both developed and developing countries. The social and economic burden of this disease is estimated to cost 14 billion dollars worldwide. In the USA alone, 15 million individuals are diabetic, nearly half of them unaware of their condition. Complications of diabetes mellitus are the leading causes for blindness, limb amputation and chronic renal failure and kidney transplantation in industrialized countries. Further, diabetes mellitus per se and the metabolic derangement associated with diabetes are important risk factors for cardiovascular disease. Diabetes, as defined by an elevated fasting blood glucose level is presently subdivided in etiologically distinct groups. The most prevalent being type 2 (adult onset) diabetes characterized by insulin resistance and failure of the ~-cell to supply insulin in amounts sufficient to meet the body's needs. Type 1 (juvenile) diabetes, most commonly with an onset during childhood and adolescence, is caused by an auto-immune destruction of the pancreatic ~-cells. The causations of both type 1 and type 2 diabetes involve a combination of complex genetic traits and environmental influences. A third category are the mature onset diabetes of the young (MODY). This comparatively small group of patients (-10% of diabetes) presents relative early in life «30 years of age) compared to the more common late onset type 2 diabetes.
