Formation of transmembrane pores is a very effective way of killing cells. It is thus not surprising that many bacterial and eukaryotic toxic agents are pore-forming proteins. Pore formation in a target membrane is a complex process composed of several steps; proteins need to attach to the lipid membrane, possibly aggregate in the plane of the membrane and finally form a pore by inserting part of the polypeptide chain across the lipid bilayer. Structural information about toxins at each stage is indispensible for the biochemical and molecular biological studies that aim to - derstand how pores are formed at the molecular level. There are currently only two Staphylococcus aureus and hemolysin E from Escherichia coli. Therefore, what we know about these proteins was obtained over many years of intense experimentation. We have nevertheless, in the last couple of years, witnessed a significant rise in structural information on the soluble forms of pore-forming proteins. Surprisingly, many unexpected similarities with other proteins were noted, despite extremely low or insignificant sequence similarity. It appears that lipid membrane binding and formation of transmembrane channels is achieved in many cases by a limited repertoire of structures. This book describes how several of the important pore forming toxin families achieve membrane bi- ing and which structural elements are used for formation of transmembrane pores. Our contributors have thus provided the means for a comparative analysis of several unrelated families.

When the six of us gathered to start planning for what was to be the Third Edition of Physiology of Membrane Disorders, it was clear that since 1986, when the Second Edition appeared, the field had experienced the dawning of a new era dominated by a change in focus from phenomenology to underlying mechanisms propelled by the power of molecular biology. In 1985, detailed molecular information was available for only three membrane transporters: the lac permease, bacterial rhodopsin, and the acetylcholine receptor. During the decade that has since elapsed, almost all of the major ion channels and transport proteins have been cloned, sequenced, mutagenized, and expressed in homologous as well as heterologous cells. Few, if any, of the transporters that were identified during the previous era have escaped the probings of the new molecular technologies and, in many instances, considerable insight has been gained into their mechanisms of function in health and disease. Indeed, in some instances novel, unexpected transporters have emerged that have yet to have their functions identified. In plant cells, the plasma membrane is a highly elaborated structure that functions as the point of exchange with adjoining cells, cell walls and the external environment. Transactions at the plasma membrane include uptake of water and essential mineral nutrients, gas exchange, movement of metabolites, transport and perception of signaling molecules, and initial responses to external biota. Selective transporters control the rates and direction of small molecule movement across the membrane barrier and manipulate the turgor that maintains plant form and drives plant cell expansion. The plasma membrane provides an environment in which molecular and macromolecular interactions are enhanced by the clustering of proteins in oligimeric complexes for more efficient retention of biosynthetic intermediates, and by the anchoring of protein complexes to promote regulatory interactions. The coupling of signal perception at the membrane surface with intracellular second messengers also involves transduction across the plasma membrane. Finally, the generation and ordering of the external cell walls involves processes mediated at the plant cell surface by the plasma membrane. This volume is divided into three sections. The first section describes the basic mechanisms that regulate all plasma membrane functions. The second describes plasma membrane transport activity. The final section of the book describes signaling interactions at the plasma membrane. These topics are given a unique treatment in this volume, as the discussions are restricted to the plasma membrane itself as much as possible. A more complete knowledge of the plasma membrane’s structure and function is essential to current efforts to increase the sustainability of agricultural production of food, fiber, and fuel crops.
