In spite of the continuing progress of research in the fields of cellular and molecular biology, which has oriented many embryologists towards molecular biology, no concrete explanation of morphogenesis has yet been found. The present state of knowledge of heart development is characterized by an enormous discrepancy between the qualitative descriptions of what happens on the organ level and the more or less quantitative information on subcellular and molecular events. It is generally not understood how cells form tissues and how tissues generate particular forms of an organ. In an attempt to fill the gaps we systematically studied in the period 1968 to 1973 one of the general but rather neglected morphogenetic mechanisms which integrates cells into tissues and organs-cell death. Only a small part of our research on cell death in the development of chick, rat and human embryo hearts has as yet been published in extenso. Most of it has been communicated in papers delivered at different scientific meetings. We would like to use the opportunity offered by Advances to present a syn thesis and integrative review of our results. In this way the actual period of discovery of the existence of cell death and of its morphogenetic role in the heart development come to an end. This opens up the next phase of our research which consists in studies of how cell death is integrated with other morphogenetic mechanisms.

It is clear that lysosomal enzymes often play a role in the destruction of the cytoplasm, but very few authorities feel that they initiate the process (Chapters 1, 2, 3, 5 -8, 12, 13). The cells show many forms of damage, and sometimes even complete destruction, before Iysosomes become a dominant part of the environ ment. What initiates the process is still unclear, although in several instances it appears that the death of a cell may arise from anyone of several pathways (Chapters, 10, II). It is rather interesting that evolution has chosen to achieve the same goal by different means. Apparently no one point is exceptionally or pre ferentially vulnerable, though a common pathway, such as permeability of the plasma membrane to calcium (Chapter 7), might currently be too subtle for routine identification. Factors which affect membrane stability and which induce mem brane bending can lead to blebing, cell fragmentation and death. Thus, more work on the changing chemistry of the plasma membrane in relation to environmental fluctuations would be welcomed. Space requirements and the major orientation of the book forced the exclusion of several very interesting topics: an evolutionary treatment of the advantages of cell death as a means of eliminating vestigial organs or embryonic scaffolding; or consider ation of the merits of body sculpting by cell death rather than cell growth.

The study of birth defects has assumed an importance even greater now than in the past because mortality rates attributed to congenital anomalies have declined far less than those for other causes of death, such as infectious and nutritional diseases. It is estimated that as many as 50% of all pregnancies terminate as miscarriages. In the majority of cases this is the result of faulty development. Major congenital malformations are found in at least 2% of all liveborn infants, and 22% of all stillbirths and infant deaths are associated with severe congenital anomalies. Teratological studies of an experimental nature are neither ethical nor justifiable in humans. Numerous investigations have been carried out in laboratory animals and other experimental models in order to improve our understanding of abnormal intrauterine development. In less than two decades the field of experimental teratology has advanced phenomenally. As a result of the wide range of information that is now accumulating, it has become possible to obtain an insight into the causes, mechanisms, and prevention of birth defects. However, considerable work will be needed before these problems can be resolved. The comributions in this volume include some of the more recent and exciting observations on the cellular and molecular aspects of developmental defects. It is not only a documentation of the latest experimental work, but it also indicates new and important areas for future research.
