Antifreeze proteins (AFPs) have the characteristics of inhibiting ice growth and recrystallization by binding to the surfaces of ice crystals. There are findings to support the use of AFPs as a potential cryoprotectant. Lactate dehydrogenase (LDH) is an enzyme found in almost all living cells catalyzing the conversation of lactate to pyruvate (and back). LDH is often used as a model enzyme for research and a biomarker for cell or tissue damages by various diseases. LDH denatures and loses its catalytic activity under harsh conditions. Polyols, carbohydrates, and certain polymers have been demonstrated to protect the activity of LDH under repeated freeze thaw cycles. In this study, we investigate the effect of a hyperactive beetle AFP from Dendroides Canadensis (DAFP-1) on cryoprotection of LDH. This study shows that DAFP-1 can protect catalytic activity of LDH under repeated freeze thaw cycles. Concentrations as low as 0.05 microg/ml of DAFP-1 were sufficient enough to provide protection of LDH activity during freezing and thawing. There was no significant difference observed in LDH activity when comparing different freeze thaw rates. High performance liquid chromatography (HPLC) and circular dichroism (CD) spectroscopy studies suggest possible interactions between LDH and DAFP-1. The experiments of LDH protection by a previously made DAFP-1 mutant, where all the arginine residues were replaced by alanines, suggest that arginine residues of DAFP-1 are important for LDH protection. The results suggest that DAFP-1 may interact with LDH through arginines and protect the active site of LDH. This study also suggests the cryogenic potential of AFPs for biopharmaceuticals, cells and organs.
