Abstract: Background: Sweet sorghum is a multi-purpose crop that can be used for human food, animal feed, and bio-fuel production. This crop is drought-tolerant and can therefore be grown in arid and semi-arid regions especially in marginal areas in Egypt. Nevertheless, production of Sweet sorghum cannot coup with the increasing demand in consumption as a food, and fuel due to a lag in scientific research regarding its genetic improvement. Therefore, genetic improvement of Sweet sorghum is innately required so that new traits of economic importance could be introduced; however, this can only be achieved through the establishment of a robust in-vitro tissue culture system. Thus, this study aimed to establish an effective regeneration system for three different Sweet sorghum cultivars namely; Rex, Sugar Drip, and Ramada, using immature inflorescence explants. Subsequently, the cultivar Sugar was selected to examine its transformation efficiency using the microprojectile bombardment technology. Methods: For callus induction media, explants from immature inflorescences of different lengths (1.5- 16 cm), were grown over a combination of 2,4-D (0, 2, 4 & 6mg/L) with kinetin (0, 0.2 & 0.5mg/L). For regeneration media, BAP was the main cytokinin implemented in the three different treatments along with either: NAA, IAA, or IAA & TDZ, whereas TDZ was employed alone in a fourth treatment. The transformation system of the cultivar Sugar Drip was also developed using immature inflorescence explants. Co-bombardment was performed using the neomycin phosphotransferase II (emnptII) gene under the control of maize ubiquitin (emUbi1) promoter. The selection of putative transgenic plants was performed using paromomycin antibiotic. Results: The best embryogenic callus induction frequency was observed in Rex (77%) on modified MS media supplemented with 4mg/L 2,4-D + 0.2mg/L kinetin after six weeks from culture. However, Ramada and Sugar Drip had their highest callus induction rates of 93 and 94%, respectively, when 6mg/L 2,4-D + 0.2mg/L kinetin was used. The addition of 1mg/L IAA + 0.5mg/L BAP + 0.1mg/L TDZ had its best outcome in terms on shoot induction, shoot/callus number, and root formation. In Sugar Drip transformation, paromomycin eliminated most the non-transgenic plants from the putative transgenic ones. Eventually, 6 out of 348 bombarded samples were found to be transgenic after PCR screening. The percentage of transformation from two independent experiments was around 1.724%. Conclusion: PCR analysis of putative transformants revealed a transformation efficiency of 1.724%. Therefore, genetic transformation using particle bombardment has shown to be a successful method for the transformation of immature Sweet sorghum inflorescences with relatively high success rates.

Recently designated as a bioenergy crop, Sorghum is rather unique as it can produce large quantities of cellulose or sugar which can be used to produce advanced biofuels or compounds. Sweet sorghum contains high levels of sugars and biomass sorghums consist primarily of ligno-cellulosic biomass. Improvement of both sorghum types is essential for maximizing production and conversion efficiency. Photoperiod sensitive sorghum is thought to maximize biomass production yet maturity influence on biomass production and composition is not fully understood. Utilizing sorghum for sugar production has increased efforts to develop sweet sorghums with sugar yields similar to sugarcane. Hybridization of these species has been investigated with, until recently, little success. Testing newly developed intergeneric hybrids and improvement of parents used in their creation will determine their feasibility and improve hybrid performance. Objectives of this research are multifaceted. First, analyze photoperiod sensitive sorghum in varying day length environments to determine maturity effects on plant phenotype, composition, and QTL detection. Second, analyze intergeneric sorghum x sugarcane hybrids to determine agronomic performance in relation to sugarcane. Lastly, introgress the iap allele into sweet sorghum females for use in intergeneric hybrid creation. Photoperiod sensitive sorghum RILs were evaluated in College Station and Weslaco, Texas and Puerto Rico which caused differential expression of plant maturity. Genetic control of trait expression was high for each location. Results indicate gradual induction of plant maturity increases detection of phenotypic QTL and detection of compositional QTL increases when maturity effects on plant phenotype are reduced. Intergeneric sorghum x sugarcane F1 hybrids were compared to sugarcane in Weslaco, Texas in 2011. Each hybrid expressed agronomic traits similar or better than that of the sugarcane variety. High levels of repeatability and genetic influence on trait expression were observed. Overall performance of the sugarcane variety was better than any individual hybrid tested. Introgression of iap into sweet sorghum was successful and generated seventeen new sweet sorghum female genotypes possessing the allele. Only two genotypes exhibited higher brix readings and both were later maturing than Tx3361. Height and maturity of all developed genotypes varied and desirability of developed lines was similar to Tx3361. Sweet sorghum (Sorghum bicolor) has potential as a bioenergy feedstock due to its high yield potential and the production of simple sugars for fermentation. Sweet sorghum cultivars are typically tall, high biomass types with juicy stalks and high sugar concentration. These sorghums can be harvested, milled, and fermented to ethanol using technology similar to that used to process sugarcane. Sweet sorghum has advantages in that it can be planted by seed with traditional planters, is an annual plant that quickly produces a crop and fits well in crop rotations, and it is a very water-use efficient crop. Processing sweet sorghum is capital intensive, but it could fit into areas where sugarcane is already produced. Sweet sorghum could be timed to harvest and supply the sugar mill during the off season when sugarcane is not being processed, be fit into crop rotations, or used in water limiting environments. In these ways, sweet sorghum could be used to produce ethanol in the Southern U.S and other tropical and subtropical environments. Traditionally, sweet sorghum has been grown as a pureline cultivar. However, these cultivars produce low quantities of seed and are often too tall for efficient mechanical harvest. Sweet sorghum hybrids that use grain-type seed parents with high sugar concentrations are one way to overcome limitation to seed supply and to capture the benefits of heterosis. There are four objectives of this research. First to evaluate the importance of genotype, environment, and genotype-by-environment interaction effects on the sweet sorghum yield and composition. The second objective is to determine the presence and magnitude of heterosis effects for traits related to sugar production in sweet sorghum. Next: to study the ability of sweet sorghum hybrids and cultivars to produce a ratoon crop and determine the contribution of ratoon crops to total sugar yield. The final objective is to evaluate variation in composition of sweet sorghum juice and biomass. Sweet sorghum hybrids, grain-type sweet seed parents, and traditional cultivars that served as male parents were evaluated in multi-environment trials in Weslaco, College Station, and Halfway, Texas in 2007 and 2008. Both genotype and environment influenced performance, but environment had a greater effect than genotype on the composition of sweet sorghum juice and biomass yield. In comparing performance, elite hybrids produced fresh biomass and sugar yields similar to the traditional cultivars while overcoming the seed production limitations. High parent heterosis was expressed among the experimental hybrids for biomass yield, sugar yield and sugar concentration. Additional selection for combining ability would further enhance yields and heterosis in the same hybrid. Little variation was observed among hybrids for juice and biomass composition suggesting that breeding efforts should focus on yield before altering plant composition.
