In comparison with antibacterial chemotherapy the clinical use of specific antiviral drugs is a fairly recent development. By the 1950s a number had been discovered, but their clinical application was delayed until 1962, when methisazone was used for the treatment of infective complications of smallpox vaccination and idoxuridine in the treatment of herpetic keratitis. In retrospect the reasons for this slowness in development is now evident. It was due in large part to the prevailing view held at the time that virus infections could never be treated with specific agents, since the multiplica tion of virus is so closely integrated with the metabolic processes of the host cell that specificity of action seemed impossible of achievement. In contrast, bacteria were independent entities with metabolic systems differing widely from those of the mammalian host, which afforded points of attack by specific agents which had no action upon mammalian systems. Although the mode of action of the sulphonamides and penicillin was not known for some time, they afforded perfect examples of the success of this mode of attack.

It is now widely acknowledged that at the beginning of this century Claude von Pirquet first pointed out that a viral disease, i. e. , measles, resulted in an anergy or depression of preexisting immune response, namely, delayed continuous hypersensitivity to PPD derived from Mycobacterium tuberculosis. Thereafter ob servations that viral infections may result in immunosuppression have been recorded by many clinicians and infectious disease investigators for six or seven decades. Nevertheless, despite sporadic reports that infectious diseases caused by viruses may result in either transient or prolonged immunodepression, investigation of this phenomenon languished until the mid-1960s, when it was pointed out that a number of experimental retroviral infections of mice with tumor viruses may result in marked immunosuppression. However, it was not until the recognition of the new epidemic of acquired immunodeficiency syn drome (AIDS) caused by the human immunodeficiency virus and related vi ruses that acquired immunodeficiencies associated with virus infection became general knowledge among biomedical investigators as well as the lay public. A number of reviews published during the past decade or so pointed out that numerous viruses may affect humoral and cellular immune responses. Furthermore, expanding knowledge about the nature and mechanisms of both humoral and cellular immunity and pathogenesis of viral infections has pro vided clinical and experimental models for investigating in depth how and why viruses of man and animals profoundly affect immune responses.

Foot-and-mouth disease (FMD) has been recognized in printed records dating from the sixteenth century, and since the eradication of rinderpest (cattle plague) in the early part of the twentieth century it has been rec- nized as the most important and feared disease of cattle and other dom- tic livestock. The beginning of the twenty-first century brought the worst outbreak of FMD ever experienced in England, which had been completely free of the disease for 33 years. This tragic epidemic, which spread to Northern Ireland, Scotland, France and the Netherlands with severe e- nomic consequences, emphasized the need for further research into better methods for the detection and control of the disease. FMD is caused by a small RNA virus which is highly contagious and can survive in meat and other animal products for long periods at normal pH levels. The virus typically infects cloven-hoofed animals, including c- tle, goats, pigs and sheep, as well as a wide range of non-domesticated a- mals in regions of the world where FMD virus is endemic, such as the Af- can continent. There are seven recognized serotypes of FMD virus, with numerous subtypes, and as a consequence vaccine production and administration is complex and a major debate surrounds every disease outbreak regarding the relative merits of vaccination as opposed to the slaughter of all infected animals.
