The last decade has witnessed remarkable developments in antibody research and its therapeutic applications. With the methods of molecular biology it is now possible to manipulate the specificities and activities of antibody molecules to generate an almost limitless array of structures for both basic investigations and the clinical setting. The contributions to this volume cover all three domains of the antibody: the variable regions, the relatively neglected but crucial hinge, and the constant region. These studies provide critical structural and functional information about antibodies, while also pointing the way to the construction of molecules with enhanced or even novel properties. Bringing together major experts on antibody engineering, this book is highly recommended to faculty, postdoctoral fellows and graduate students in molecular biology, microbiology, immunology, cancer research and genetics.

The field of antibody engineering has become a vital and integral part of making new, improved next generation therapeutic monoclonal antibodies, of which there are currently more than 300 in clinical trials across several therapeutic areas. Therapeutic antibody engineering examines all aspects of engineering monoclonal antibodies and analyses the effect that various genetic engineering approaches will have on future candidates. Chapters in the first part of the book provide an introduction to monoclonal antibodies, their discovery and development and the fundamental technologies used in their production. Following chapters cover a number of specific issues relating to different aspects of antibody engineering, including variable chain engineering, targets and mechanisms of action, classes of antibody and the use of antibody fragments, among many other topics. The last part of the book examines development issues, the interaction of human IgGs with non-human systems, and cell line development, before a conclusion looking at future issues affecting the field of therapeutic antibody engineering. - Goes beyond the standard engineering issues covered by most books and delves into structure-function relationships - Integration of knowledge across all areas of antibody engineering, development, and marketing - Discusses how current and future genetic engineering of cell lines will pave the way for much higher productivity

Biophysical Characterization of Proteins in Developing Biopharmaceuticals is concerned with the analysis and characterization of the higher-order structure (HOS) or conformation of protein based drugs. Starting from the very basics of protein structure this book takes the reader on a journey on how to best achieve this goal using the key relevant and practical methods commonly employed in the biopharmaceutical industry today as well as up and coming promising methods that are now gaining increasing attention. As a general resource guide this book has been written with the intent to help today's industrial scientists working in the biopharmaceutical industry or the scientists of tomorrow who are planning a career in this industry on how to successfully implement these biophysical methodologies. In so doing a keen focus is placed on understanding the capability of these methodologies in terms of what information they can deliver. Aspects of how to best acquire this biophysical information on these very complex drug molecules, while avoiding potential pitfalls, in order to make concise, well informed productive decisions about their development are key points that are also covered. - Presents the reader with a clear understanding of the real world issues and challenges in using these methods. - Highlights the capabilities and limitations of each method. - Discusses how to best analyze the data generated from these methods. - Points out what one needs to look for to avoid making faulty conclusions and mistakes. - In total it provides a check list or road map that empowers the industrial scientists as to what they need to be concerned with in order to effectively do their part in successfully developing these new drugs in an efficient and cost effective manner.

Despite the phenomenal clinical success of antibody-based biopharmaceuticals in recent years, discovery and development of these novel biomedicines remains a costly, time-consuming, and risky endeavor with low probability of success. To bring better biomedicines to patients faster, we have come up with a strategic vision of Biopharmaceutical Informatics which calls for syncretic use of computation and experiment at all stages of biologic drug discovery and pre-clinical development cycles to improve probability of successful clinical outcomes. Biopharmaceutical Informatics also encourages industry and academic scientists supporting various aspects of biotherapeutic drug discovery and development cycles to learn from our collective experiences of successes and, more importantly, failures. The insights gained from such learnings shall help us improve the rate of successful translation of drug discoveries into drug products available to clinicians and patients, reduce costs, and increase the speed of biologic drug discovery and development. Hopefully, the efficiencies gained from implementing such insights shall make novel biomedicines more affordable for patients. This unique volume describes ways to invent and commercialize biomedicines more efficiently: Calls for digital transformation of biopharmaceutical industry by appropriately collecting, curating, and making available discovery and pre-clinical development project data using FAIR principles Describes applications of artificial intelligence and machine learning (AIML) in discovery of antibodies in silico (DAbI) starting with antigen design, constructing inherently developable antibody libraries, finding hits, identifying lead candidates, and optimizing them Details applications of AIML, physics-based computational design methods, and other bioinformatics tools in fields such as developability assessments, formulation and excipient design, analytical and bioprocess development, and pharmacology Presents pharmacokinetics/pharmacodynamics (PK/PD) and Quantitative Systems Pharmacology (QSP) models for biopharmaceuticals Describes uses of AIML in bispecific and multi-specific formats Dr Sandeep Kumar has also edited a collection of articles dedicated to this topic which can be found in the Taylor and Francis journal mAbs.
