ATP, the intracellular energy source, is also an extremely important cell–cell signalling molecule for a wide variety of cells across evolutionarily diverse organisms. The extracellular biochemistry of ATP and its derivatives is complex, and the multiple membrane receptors that it activates are linked to many intracellular signalling systems. Purinergic signalling affects a diverse range of cellular phenomena, including ion channel function, cytoskeletal dynamics, gene expression, secretion, cell proliferation, differentiation and cell death. Recently, this class of signalling molecules and receptors has been found to mediate communication between neurons and non-neuronal cells (glia) in the central and peripheral nervous systems. Glia are critical for normal brain function, development and response to injury. Neural impulse activity is detected by glia and purinergic signalling is emerging as a major means of integrating functional activity between neurons, glia and vascular cells in the nervous system. These interactions mediate effects of neural activity on the development of the nervous system and in association with injury, neurodegeneration, myelination and cancer. Glial cells are no longer considered passive bystanders in neuronal brain circuits. Not only are they required for housekeeping and brain metabolism, they are active participants in regulating the physiological function and plasticity of brain circuits and the online control of behavior both in invertebrate and vertebrate model systems. In invertebrates, glial cells are essential for normal function of sensory organs (C. elegans) and necessary for the circadian regulation of locomotor activity (D. melanogaster). In the mamallian brain, astrocytes are implicated in the regulation of cortical brain rhythms and sleep homeostasis. Disruption of AMPA receptor function in a subset of glial cell types in mice shows behavioral deficits. Furthermore, genetic disruption of glial cell function can directly control behavioral output. Regulation of ionic gradients by glia can underlie bistability of neurons and can modulate the fidelity of synaptic transmission. Grafting of human glial progenitor cells in mouse forebrain results in human glial chimeric mice with enhanced plasticity and improved behavioral performance, suggesting that astrocytes have evolved to cope with information processing in more complex brains. Taken together, current evidence is strongly suggestive that glial cells are essential contributors to information processing in the brain. Over the last two decades, the recognition that astrocytes - the predominant type of cortical glial cells - could sense neighboring neuronal activity and release neuroactive agents, has been instrumental in the uncovering of many roles that these cells could play in brain processing and the storage of information. These findings initiated a conceptual revolution that leads to rethinking how brain communication works since they imply that information travels and is processed not just in the neuronal circuitry but in an expanded neuron-glial network. On the other hand the physiological need for astrocyte signaling in brain information processing and the modes of action of these cells in computational tasks remain largely undefined. This is due, to a large extent, both to the lack of conclusive experimental evidence, and to a substantial lack of a theoretical framework to address modeling and characterization of the many possible astrocyte functions. This book that we propose aims at filling this gap, providing the first systematic computational approach to the complex, wide subject of neuron-glia interactions. The organization of the book is unique insofar as it considers a selection of “hot topics” in glia research that ideally brings together both the novelty of the recent experimental findings in the field and the modelling challenge that they bear. A chapter written by experimentalists, possibly in collaboration with theoreticians, will introduce each topic. The aim of this chapter, that we foresee less technical in its style than in conventional reviews, will be to provide a review as clear as possible, of what is “established” and what remains speculative (i.e. the open questions). Each topic will then be presented in its possible different aspects, by 2-3 chapters by theoreticians.
