This Thesis presents the development and characterisation of a novel wide field, time-correlated single photon technique for luminescence lifetime imaging, and its possible use for life sciences imaging. -- The primary device which was developed is based on a 3-stage image intensifier coupled with a high speed CMOS camera, mounted on a conventional microscope. The post-acquisition image processing software developed for this purpose, in¬volving photon detection and centroiding algorithms, is described and discussed. Lifetime images of artificial ruby and europium compounds are reported, as well as other applications of single photon counting imaging for fluorescence microscopy. Another wide-field TCSPC technique, based on an Electron-Bombarded CCD de¬tector, is presented, along with the characterisation of the device, and its uses as a single photon counting detector for wide-field microscopy. -- This work also presents results of Fluorescence Lifetime Imaging Microscopy and Time-Resolved Anisotropy measurements of biological samples on a commercial laser scanning microscope system. -- Finally, this work presents suggestions of improvement and further developments on these wide-field TCSPC detectors.

Fluorescence lifetime imaging microscopy (FLIM) is a key technique to image cells as, in addition to the advantages of standard fluorescence microscopy, it allows to study the environment and probe interaction in living specimens. Implementing FLIM via Single Photon Counting (SPC) proved to be the most effective technique considering the fiuorophores limited photon budget before being irreversibly bleached. This thesis focuses on the development and application of Single Photon Counting techniques to imaging systems and to spectroscopy. , Firstly, the BODIPY-C12 molecular rotor was used to determine dyeconcentrations between 3 and 16 ~tM for living cells and lipid droplets via FLIM and intensity measurements A novel compound, named ET, was tested for the first time on living cells and its possible applications as a molecular rotor discussed. The use of an Electron Bombarded Charge-Coupled Device (EBCCD) camera as a parallel-processing Time to Amplitude Converter device for SPC Imaging with sub-frame exposure time resolution was investigated and, although not implemented, the results supports the proposed method. In order to design a wide field time-correlated single photon counting system, a Complementary Metal-Oxide Semiconductor (CMOS) FastCamera was coupled with an 3-stage image intensifier. This proved to be suitable for luminescence lifetime measurements of a Ruthenium complex, with results of 1. 7~ts comparable with confocal scanning Time-Correlated SPC (TCSPC). Finally, conclusions and future work are discussed.

In 1984 Desmond O’Connor and David Phillips published their comprehensive book „Time-correlated Single Photon Counting“. At that time time-correlated s- gle photon counting, or TCSPC, was used primarily to record fluorescence decay functions of dye solutions in cuvettes. From the beginning, TCSPC was an am- ingly sensitive and accurate technique with excellent time-resolution. However, acquisition times were relatively slow due to the low repetition rate of the light sources and the limited speed of the electronics of the 70s and early 80s. Moreover, TCSPC was intrinsically one-dimensional, i.e. limited to the recording of the wa- form of a periodic light signal. Even with these limitations, it was a wonderful te- nique. More than 20 years have elapsed, and electronics and laser techniques have made impressive progress. The number of transistors on a single chip has approximately doubled every 18 months, resulting in a more than 1,000-fold increase in compl- ity and speed. The repetition rate and power of pulsed light sources have increased by about the same factor.

Fluorescence methods are being used increasingly in biochemical, medical, and chemical research. This is because of the inherent sensitivity of this technique. and the favorable time scale of the phenomenon of fluorescence. 8 Fluorescence emission occurs about 10- sec (10 nsec) after light absorp tion. During this period of time a wide range of molecular processes can occur, and these can effect the spectral characteristics of the fluorescent compound. This combination of sensitivity and a favorable time scale allows fluorescence methods to be generally useful for studies of proteins and membranes and their interactions with other macromolecules. This book describes the fundamental aspects of fluorescence. and the biochemical applications of this methodology. Each chapter starts with the -theoreticalbasis of each phenomenon of fluorescence, followed by examples which illustrate the use of the phenomenon in the study of biochemical problems. The book contains numerous figures. It is felt that such graphical presentations contribute to pleasurable reading and increased understand ing. Separate chapters are devoted to fluorescence polarization, lifetimes, quenching, energy transfer, solvent effects, and excited state reactions.
