Viral Transformation and Endogenous Viruses is a collection of papers presented at the symposium on ""Viral Transformation and Endogenous Viruses"" held at Vanderbilt University on April 1-2, 1974. Contributors discuss the viral function(s) responsible for the transformation of the cells that are infected with oncogenic viruses, emphasizing the type of changes that characterize transformed cells and the regulatory mechanisms that are altered after malignant transformations. This volume is organized into 10 chapters and begins with an overview of DNA-containing tumor viruses, particularly Simian Virus 40 (SV40) and its mutants with DNA deletions, insertions, and duplications. The integration and transcription of adenovirus DNA is considered, along with the characteristics of temperature-sensitive mutants of these viruses. The reader is then introduced to the chemistry and biology of RNA-containing tumor viruses, which are useful reagents to study neoplastic transformation both in vivo and in vitro. A major aspect of these viruses concerns the analysis of their genome at the chemical and genetic levels. The last part of the book is devoted to biochemical and genetic analyses of endogenous viruses isolated from avian cells. This book is a valuable resource for scientists and investigators in fields such as pathology, molecular virology, molecular biology, microbiology, oncology, and biochemistry.

All but one* of the following articles represent comprehensive reports on a workshop held between 7 and 9 May 1981 at the Institute of Virology and Immunobiology, University of Wfuzburg, Federal Republic of Germany. The title of the workshop was "The Involvement of Endogenous Retroviruses inN ormalFunction and Pathological Growth of Lymphocytes." Rather than collecting and printing manuscripts of the individual communications, the organizers asked selected parti cipants to write, after the workshop, concise articles each compris ing several contributions and discussions on major topics. In so doing, we hope to present to a larger audience a synopsis of the various information and views exchanged at the meeting. Such a procedure seemed the more appropriate as the workshop was intended to bring together specialists from two rather diverse fields: RNA-tumor virology and immunobiology. While this created some initial problems of terminology, it was quite effective in making representatives of one field more aware of the significance and the contributions ofthe other. It also great ly contributed to realization of the complexity of the problems involved in virus-induced leukemogenesis.

l RNA tumor viruses have become increasingly utilized in studies of cellular transformation and gene regulation. The genes of retroviruses exist in two forms; as extrachromosomal, RNA-containing, infectious particles and as DNA pro 2 viruses stably associated with cell genes. Components from the extracellular form can be collected in large quantity and purified for the preparation of molec ular probes. These probes can be used to dissect the sequence of events required for the establishment and expression of the integrated form. Furthermore the 2 genomes of retroviruses originated from normal cell genes, genes called virogenes • The nucleic acid and protein probes isolated from these viruses are therefore useful for studying the nature and expression of this normal cell gene and in elucidating the physiological role of its products. RNA tumor viruses perhaps offer us one of the most complete sets of biochemical reagents and biological responses for examining gene regulation in vertebrates and for studying the consequences of aberrant gene regulation on cell growth in tissue culture and in animals. Further more, there is an increasing conviction that virogenes play an important role in normal development and/or differentiation (RISSER, STOCKERT and OLD, 1978). Consequently, there is a growing feeling that DNA proviruses are altered viro genes and are capable of interfering with normal development or differentiation, causing reprogrammed growth or the incapacity to specialize.

1. 1 Scope of the Review This review was intended initially as a reference source for those interested in the origins and fITst descriptions ofthe defective avian sarcoma viruses. Quite a few of these viruses have been characterized in the past few years and their varied nomenclature according to source, discoverer, date of isolation or biological properties could result in some con fusion among those attempting to follow the literature. Information will be included on the molecular biology of the sarcoma viruses, rather more of which is available than when the review was fITst conceived, although in this respect the review will inevitably be out of date by the time of publication. If any bias of content is introduced, this will be towards a more detailed coverage of the author's own area of interest, the gene products of the defective sarcoma viruses. Rous sarcoma virus (RSV) serves as the model for much of this work and will frequently be referred to for comparative purposes, as will the mammalian defective transforming viruses. Recent reviews provide more complete coverage of these topics (l-4a). This review is complemented by a discussion ofthe avian acute leukaemia viruses, which appears elsewhere in this volume [5]. As will be seen, concentrating primarily on the defective sarcoma viruses and comparing them to RSV can be justified in terms of their biochemical properties as well as their similar biology.

For decades retroviruses have been riding the crest of a wave of experimental research directed toward the identification of an infectious agent of human neoplastic diseases. In the early 1970s, several scientists successfully demonstrated the presence of retroviruses in numerous animal species and proved their etiological role in some related diseases. Corresponding findings in humans were somewhat discouraging. Although financial support for this line· of research declined, a few dedicated retrovirologists survived and continued to collect more biological information and technological expertise that opened a new approach to the search for a human retrovirus. The rewards came with the discovery that the genes responsible for neoplastic transformation (oncogenes) are of cellular origin and can be shuttled about by retroviruses, and with the identification of a new family of Human T-cell Lymphotrophic retroViruses (HTLV) from patients with diseases ranging from leukemia to the acquired immunodeficiency syndrome (AIDS). An understanding of the role and significance of retroviruses in human pathology requires basic knowledge of the major animal systems studied. With this perspective in mind, we present here a survey that includes general overviews, minireviews on each animal system studied with selected experimental reports and, finally, a stimulating review of the field of human retrovirology by many of the pioneer scientists who created it.
