This book, like other monographs of the Cellular Organelles series, is not a comprehensive review, but an introduction to the study of cytoskeleton. Accordingly, we describe only the main facts and concepts related to cyto skeleton. Needless to say, selection and interpretation was influenced by the personal interests and opinions of the authors, although we attempted to be as fair as possible. We wished to familiarize the reader not only with well established facts, but with current unsolved problems. Therefore, the words "possibly," "maybe," "not known," and "not clear" are much more frequent in this text than in many others. In accordance with the style of the series, relatively short lists of addi tional readings are given at the end of each chapter; these lists contain mostly the recent reviews and a few original papers describing certain phenomena in detail. Few references are cited in the text; these citations are given to help the reader find the source of certain new data and theories, which are not discussed at length in the reviews. In contrast, many well-established facts and widely known theories are not cited.

The student of biological science in his final years as an undergraduate and his first years as a graduate is expected to gain some familiarity with current research at the frontiers of his discipline. Over the past two decades experimental studies have solidified the int- pretation of the cytoskeleton as a highly dynamic network of microtubules, actin microfilaments, intermediate filaments, and myosin filaments. Rather than a network of disparate fibers, these polymers are often interconnected and display synergy, which is the combined action of two or more cytoskeletal polymers to achieve a specific cellular structure or function. Cross-commu- cation among cytoskeletal polymers is thought to be achieved through cytoskeletal polymer accessory proteins and molecular motors that bind two or more cytoskeletal polymers. Development of the modern concept of the cytoskeleton is a direct o- growth of advances in experimental tools and reagents that are available to cell and molecular biologists. Technological advances and refinements in cell imaging have made it possible to selectively image a single cytoskeletal po- mer and monitor its dynamics through the use of fluorescence probes in vitro and in vivo. Two decades ago, cytoskeletal research was limited to a few perturbation reagents that included colchicine and cytochalasin. Today, the perturbation arsenal has expanded to a highly selective group of reagents that includes Taxol, nocodazole, benomyl, latrunculin, jasplakinolide, and such endogenous proteins as gelsolin. These reagents enable the investigator to selectively perturb or destroy a cytoskeletal polymer while leaving other cytoskeletal polymers intact. Site-specific monoclonal antibodies that target a specific cytoskeletal polymer have proven to be highly selective affinity tools for cytoskeletal research.

The term cytoskeleton has become firmly established in today's scientific vocabulary. Indeed, it is difficult to believe that only ten years ago, it was virtually non-existent. Since then, the modern field of research on the structural organization of the cytoplasm has turned into one of the most productive and rapidly expanding research areas in Cell Biology today. Considerable progress has been made towards the identification of the various structural components of the cytoskeleton and their interactions with one another and with membranes. The first attempts to understand, in molecular terms, complex cellular processes such as shape changes, locomotion, division, and organelle movements have been made. And it is now apparent that the cytoskeleton has impact on other biological processes such as the control of gene expression, protein synthesis, cell cycle regulation, and development. This monograph outlines the basic properties of the major components of the polymeric filament networks and their interactions and associations. Wherever possible, emphasis is placed on more recent references. Any attempt to cover a research field this complex in an introductory mono graph is, by necessity, fragmentary, and oversights or omissions are inevitable. The mammalian cytoskeleton is an internal framework of actin, tubulin, and intermediate filament proteins. Proteins of these three classes assemble non-covalently into filamentous polymers that support the structural integrity of the relatively fragile lipid plasma membrane. In addition, cytoskeletal polymers provide the mechanical strength that hold a cell together and anchor it to its growth substrate. The cytoskeleton must also have the capacity for rapid and substantial remodeling and provide the motive and tractor force necessary to drive motility. As such, the cytoskeleton has a functional duality: sufficiently rigid to prevent plasma membrane deformation but pliable enough to allow for cytokinesis; sufficiently adhesive to allow for traction but dynamic enough to allow movement from one place to another. A major research challenge in cytoskeleton biology is to understand how cytoskeletal proteins assemble and dissemble in support of physiological processes. This chapter will focus on the role of the actin cytoskeleton in cell migration. More specifically, we will focus on the actin cytoskeleton of vertebrate cells.
