This SpringerBrief explores unconventional functions of eight different transcription factors and concludes with a discussion of their biological significance and impact, including effects on processes within the cell nucleaus during development and in adult organisms. Chapter One details unconventional functions of the transcription factors GAGA, HP1, Rb, STAT, ATF-2 and NF-kB. Surprisingly, all of these transcription factors can be found in association with heterochromatin as well as euchromatin, and in some cases unconventional functions have been demonstrated for these heterochromatin-associated factors. Chapter Two focuses on the unconventional functions of STAT and HP1 and discusses their roles in the promotion of longevity, and in protection from cancer and DNA damage. Chapter Three explores the biological significance of the findings presented in the first two chapters and considers how global changes in the epigenome brought about by factors such as STAT and HP1 might affect processes within the cell nucleus during development and in adult organisms. This succinct yet thorough SpringerBrief is essential for researchers studying epigenetics, and to instructors of the subject. It should also appeal to people interested in the control of gene transcription and other processes in the cell nucleus, and to those interested in development.

Several general principles have emerged from the study of human transcription factors. First, germline mutations in genes encoding transcription factors result in malformation syndromes in which the development of multiple body structures is affected. Second, somatic mutations involving many of the same genes contribute to tumorigenesis. Third, transcriptional regulatory mechanisms demonstrate remarkable evolutionary conservation. Fourth, prenatal development and postnatal physiology are unified by the demonstration that a single transription factor can control the proliferation of progenitor cells during development and the expression within the differentiated cells of gene products that participate in specific physiologic responses. Transcription Factors and Human Disease presents the basic science of transcriptional regulation and then describes inherited human diseases attributable to mutations in DNA sequences encoding transcription factors or their cognate binding sites. The involvement of transcription factors in somatic cell genetic diseases (cancer) and epigenetic disease (teratogenesis) is briefly discussed. The effect of specific mutations on transcription factor activity and the relationship between transcriptional dysregulation, dominant or recessive inheritance patterns, and disease pathogenesis are also explored. This book thus provides a direct connection between molecular defects in transcriptional regulation and human pathophysiology.

Transcription factors are important in regulating gene expression, and their analysis is of paramount interest to molecular biologists studying this area. This book looks at the basic machinery of the cell involved in transcription in eukaryotes and factors that control transcription in eukaryotic cells. It examines the regulatory systems that modulate gene expression in all cells,a s well as the more specialized systems that regulate localized gene expression throughout the mammalian organism. Transcription Factors updates classical knowledge with recent advances to provide a full and comprehensive coverage of the field for postgraduates and researchers in molecular biology involved in the study of gene regulation.

In the last few years, significant breakthroughs in transcription research expanded our appreciation for the complexity of molecular controls on gene expression in mammalian cells. In Transcription Factors: Methods and Protocols, experts in the field describe state-of-the-art approaches that investigators can use to probe critical mechanisms underlying transcription factor nuclear-cytoplasmic trafficking as well as to assess the functional impact of post-translational modifications on transcription factor function. The chapters are written by prominent scientists, many of whom developed these methods, and highlight protocols that focus on specific transcription factor family members with particular relevance to human disease. Composed in the highly successful Methods in Molecular BiologyTM series format, each chapter contains a brief introduction, step-by-step methods, a list of necessary materials, and a Notes section which shares tips on troubleshooting and avoiding known pitfalls. Comprehensive and current, Transcription Factors: Methods and Protocols compiles the latest techniques for elucidating controls on transcription factor intracellular localization and activity, and consequently is unlike any other methods-based text on transcriptional regulation today.

Acclaimed biologist Lewis Wolpert eloquently narrates the basics of human life through the lens of its smallest component: the cell. Everything about our existence-movement and memory, imagination and reproduction, birth, and ultimately death-is governed by our cells. They are the basis of all life in the universe, from bacteria to the most complex animals. In the tradition of the classic Lives of a Cell, but with the benefit of the latest research, Lewis Wolpert demonstrates how human life grows from a single cell into a body, an incredibly complex society of billions of cells. Wolpert goes on to examine the science behind topics that are much discussed but rarely understood—stem-cell research, cloning, DNA, cancer—and explains how all life on earth evolved from just one cell. Lively and passionate, this is an accessible guide to understanding the human body and life itself.

How do we move, think and remember? Why do we get ill, age and die? Distinguished biologist Lewis Wolpert explains how cells provide the answers to the fundamental questions about our lives. Cells are the basis of all life in the universe. Our bodies are made up of billions of them: an incredibly complex society that governs everything, from movement to memory and imagination. When we age, it is because our cells slow down; when we get ill, it is because our cells mutate or stop working. In How We Live and Why We Die, Wolpert provides a clear explanation of the science that underpins our lives. He explains how our bodies function and how we derive from a single cell - the egg. He examines the science behind the topics that are much discussed but rarely understood - stem-cell research, cloning, DNA - and explains how all life evolved from just one cell. Lively and passionate, How We Live and Why We Die is an accessible guide to understanding the human body and, essentially, life itself.
