This book was written in the context of the daily confrontation with problems in the utilization of polymeric supports for the synthesis of pep tides. Therefore, views and experiences which usually are not mentioned in scientific journals are collected in these pages. The author has deliberately discussed in detail the possible influence of the polymer phase on the varying reaction conditions in the Merrifield synthesis; this aspect is neglected in most publications dealing with peptide synthesis. However, in view of the growing body of information on the chemistry of polymer-supported peptide syntheses, the international readership should regard the author's arguments as open to discussion. I am very much indebted to all of my colleagues with whom I have had the opportunity to cooperate in studying the potential of the Merrifield synthesis. Above all I like to express my gratitude to my teacher, Professor Dr. Theodor Wieland, Heidelberg, for his boundless encouragement and support in my ef forts in the field of peptide synthesis, particularly in its polymer phase bound version. During the years 1980-81, as guests of the Deutsches Woll forschungsinstitut in Aachen, Germany, we were working on a small book entitled, "Principles of Peptide Synthesis". In the library of the Institute we noted that the volumes of Houben-Weyl's Handbuch der Organischen Chemie dealing with peptide synthesis were so much in use that they were ready to fall apart because the researchers of the Institute consulted them with amazing regularity. They were looking for references, but even more for experimental details which could be adapted to the particular problem they happened to face. In planning a new synthetic endeavor they tried to lean on the experience of others in analogous situations. This suggested to us that a smaller and hence more tractable book may be needed, a volume which can be kept on or near the bench to make examples of funda mental methods readily available in the laboratory. Such a collection could save numerous short trips to the library, a point particularly important where a library well equipped with the sources of the literature of peptide synthesis is not near at hand. Also, we thought that the envisaged book may be welcome by those who are more versed in English than in German. To our best knowledge no similar publi cation is available.

In the years since the publication of Atherton and Sheppard's volume, the technique of Fmoc solid-phase peptide synthesis has matured considerably and is now the standard approach for the routine production of peptides. The basic problems at the time of publication of this earlier work have now for the most part, been solved. As a result, innovators in the field have focussed their efforts to develop methodologies and chemistry for the synthesis of more complex structures. The focus of this new volume is much broader, and covers the essential procedures for the production of linear peptides and more advanced techniques for preparing cyclic, side-chain modified, phospho- and glycopeptides. Many other methods also deserving attention have been included: convergent peptide synthesis; peptide-protein conjugation; chemoselective ligation; and chemoselective purification. The difficult preparation of cysteine and methionine-containing peptides is also covered, as well as methods for overcoming aggregation during peptide chain assembly. Leonidas Zervas, Emeritus Professor of organic chemistry at the University of Athens, and past president of the National Academy of Greece, cele brated his seventieth birthday this past year.
