Antigen processing and presentation, as a field, explores a broad range of protein interactions and functions, both intracellular (in the cytoplasm and in the endoplasmic reticulum) and at the cell surface (between T cells and MHC molecules). To investigate such a diverse array, it is necessary that biochemical, cell biology, and immunological techniques all be employed. The purpose of Antigen Processing and Presentation Protocols is therefore to detail the most up-to-date techniques being used in this burgeoning field. Such techniques include those used to question how MHC-binding peptides are generated, to test how peptides are delivered to MHC molecules, to analyze MHC peptide-binding patterns, and to assay the T-cell response to MHC/peptide. Antigen Processing and Presentation Protocols should aid both those new and those experienced in this area of research in extending the questions that can be asked and answered by the application of these current methods. For editorial assistance, I would like to thank Angela Beninga and Rachael Turnquist.

This book provides a state-of-the-art review of the processing, presentation, and subsequent recognition of antigens by T cells. Topics discussed include the structure of MHC molecules, the nature and specificity of human class II molecule interactions with peptide antigens, the class II invariant chain, antigen processing and presentation by class I MHC molecules, the biology of antigen processing, and the presentation and role of accessory molecules in T cell recognition. Other chapters feature discussions about the T cell allorecognition of MHC molecules, the recognition of minor antigens, and the concept of "superantigens." Color plates demonstrate the three-dimensional structure of MHC molecules and peptide antigen interaction with MHC molecules. Diagrams illustrate antigen presentation pathways, T cell receptor-MHC interaction and accessory molecule interactions. Antigen Processing and Recognition will be a valuable addition to the libraries of students and teachers of immunology, as well as cell biologists who are looking for a unified view of this rapidly expanding subject.

Antigen presentation is central to the immune response, and is instrumental in ensuring that the response mounted is that best suited to the eradication of the particular microbe faced. In this volume, experts in the field provide state-of-the-art descriptions of the antigen presentation pathways. How do viruses disrupt these critical pathways, and to what effect? Do all tissues present antigen in the same way? If not, why? What are the consequences of dysfunctional antigen presentation, seen in certain genetic disorders? This book considers not only the molecular details, but also their relevance to the whole organism.

Processing And Presentation Of Antigens written by Benvenuto Pernis and has been published by Elsevier this book supported file pdf, txt, epub, kindle and other format this book has been release on 2012-12-02 with Health & Fitness categories.

Processing and Presentation of Antigens brings together the views of immunologists, biochemists, cell biologists, and virologists who have a common interest in the early phases of the immune response. Antigen processing and presentation comprise a series of events that are much more complex and sophisticated than had been thought for a long time, and constitute an essential part of the biology of the immune response to T-dependent protein antigens. The book is organized into seven parts. Part I discusses the major advances and insights in antigen processing, some results on the interaction of antigen with class II MHC molecules, and the precise mechanism involved in antigen processing. Part II presents papers on endosomes, lysosomes, and recycling. Part III deals with studies on presentation in the context of class I MHC molecules. The chapters in Part IV examine interactions of antigens with class II MHC molecules. Part V contains papers on macrophages and dendritic cells as accessory cells. Part VI examines antigen presentation by B cells. Part VII covers studies on T cells.

The activation of adaptive immune responses requires the processing and presentation of protein antigens to lymphocytes. Especially dendritic cells are effective at display of antigen-derived peptides in the form of immunogenic peptide/MHC complexes to CD4 and CD8-positive T cells, and can stimulate even naive T cells to clonally expand. During the last 40 years, mechanisms that facilitate antigen processing and presentation were clarified, mostly from work in cell lines and mouse models. From mouse-based work, it is now clear that dendritic cells represent a collection of specialized cell subsets that are particularly well endowed to stimulate antigen transport to distinct tissue locations, to transfer antigens between cellular subsets or to trigger T cell responses. Dendritic cell subsets hold great promise for therapeutic application, for example as dendritic cell-based vaccines to bolster immune responses against viruses or malignant growths. Hurdles remain that preclude the efficient application of high quality pre-clinical research into standardized patient care. In this research topic, efforts in dendritic cell research and dendritic cell-based vaccines are discussed, from both pre-clinical and application points of view.
