Corynebacterium glutamicum was discovered in Japan in 1956 as a natural glutamate producer. Its “microbial factory” qualities, such as its physiological plasticity and robust catalytic functionalities, have since facilitated the development of efficient production processes for amino acids, nucleotides and vitamins. This monograph illustrates how the information gleaned from complete genome sequencing allows the rational engineering of the entire cellular metabolism and how systems biology permits the further optimization of C. glutamicum as a biocatalyst. Aspects of gene regulation, metabolic pathways, sugar uptake, protein secretion, cell division and biorefinery applications highlight the enormous biotechnological and biorefinery potential.

This updated second edition covers the molecular biology, genome engineering tools and comprehensive analysis techniques for Corynebacterium glutamicum. Aside from modern omics-based approaches, the authors also focus on cell physiology, including cell division, central carbon metabolic pathways, and the respiratory chain. Readers will learn how primary mechanisms like energy metabolism can be applied in processes like biorefinery. Newly added topics include cell envelope structures and aromatic compound metabolism in C. glutamicum. These chapters will be particularly useful for those interested in the microbial production of commodity chemicals, fuels, and proteins. Corynebacteriacea are already some of the most important industrial microorganisms. Understanding the cell physiology of C. glutamicum will help manufacturers to increase their product range and productivity through efficient metabolic engineering.

Corynebacterium glutamicum is most widely known for its role in the industrial production of L-glutamate and L-lysine and as a platform organism for the production of a variety of fine chemicals, biofuels, and polymers. The organism's accessibility to genetic manipulation has resulted in a wealth of data on its metabolism and regulatory networks; this in turn makes C. glutamicum the model organism of choice in white biotechnology. A key development in recent years has been the engineering of C. glutamicum to utilize a broader spectrum of carbon sources (e.g. glycerol, galactose, and pentose sugars), in addition to the traditional hexoses. Given its unique ability to co-utilize mixed carbon sources, C. glutamicum could be used for the clean-up of wastes from agricultural or other industries, simultaneously producing useful compounds such as L-lysine or putrescine. This book provides a comprehensive overview of current knowledge and research on C. glutamicum systems biology and biotechnological applications. Written by a team of prominent scientists, the topics covered include: proteomics * flux analysis technology for metabolic analysis * metabolic engineering for alternative carbon source utilization * manipulation of nitrogen metabolism * transport, degradation, and assimilation of aromatic compounds and their regulation * engineering for production of organic acids and alcohols * microbial factory for the production of polyesters * biotechnological application under oxygen deprivation * the secretory production of heterologous proteins * genetically-encoded biosensors. Packed with practical information and state-of-the-art science, this concise volume is an essential handbook for everyone working with Corynebacterium and related organisms in academia, biotechnology companies, and the pharmaceutical industry, It is a recommended volume for all microbiology libraries. More than fifty years of scientific and industrial developments focused on the amino acid producer bacterium Corynebacterium glutamicum has successfully yielded up to 2.93 and 1.95 million tons/year of L glutamic acid and L-lysine, respectively. As a result of this biotechnological effort, a massive amount of knowledge decidedly applicable to the development of new products has been gained. Nowadays, this knowledge is used to enlarge the portfolio of C. glutamicum industrial products, since they are highly relevant as chemicals, biodegradable polymers, ingredients, or additives in food, feed, cosmetics and pharmaceuticals. This book addresses production of these new metabolites (C. glutamicum applications) and their potential under the expert point of view of those groups directly involved in their developments. Thus, amino acids out of the traditional glutamic acid or lysine, as L valine; food colorants such as carotenoids; dicarboxylic acids used as building blocks (e.g: succinate); alpha hydroxy acids as L lactate, D lactate or glycolate; biodegradable polyester (eg.: polyhydroxybutyrate); alcohols for biofuel as isobutanol as well as bioremediation applications of the new developments around C. glutamicum described in this book. Those industrial relevant compounds come to the market parallel with the developing application of new technologies. Hence, cell activity as a whole by means of the Proteomics or Transcriptomics approaches, as well as the methodologies that mimic the large fermetor conditions in a scale-down format are also reviewed in the book. Additionally, a historical description and an updated review of the taxonomical environment of Corynebacterium genus open an essential manual for enterprises, researchers and academia involved in Actinobacteria, secondary metabolite production and industrial applications. This book reviews the evolution of almost sixty years of development, initially focused on amino acid production, which unleashes the idea of C. glutamicum as a biorefinery due to its ability to generate hundreds of products demanded by the market.

L-lysine is an essential amino acid which is mainly applied as supplement in animal feed. With a world market of around 1,000,000 t/a it is one of the most important fermentation products. The gram positive soil bacterium Corynebacterium glutamicum thereby serves as major production organism. The aim of the present work was rational strain engineering of lysine production by C. glutamicum using a systems-oriented approach. Detailed studies on this organism exposed lysine biosynthesis, NADPH metabolism, TCA cycle and precursor supply as key pathways. The central role for strain characterization can here be assigned to 13C metabolic flux analysis, which was also applied in this work to obtain a detailed insight into the cellular physiology. Based on an existing model, the network for flux determination was extended in the present study to increase the number of estimable flux parameters. With regard to strain optimization, targeted modulation of enzyme activities within the cell is crucial to increase or reduce carbon conversion of the selected pathways. The common toolbox such as over expression by promoter exchange or gene deletion was complemented with the method of start codon exchange allowing permanent modulation of enzyme activity. As host-strains for target evaluation, genetically defined, wild type based lysine producers were used which all express a feedback-deregulated variant of aspartate kinase. The most beneficial modifications identified in this work comprised over expression of diaminopimelate dehydrogenase, down-regulation of TCA cycle by attenuation of isocitrate dehydrogenase and engineering of the NADPH metabolism. By subsequent implementation of beneficial targets in a single strain, a rationally designed lysine hyper-producer was created. This exhibited remarkable production properties such as a lysine-HCl titre of 120 g L-1 and a carbon conversion yield of up to 55 %.
