The plan for this atlas evolved from the necessity of providing the biology student interested in protozoology, cytology, and para sitology with an introduction to the study of fine structure in Protozoa. To reduce the book's extend, a selection of characteristic protozoans had to be made, limited to those which could be regarded as representative for entire groups. Interest in parasitic protozoans has been steadily on the increase over the last 10 years. This particular group of organisms thus seemed a very suitable choice. The" Apicomplexa" were selected as an area of emphasis. These once were part of the collective group called Sporozoa which included many parasitic protozoans of uncertain taxonomy. Fine structural research has been of especial significance for the Apicomplexa, since Protozoa belonging to this subgroup can now be named, characterized, and classified by features recognizable by electron microscopy. Only the fine structure of whole cells is represented in this atlas, so that the ciliates have been of necessity excluded. Their cells are too large in diameter in any case for our purpose here. They also play only a minor role as parasitic organisms. This book utilizes a new method to facilitate the analysis of protozoan fine structure. An electron micrograph, a descriptive text, and an analytic drawing are arranged on two facing pages so that the electron micrograph and the drawing can be compared.

In the continuing quest to explore structure and to relate struc tural organization to functional significance, the scientist has developed a vast array of microscopes. The scanning electron microscope (SEM) represents a recent and important advance in the development of useful tools for investigating the structural organization of matter. Recent progress in both technology and methodology has resulted in numerous biological publications in which the SEM has been utilized exclusively or in connection with other types of microscopes to reveal surface as well as intracellular details in plant and animal tissues and organs. Because of the resolution and depth of focus presented in the SEM photograph when compared, for example, with that in the light microscope photographs, images recorded with the SEM have widely circulated in newspapers, periodicals and scientific journals in recent times. Considering the utility and present status of scanning electron microscopy, it seemed to us to be a particularly appropriate time to assemble a text-atlas dealing with biological applications of scanning electron microscopy so that such information might be presented to the student and to others not yet familiar with its capabilities in teaching and research. Plants, fungi, and viruses were among the first biological objects studied with an electron microscope. One of the two first instruments built by Siemens was used by Helmut Ruska, a brother of Ernst Ruska, the pioneer in constructing electron microscopes. H. Ruska published numerous papers on different biological objects in 1939. In one of these, the pictures by G. A. Kausche, E. Pfankuch, and H. Ruska of tobacco mosaic virus opened a new age in microscopy. The main problem was then as it still is today, to obtain an appropriate preparation of the specimen for observation in the electron microscope. Beam damage and specimen thickness were the first obstacles to be met. L. Marton in Brussels not only built his own instrument, but also made considerable progress in specimen preparation by introducing the impregnation of samples with heavy metals to obtain useful contrast. His pictures of the bird nest orchid root impregnated with osmium were revolutionary when published in 1934. It is not the place here to recall the different techniques which were developed in the subsequent years to attain the modern knowledge on the fine structure of plant cells and of different plant pathogens. The tremendous progress obtained with tobacco mosaic virus is reflected in the chapter by M. Wurtz on the fine structure of viruses in this Volume. New cytochemical and immunological techniques considerably surpass the morphological information obtained from the pathogens, especially at the host-parasite interface.
