By means of quantitative analysis of the tissue components in the cortex of the mouse, this book presents an overall picture of the cortical network which is then related to various theories on cortical function. Centering around the idea of a diffuse network in a fairly homogeneous population of excitatory neurons, that of the pyramidal cells, it shows that the whole organisation in the cortical skeleton of pryramidal cells corresponds well with the idea of an associative memory and with the theory of cell assemblies. In this essay we propose a view of the cerebral cortex from an unusual angle, determined by our search for the most general statements on the relation between cortical structure and function. Our results may be at variance with the ideas of those physiologists who have specialized subsystems of the cortex in mind, but are not in contrast with them, as we hope to show. Our view is akin to one presented by Moshe Abeles in his monograph Local Cortical Circuits ( 1982). It is related to the theory developed by Gunther Palm, for many years our partner, in Neural Assemblies (1982). It owes much to the ideas which inspire the work of George Gerstein (beginning with the classic: Gerstein 1962) and especially to the insights gained by our colleague Ad Aertsen and his group in TUbingen. Our ideas on the cortex find some resonance in a recent trend in Artificial Intelligence (Kohonen 1977; Hopfield 1982; Rumelhart et al. 1986), and indeed the diagram (Braitenberg 1974a) representing the dominant idea of that trend, the associative matrix with feedback, has served as an emblem for the editorial enterprise Studies of Brain Function since its inception in 1977. Since our earlier reports (Braitenberg 1974a,b, 1977, 1978a,b, 1986; Schilz 1976, 1978, 198la,b, 1986) enough new experimental evidence has accumulated in our own laboratory (summarized by Schilz 1989) and elsewhere to justify a more extended and more confident presen tation.

The true revolution in the age of digital neuroanatomy is the ability to extensively quantify anatomical structures and thus investigate structure-function relationships in great detail. Large-scale projects were recently launched with the aim of providing infrastructure for brain simulations. These projects will increase the need for a precise understanding of brain structure, e.g., through statistical analysis and models. From articles in this Research Topic, we identify three main themes that clearly illustrate how new quantitative approaches are helping advance our understanding of neural structure and function. First, new approaches to reconstruct neurons and circuits from empirical data are aiding neuroanatomical mapping. Second, methods are introduced to improve understanding of the underlying principles of organization. Third, by combining existing knowledge from lower levels of organization, models can be used to make testable predictions about a higher-level organization where knowledge is absent or poor. This latter approach is useful for examining statistical properties of specific network connectivity when current experimental methods have not yet been able to fully reconstruct whole circuits of more than a few hundred neurons.

Supercomputing facilities are becoming increasingly available for simulating activity dynamics in large-scale neuronal networks. On today's most advanced supercomputers, networks with up to a billion of neurons can be readily simulated. However, building biologically realistic, full-scale brain models requires more than just a huge number of neurons. In addition to network size, the detailed local and global anatomy of neuronal connections is of crucial importance. Moreover, anatomical connectivity is not fixed, but can rewire throughout life (structural plasticity)—an aspect that is missing in most current network models, in which plasticity is confined to changes in synaptic strength (synaptic plasticity). The papers in this Ebook, which may broadly be divided into three themes, aim to bring together high-performance computing with recent experimental and computational research in neuroanatomy. In the first theme (fiber connectivity), new methods are described for measuring and data-basing microscopic and macroscopic connectivity. Neuronal dendritic trees are complex structures that endow the cell with powerful computing capabilities and allow for high neural interconnectivity. Studying the function of dendritic structures has a long tradition in theoretical neuroscience, starting with the pioneering work by Wilfrid Rall in the 1950s. Recent advances in experimental techniques allow us to study dendrites with a new perspective and in greater detail. The goal of this volume is to provide a résumé of the state-of-the-art in experimental, computational, and mathematical investigations into the functions of dendrites in a variety of neural systems. The book first looks at morphological properties of dendrites and summarizes the approaches to measure dendrite morphology quantitatively and to actually generate synthetic dendrite morphologies in computer models. This morphological characterization ranges from the study of fractal principles to describe dendrite topologies, to the consequences of optimization principles for dendrite shape. Individual approaches are collected to study the aspects of dendrite shape that relate directly to underlying circuit constraints and computation.
