During the past decade, great strides have been made in our un derstanding of the biochemistry and pharmacology of the alpha-l adrenergic receptor. The alpha-l adrenergic receptor plays a key role in biological function. This is evidenced by the fact that the alpha-l adrenergic receptor plays a prominent functional role in most organs of the body and in the key systems responsible for survival of the organism and maintenance of optimum biological activity. This is most apparent in the cardiovascular system, in which alpha-l adrenergic receptors are the single most important receptor involved in the maintenance of blood pressure and circu latory function. It is appropriate, therefore, that recent findings related to the pharmacology and biochemistry of the alpha-l adrenergic receptor be compiled, since this subject has not been reviewed in detail in recent years. It is the purpose of this book to present a series of reviews of key experimental findings that shed new light on the alpha-l adrenergic receptor and the manner in which it functions. Classically, most receptors have been characterized based on structure-activity relationships obtained for selective agonists and antagonists interacting with the receptor. Although there are many newer and more sophisticated approaches to receptor char acterization, structure-activity relationships still provide impor tant information regarding the chemical requirements made by the receptor for its occupation by ligands and its subsequent acti vation by those ligands possessing intrinsic efficacy and, there fore, agonist activity.

Experimental evidence indicates that catecholamines are able to modulate information processing in the amygdaloid complex. The dense catecholaminergic innervation of the amygdala may thus exert significant influence upon its well-documented functions in emotion, attention, learning and memory. Dysfunctions of the systems have been suggested to be involved in the etiology of neuropsychiatric disorders including senile dementia, Parkinson's disease and schizophrenia. To further insight into neural mechanisms of the innervation, the present study provides a detailed documentation of ultrastructural and neurochemical characteristics of dopanminergic, noradrinergic and adrenergic afferent fibers and of their target structures in central, basal and intercalated amygdaloid nuclei. A high degree of heterogeneitiy is observed between the nuclei concerning the different catecholaminergic innervation patterns and the peptide content of individual catecholaminergic fibers, as well as concerning the neurotransmitter/-modulator and receptor expression of possible amygdaloid target neurons. The results are discussed with regard to previously documented properties of the nuclei.

Not much more than a decade has passed since the appearance of the outstanding handbook, Catecholamines, edited by BLASCHKO and MUSCHOLL, in the series: Handbook of Experimental Pharmacology. However, this extremely well organized volume dealt mainly with the origin, molecular actions, and fate of the naturally occuring catecholamines. It was felt that a separate volume should be dedicated to the remarkable and exciting progress made in the field of agents influencing the adrenergic system, both in physiologic and pharmacologic respect. The editor of the present volume considers himself lucky to have been able to persuade a number of eminent specialists to collaborate. The main concept of the present handbook is a systematic approach to the various effects of adrenergic activators and inhibitors starting with the chemistry and structure actitivity relationship, followed by the evaluation of adrenergic activators and inhibitors, and discussing their mode of action. The most voluminous part is the chapter dealing with the systemic pharma cology of these agents analyzing the effects on the central nervous system, on the autonomic nervous system, on the cardiovascular, the respiratory, the digestive, the endocrine system, on the skeletal muscle, and on metabolism. Kinetics and bio transformation, further toxic effects are discussed in the following chapters. A special chapter on clinical features concludes the monograph.
