Cryptic Enzymes and Moonlighting, a new volume in the Foundations and Frontiers in Enzymology series, offers a thorough overview of cryptic enzymes and moonlighting proteins in signalling cascades. It explores recent advances in research, and brings together an array of information across different fields to enable better targeting of these exciting enzymes and their interactomes. With a clear focus on the role of moonlighting and cryptic enzymes in signal transduction, the book considers examples of cryptic enzymes across species, as well in human healthy biology and pathogenesis. In early chapters, leading international contributors discuss evolutionary considerations for moonlighting proteins, moonlighting interactions in the extracellular matrix, eukaryotic moonlighting proteins, modulating, moonlighting kinases, moonlighting proteins in neurobiology signalling, metabolic enzyme moonlighting, and RNA binding and regulatory proteins. Later, methods-driven chapters discuss practical aspects of identifying hidden moonlighting domains in proteins, computational approaches and bioinformatic tools for the identification of cryptic enzymes, establishing cryptic enzyme interactomes, and assessing contributions of moonlighting proteins to signal cascades.

The past 25 years has seen the emergence of a wealth of data suggesting that novel biological functions of known proteins play important roles in biology and medicine. This ability of proteins to exhibit more than one unique biological activity is known as protein moonlighting. Moonlighting proteins can exhibit novel biological functions, thus extending the function of the proteome, and are also implicated in the pathology of a growing number of idiopathic and infectious diseases. This book, written by a cell biologist, protein evolutionary biologist and protein bioinformatician, brings together the latest information on the structure, evolution and biological function of the growing numbers of moonlighting proteins that have been identified, and their roles in human health and disease. This information is revealing the enormous importance protein moonlighting plays in the maintenance of human health and in the induction of disease pathology. Protein Moonlighting in Biology and Medicine will be of interest to a general readership in the biological and biomedical research community.

Moonlighting Proteins: Novel Virulence Factors in Bacterial Infections is a complete examination of the ways in which proteins with more than one unique biological action are able to serve as virulence factors in different bacteria. The book explores the pathogenicity of bacterial moonlighting proteins, demonstrating the plasticity of protein evolution as it relates to protein function and to bacterial communication. Highlighting the latest discoveries in the field, it details the approximately 70 known bacterial proteins with a moonlighting function related to a virulence phenomenon. Chapters describe the ways in which each moonlighting protein can function as such for a variety of bacterial pathogens and how individual bacteria can use more than one moonlighting protein as a virulence factor. The cutting-edge research contained here offers important insights into many topics, from bacterial colonization, virulence, and antibiotic resistance, to protein structure and the therapeutic potential of moonlighting proteins. Moonlighting Proteins: Novel Virulence Factors in Bacterial Infections will be of interest to researchers and graduate students in microbiology (specifically bacteriology), immunology, cell and molecular biology, biochemistry, pathology, and protein science.

Does the field of evolution differ from other sciences? The author, a reviewer for a major medical journal, scrutinized hundreds of scientific references in evolutionary literature, adopting the same standards used for studies submitted for medical publication. The data show that there are two types of evolution, microevolution and macroevolution, with a clear boundary between them based upon the presence and absence of empirical evidence, respectively. The surprising results show that there is a universal disconnect between the data and the conclusions that claim to show the larger changes of macroevolution. The author reveals patterns of deviations from standard scientific methods in these studies. For the first time, evolutionary data have been summarized to describe both what evolution can and cannot accomplish. The author shows the reader how to recognize the different ways in which the evidence for microevolution within and between some species differs from the unsupported macroevolution of most species. Previous critiques of macroevolution have been debunked by advocates who have cited a multitude of scientific studies. This book goes beyond previous critiques by directly addressing the data from these studies to see if they do, in fact, support macroevolution-focused conclusions. Many expert counterarguments against this book’s thesis are presented and examined in the context of scientific research to reassure the reader that the author has left no stone unturned in the macroevolution debate. A theory is proposed as to why there may be no empirical evidence for macroevolution. The book concludes with a section entitled “What we see differently.” There, the author shows the reader the differences in perspective between the evolutionist and macroevolution critic as they look at and interpret the very same set of data.
