The number of mouse models that are available for the study of human genetic neurological disorders is large and growing rapidly. Therefore, it was difficult to select the models that were reviewed in this volume. Clearly, there are important models that are not discussed, and perhaps a volume twice this size would have been more appropriate. Moreover, the pace at which new models are being developed and analyzed is rapid. As this volume goes to press, I am sure that additional mouse genes responsible for naturally occurring neurological disorders are being discovered and that many new transgenic and mutant mouse strains are being developed. Therefore, this volume should not be viewed as a comprehensive compendium, but rather as an update of work in progress. It is exhilarating to witness the fast pace at which these models are being established as important tools in the study of basic neuroscience and neurological disorders. It will be even more exciting to see their utilization in the development and testing of therapeutic interventions for these diseases. I would like to thank each of the authors who have contributed to this volume for their time and their expertise. I would also like to thank Drs. Timothy Coetzee and Joshua Corbin for their advice in the selection of the topics covered. erebellar ataxia is a failure in muscular coordination that re C sults from a slow, progressive deterioration of neurons in the cerebellum. An estimated 150,000 people are affected by the he reditary ataxias and related disorders in the United States. At present, there is no known cure. In an experimental treatment aimed at reconstructing the damaged pathway through exog enous neuronal supplementation, genetically ataxic mice have been used for intracerebral grafting of genetically healthy cerebellar neuroblasts, and evidence has been obtained for graft-induced en hancement of behavioral responses after bilateral cerebellar grafts. Such results are encouraging and underscore the potential of the neural grafting technique in restoring cerebellar function. How ever, many of the pathological and biochemical mechanisms in the interaction between grafted tissue and the host brain need to be further elucidated in extensive experimental studies, and great cau tion must be used in contemplating the theoretical feasibility of a possible application in humans.

Neuroimmunology could be defined as the application of immunological methods to problems in neurobiology but such a definition is so all encompassing as to be unhelpful. It is not a precisely circumscribed discipline but it seems worthwhile at the outset to point to three of the major areas of activity. One rather early use of the term was in connection with studies on the immune response to antigens in the nervous system. This includes topics such as autoimmunity in the central and peripheral nervous sys tems, the response to neural tumors or viral infections, and the im munopathology of such processes. Although not at the forefront of the currently fashionable preoccupation with neuroimmunology, this area continues to be a vital and interesting one from both clinical and basic perspectives. A second very active area is the exploitation of antibodies to iden tified components of neural cells, and in particular to those molecules involved as neurotransmitters, in transmitter synthesis and breakdown, and as synaptic receptors. The immunohistochemical detection of these antigens has led to new insights into the functional organization of the nervous system, and reference to such studies is almost a sz'ne qua non for discussions of most central and peripheral synapses.

The purpose of the first four volumes of the Handbook of Genetics is to bring together collections of relatively short, authoritative essays or an notated compilations of data on topics of significance to geneticists. Many of the essays will deal with various aspects of the biology of certain species or species groups selected because they are favorite subjects for genetic investigation in nature or the laboratory. Often there will be an encyclo pedic amount of information available on such species, with new papers appearing daily. Most of these will be written for specialists in a jargon that is bewildering to a novice, and sometimes even to a veteran geneticist working with evolutionarily distant organisms. For such readers what is needed is a written introduction to the morphology, life cycle, reproductive behavior, and culture methods for the species in question. Lists giving the symbolism and descriptions for selected mutants that have been retained and are thus available for future studies are provided whenever possible.
