Schild University College London, 1962 An intact immune response is essential for survival, as is evidenced by the various innate immune deficiency syndromes and by the emergence of the acquired immune deficiency syndrome (AIDS) as a pandemic during the last decade. Substances which stimulate the immune response might contribute to the therapy of AIDS and its precursor, AIDS-related syndrome, as well as of other clinical conditions in which immune responses can be diminished, such as carcinoma and infections. In other circumstances, an intact or heightened immune response may pose clinical problems; hence there is need to suppress, or diminish, components of the immune response. For instance, it is necessary to impair cellular immunity in order to ensure lasting acceptance of heterografts and it is already established that agents effective in transplantation are therapeutically effective in an range of autoimmune diseases. More recently, experimental studies have indicated that aberrant manifestations of humoral immunity, as in allergies, may also be amenable to pharmacological intervention.

Lymphocytes constitute the central cell type of the immune system. Over the last two decades, very powerful tools have become available with which to define their characteristics and functions. For example, monoclonal antibodies have allowed fine phenotypic characterization of various subpopulations of lymphocytes. The discovery of an ever-growing number of cytokines and growth factors, along with the structural elucidation of their receptors, was a result of refined bioassays, cloning and sequencing techniques. Lymphocytes, more than any other cell type, function in a network of cellular and humoral interactions. These humoral interactions include not only cytokines, but also hormones, neurotransmitters and lipid mediators. In addition, the manipulation of the immune system is effected by numerous exogenous substances with the properties of agonists, antagonists, metabolic inhibitors or biological response modifiers. It is in this context that this volume forms an essential part of the Handbook of Immunopharmacology series. The main focus of the book is on T lymphocytes and NK cells. The physiology and immunopharmacology of B cells, although not covered in a separate chapter, form an integral part of the sections on modulation by inflammatory mediators, hormones and neuropeptides. Signaling through antigen receptor initiates a complex series of events resulting in the activation of genes that regulate the development, proliferation and differentiation of lymphocytes. During the past few years, rapid progress has been made in understanding the molecular basis of signaling pathways mediated by antigen and cytokine receptors. These pathways involve protein tyrosine kinases which are coupled to downstream regulatory molecules, including small guanine nucleotide binding proteins (e. g. p21'OS), serine threonine kinases (e. g. , members of the ERK family), and a large group of transcription factors. More recently, there have been breakthroughs in elucidating the genetic defects underlying three X-linked primary immunodeficiency diseases in humans. This volume surveys aspects of these rapidly developing areas of research. The book is divided into 5 different sections. Section I deals with signaling pathways in B lymphocytes. It includes a contemporary assessment of B cell antigen receptor structures, and discussion of the role of Ig-a/lg-B polypeptides in linking the antigen receptor to intracellular signal transduction pathways. The role of accessory molecules in the regulation of signaling by the B cell antigen receptor is also considered. Section II adopts a similar approach to the analysis of the antigen receptor on T lymphocytes. The importance of specialized signaling motifs in the CD3 polypeptides, mechanisms whereby these motifs may interact with the lymphocyte-specific protein tyrosine kinases, and the downstream consequences of these interactions are reviewed. In addition, the role of antigen-induced apoptosis in the generation of immunological tolerance is discussed.

It is well established that asthma is an inflammatory disease of the airway mucosa and drugs like inhaled glucocorticoids are now commonly introduced early in therapy. A characteristic feature of this disease is the vast number of eosinophils in airway tissue, although many other migratory and resident inflammatory cells with the capacity to synthesize and release cytokines and putative asthma mediators are present in the inflamed mucosa.
