In this book, authors who are experts in their fields describe current advances on commercial crops and key enabling technologies that will underpin future advances in biotechnology. Tremendous progress has been made in introducing novel genes and traits into plant genomes since the first creation of transgenic plants thirty years ago, and the first commercialization of genetically modified maize in 1996. Consequently, cultivation of biotech crops with useful traits has increased more than 100-fold from 1.7 million hectares in 1996 to over 175 million hectares globally in 2013. This achievement has been made possible by continued advances in understanding the basic molecular biology of regulatory sequences to modulate gene expression, enhancement of protein synthesis and new technologies for transformation of crop plants. This book has three sections that encompass knowledge on genetically modified (GM) food crops that are currently used by consumers, those that are anticipated to reach the market place in the near future and enabling technologies that will facilitate the development of next generation GM crops. First attempts to isolate plant genes were for those genes that are abun dantly expressed in a particular plant organ at a specific stage of devel opment. However, many important gene products are produced in a very minute quantity and in specialized cell types. Such genes can now be isolated using a variety of approaches, some of which are described in this volume. The rapid progress during the last decade in regeneration of a number of crop plants and the availability of molecular tools to introduce foreign genes in plants is allowing the engineering of specific traits of agri cultural importance. These genes must, however, be regulated in a spatial and temporal manner in order to have desired effects on plant devel opment and productivity. The habitat of plants necessitate adaptive responses with respect to the environmental changes. Starting from germination of the seed, the plant begins to sense environmental cues such as moisture, light, temperature and the presence of pathogens, and begins to respond to them. Little is known about various signal transduction pathways that lead to biochemical and morphogenetic responses, in particular, transition from vegetative to reproductive phase. With the availability of tools to generate specific mutations via transposon tagging, identification and isolation of genes affecting these processes may be facilitated. Transfer of these genes into heterologous environments will allow understanding of the complex processes that control plant development.

The intricacies of plant growth and development present a fascinating intellectual challenge, and yet our understanding of the subject has increased relatively slowly, despite the application of many different experimental approaches. Now, however, the introduction of molecular methods, coupled with genetic transformation technology, has provided a change in pace, and fundamental advances are occurring rapidly. This volume, the second in our Plant Biotechnology series, shows how we are beginning to understand the molecular basis of plant growth and development, and are thus moving from the descriptive to the predictive stage. The ability, discussed in chapter one, to generate a fivefold change in plant height by overexpression of a single gene for the photoreceptor phytochrome heralds not only a new phase in plant photobiology but also highlights the close relationship between fundamental knowledge and commercial application. Other chapters review progress in our understanding of the molecular basis of hormone action and processes such as tuber development, seed protein synthesis and deposition, fruit ripening, and self-recognition during pollination. The successful uses of antisense genes to alter the colour and pattern of flowers and to change the enzymic composition of ripening fruit are also discussed, together with identification and down regulation of a gene involved in ethylene synthesis by antisense technology. Opportunities are considered for altering the composition and quality of harvested plant organs and for using plants to synthesise novel products.

A second conference. entitled "Tailoring Genes for Crop Improvement." spon sored by the UC-Davis College of Agricultural and Environmental Sciences and the College's Biotechnology Program. was held at Davis in August. 1986. to discuss the notable advances that had been made during the intervening years in the technology for gene modification. transfer. and expression in plants. This volume contains papers that were presented at this meeting and provides readers with examples of how the new experimental strategies are being used to gain a clearer understanding of the biology of the plants we grow for food and fiber; it also discusses how molecular biology approaches are being used to introduce new genes into plants for plant breeding programs. A recent volume of this series (Signals and Signal Transduction Pathways in Plants (K. Palme, ed.) Plant Molecular Biology 26, 1237-1679) described the relay races by which signals are transported in plants from the sites of stimuli to the gene expression machinery of the cell. Part of this machinery, the transcription apparatus, has been well studied in the last two decades, and many important mechanisms controlling gene expression at the transcriptional level have been elucidated. However, control of gene expression is by no means complete once the RNA has been produced. Important regulatory devices determine the maturation and usage of mRNA and the fate of its translation product. Post-transcriptional regulation is especially important for generating a fast response to environmental and intracellular signals. This book summarizes recent progress in the area of post-transcriptional regulation of gene expression in plants. 18 chapters of the book address problems of RNA processing and stability, regulation of translation, protein folding and degradation, as well as intracellular and cell-to-cell transport of proteins and nucleic acids. Several chapters are devoted to the processes taking place in plant organelles.
