Since the initial report of the amplification of specific DNA fragments using the polymerase chain reaction (PCR) in 1985, this technique has revolutionized molecular biology. It enables the production of large quantities of DNA from minute amounts of sample material, which can then be readily analyzed. This facility has had an enormous influence on the way both fundamental and diagnostic questions are approached and its use is now considered essential for molecular work in all branches of biology. The purpose of this book is to highlight the wide-ranging applications of PCR in pure and applied mycology and to increase understanding of its potential benefits. After a brief overview, a group of internationally-renowned mycologists give definitive descriptions of the use of PCR in their own specialized fields. These include fungal gene expression and cloning, taxonomy and speciation, fungal mycobionts, mycorrhizal fungi, entomopathogenic fungi, mycotoxin-producing fungi, diagnosis of fungal infections in animals, seed-borne diseases, fungal/plant interactions and applications with industrially-important fungi. The Kingdom fungi encompass a massive diversity of taxa with wide-ranging ecologies, life cycles, and morphologies ranging from unicellular aquatic chytrids to large mushrooms. Before molecular methods came in existence, taxonomists considered this Kingdom to be a member of the plant kingdom due to certain life styles like immobility and growth habitats. Molecular markers (also known as DNA markers), facilitated a better alternative method over traditional morphological methods, employed for the identification, characterization, and to understand the evolution of fungi. The morphological methods used for identification are mainly dependent on spore color or microscopic features whereas molecular markers are based on DNA polymorphism in the genomic organization. Phylogenetic studies reported in last decade, based on molecular markers, have reshaped the classification system of Kingdom fungi, which divided into one subkingdom, seven phyla, and ten subphyla. Recent advances in molecular mycology have opened the way for researchers to identify and characterize novel fungal species from unique environments. Mycology is concerned with the systematic study of fungi, including their genetic and biochemical properties, their use to humans as a source of medicine and food, as well as their dangers, such as poisoning and infections. In the 21st century with the development of DNA sequencing technologies and phylogenetic analysis based on molecular markers, new insights into fungal taxonomy were provided. This book contains a thorough discussion of molecular characterization and detection of different groups of fungi by using PCR-based markers and provides a comprehensive view of the applications and uses of different molecular markers in molecular mycology. It also addresses the recent molecular markers employed to solve the problems of identification and discusses current approaches used in molecular characterization and detection of fungi.

In the last decade, enormous progress has been made on the physiology of plant roots, including on a wide range of molecular aspects. Much of that progress has been captured in the chapters of this book. Breakthroughs have been made possible through integration of molecular and whole-plant aspects. The classical boundaries between physiology, biochemistry and molecular biology have vanished. There has been a strong focus on a limited number of model species, including Arabidopsis thaliana. That focus has allowed greater insight into the significance of specific genes for plant development and functioning. However, many species are very different from A. thaliana, in that they are mycorrhizal, develop a symbiosis with N2-fixing microsymbionts, or have other specialised root structures. Also, some have a much greater capacity to resist extreme environments, such as soil acidity, salinity, flooding or heavy-metal toxicities, due to specific adaptations. Research on species other than A. thaliana is therefore pivotal, to develop new knowledge in plant sciences in a comprehensive manner. This fundamental new knowledge can be the basis for important applications in, e.g., agriculture and plant conservation. Although significant progress has been made, much remains to be learnt. It is envisaged that discoveries made in the recent past will likely lead to major breakthroughs in the next decade.
