This book discusses different signaling pathways involved in tumor development and progression. It discusses the pathways that allow tumor cells to proliferate, survive, and invade other tissues. Further, the book reviews the signal transduction regulating phagocytosis, production of cytokines, cell division, and differentiation. It elucidates the dysregulation of cellular signal transduction induced by the genetic and epigenetic changes that drive cancer and enumerates the signaling network encompassing the extracellular matrix, blood vessels, and the immune system. Additionally, the book provides mechanistic insights into inhibitors of the receptor tyrosine kinases, BRAF, EGFR, and ALK that improve clinical response and increase the survival of patients with cancer. The book covers the signal transduction pathways that regulate cell cycle progression and genomic stability, providing a mechanistic understanding of the major checkpoint pathways, and reviewing their diagnostic and therapeutic potential. This volume is essential reading for researchers to understand the therapeutic potential of important signaling molecules against cancer.

Molecular Mechanisms Of Cellular Stress Responses In Cancer And Their Therapeutic Implications written by Megan Chircop and has been published by Frontiers Media SA this book supported file pdf, txt, epub, kindle and other format this book has been release on 2015-03-06 with Carcinogenesis categories.
In response to stress, cells can activate a myriad of signalling pathways to bring about a specific cellular outcome, including cell cycle arrest, DNA repair, senescence and apoptosis. This response is pivotal for tumour suppression as all of these outcomes result in restriction of the growth and/or elimination of damaged and pre-malignant cells. Thus, a large number of anti-cancer agents target specific components of stress response signalling pathways with the aim of causing tumour regression by stimulating cell death. However, the efficacy of these agents is often impaired due to mutations in genes that are involved in these stress-responsive signalling pathways and instead the oncogenic potential of a cell is increased leading to the initiation and/or progression of tumourigenesis. Moreover, these genetic defects can increase or contribute to resistance to chemotherapeutic agents and/or radiotherapy. Modulating the outcome of cellular stress responses towards cell death in tumour cells without affecting surrounding normal cells is thus one of the ultimate aims in the development of new cancer therapeutics. To achieve this aim, a detailed understanding of cellular stress response pathways and their aberrations in cancer is required. This Research topic aims to reflect the broadness and complexity of this important area of cancer research.

This book comprehensively reviews the role of cancer stem cells (CSCs) in cancer initiation, progression, and resistance to anticancer therapies. The initial chapters examine the methods and procedure of the detection, isolation, and characterization of CSCs. It also introduces various epigenetic pathways that contribute to cancer initiation and tumorigenesis, particularly regarding the maintenance and survival of CSCs. It also explores the role of CSCs metabolism and the mechanisms of metabolic plasticity of CSCs in cancer biology. Further, it also presents the implications of CSCs on the origin of tumor heterogeneity and on heterogeneity of the therapeutic response. Towards the end, this book highlights the different immunotherapeutic approaches targeting CSCs with the potential of strongly improving cancer outcomes. This book offers a broad framework to scientists and clinicians into the state-of-the-art knowledge on cancer stem cell biology and highlights their therapeutic implications.

Cancer cells can change and adapt, especially within the host environment; a phenomenon known as cancer plasticity. Several factors, including the immune system can influence, and be influenced by, cancer plasticity which in turn can impact upon patient responses to treatment. As such, we currently face several challenges for implementing combination therapies as effective cancer treatment strategies. We have compiled a topic with a number of articles that emphasize the various aspects of cancer plasticity, describing in particular the important role of the tumor microenvironment. As we embark on a new era of precision medicine with multi-modal therapies for improving patient outcomes, this topic highlights some relevant points for consideration that are pertinent to the incorporation and effective use of new treatments as part of cancer treatment regimens, including immune-modulating drugs.
