Annual Plant Reviews, Volume 5 Our concept of the vacuolar compartment in plant cells has changed radically in recent years. We now know that the traditional view of a vacuole as one multifunctional organelle is incorrect, and that most plant cells contain basically two types of vacuoles - ltyic and protein storage vacuoles. These are served by separate vesicular pathways, which defines them as distinct organelles. Furthermore, it appears that the functions of each type can be modified such that several different vacuoles, each with a somewhat different function, can exist in one cell. This volume examines our current understanding of vacuolar compartments. It is directed at academic and industrial researchers in plant cell biology, biochemistry, physiology, molecular biology and developmental biology.

The papers in this book, illustating the present status of knowledge related to the vacuolar compartment of fungi and plants, were presented at an Advanced Research Workshop entitled "Plant Vacuoles. Their Importance in Plant Cell Compartmentation and their Applications in Biotechnology" held in Sophia-Antipolis, France, on July 6-11, 1986. The organizers were fortunate in being able to assemble representative leaders of all the above fields of research concerning this compartment. These scientists from all over the world were invited to present their latest results and to exchange views and plans for continued research in this highly exciting field, which is very important for the improving the industrial aspect of plant biotechnology related to the production of molecules with a high added value. To ensure maximal flexibility and opportunity for dicussion, the chairmen of the various sessions were asked to introduce their respective topics, after which they were given the freedom of organizing their sessions in a more or less improvised fashion, with brief presentations of relevant new information by participants following a schedule fixed relatively shortly before the workshop. This procedure proved to be highly successful, giving rise to most productive and stimulating discussions among the representatives of the various fields.

The yeast S.cerevisiae is not an efficient tool for in vivo studies of plant vacuolar sorting receptors Plant and yeast vacuoles, the equivalents of mammalian lysosomes, are acidic compartments involved in hydrolytic functions. They are also essential for metabolite storage and for maintaining cytosolic ion and pH homeostasis (Klionsky and Emr, 1990; Wink, 1993). The vacuoles mainly receive proteins and lipids from the biosynthetic and endocytic vesicular transport pathways. Newly synthesized vacuolar proteins transit through the early compartments of the secretory pathway and are actively sorted away from secreted proteins in the trans-Golgi network (TGN) before being delivered via prevacuolar compartments (PVC) to the vacuoles. In contrast to yeast, the plant vacuolar sorting machinery is extremely complex, since some plant cells may have up to three functionally distinct vacuoles: the lytic vacuole, the storage vacuole (Hoh et al., 1995; Paris et al., 1996) and the neutral vacuole (Di Sansebastiano et al, 1998). Transport analysis of soluble vacuolar proteins in plants identified three classes of vacuolar sorting determinants (VSDs), which likely interact with specific vacuolar sorting receptors (VSR). It is assumed that this interaction allows the VSRs to direct their specific ligands to the right vacuole. Indeed, protein transport to the lytic vacuole has been shown to depend on sequence-specific VSDs (ssVSD) and is mediated by clathrin-coated vesicles (CCVs). Transport to the neutral and (seed) storage vacuoles requires the C-terminal VSD (ctVSD) and the structural type VSD (psVSD), respectively (reviewed in Neuhaus and Rogers, 1998). VSRPS-1 (previously named BP-80) is the first vacuolar sorting receptor identified in plants. VSRPS-1 was originally isolated from pea CCVs by its ability to bind to the ssVSD from barley proaleurain in a pH-sensitive manner in an in vitro assay (Kirsch et al., 1994). Since then, several VSRs were cloned from different plant spec.

Early approaches towards an understanding of transport at the tonoplast of higher plant cells were based on the kinetics of radio-isotope uptake (Torii and Laties. 1966) and exchange (Pitman, 1963). A variety of other methods became available in due course (reviewed by Clarkson and Luttge, 1984). But even with the most modern developments, the function of. the tonoplast remained in the dark. On the basis of Hitchell's chemiosmotic $heory (Hitchell, 1967), mechanisms of membrane-bound H -translocating ATPases and H -solute co-transport were conceived for the plasmalemma of fungi and plants, based on experiments with intact tissues, cells or protoplasts . With the onset of the era of membrane fractionation it seemed obvious that plasmalemma and tonoplast vlould cause the greatest problems due to the apparent lack of intrinsic markers. Again the situation for the plasmalemma appeared to be favorable; the periodic-phosphotungstic-acid reagent was considered as a specific stain. There was also the opportunity to bind externally applied markers to the plasmalemma prior to cell homogenization. Vacuoles were initially considered as compartments for intracellular excretion or at best as storage compart­ ments, but were later understood to have important dynamic functions in cell physiology as a lytic compart­ ment comparable to animal lysosomes (Hatile, 1966). The dynamics of molecular functions of the tonoplast itself remained obscure, however, with one exception. Hotivated by the search for a better understanding of latex production by the rubber tree Hevea brasiliensis Hull. -Arg.
