Neurodevelopmental disorders are generally influenced by genetic as well as intrauterine and extrauterine factors that affect the fetal-maternal environment and/or brain development that continues after birth. Specific genetic polymorphisms may increase susceptibility to environmental factors that alter the trajectory of brain development via diverse molecular mechanisms. In particular, the pre- and post-natal exposure to neurotoxic metals, pesticides, persistent organic pollutants, and other chemicals is increasingly recognized as involved in the pathogenesis of neurodevelopmental disorders, such as autism, deficiency attention/hyperactivity disorders, neurodegenerative disorders such as Parkinson's and Alzheimer's disease, chronic multiple sclerosis, and even fetal and infant death, including SIUDS (Sudden Unexplained Intrauterine Death Syndrome) and SIDS (Sudden Infant Death Syndrome).

This eighth and final report of the Immunization Safety Review Committee examines the hypothesis that vaccines, specifically the measles-mumps-rubella (MMR) vaccine and thimerosal-containing vaccines, are causally associated with autism. The committee reviewed the extant published and unpublished epidemiological studies regarding causality and studies of potential biologic mechanisms by which these immunizations might cause autism. Immunization Safety Review: Vaccines and Autism finds that the body of epidemiological evidence favors rejection of a causal relationship between thimerosal-containing vaccines and autism. The book further finds that potential biological mechanisms for vaccine-induced autism that have been generated to date are only theoretical. It recommends a public health response that fully supports an array of vaccine safety activities and recommends that available funding for autism research be channeled to the most promising areas. As the average life expectancy of many populations throughout the world increases, so to does the incidence of such age-related neurodegenerative disorders as Alzheimer's, Parkinson's, and Huntington's diseases. Rapid advances in our understanding of the molecular genetics and environmental factors that either cause or increase risk for age-related neurodegenerative disor ders have been made in the past decade. The ability to evaluate, at the cellular and molecular level, abnormalities in postmortem brain tissue from patients, when taken together with the development of valuable animal and cell-culture models of neurodegenerative disorders has allowed the identification of sequences of events within neurons that result in their demise in specific neurodegenerative disorders.
