Diploma Thesis from the year 2005 in the subject Biology - Genetics / Gene Technology, grade: 1,1, TU Bergakademie Freiberg, language: English, abstract: 1. Introduction 1.1 Schizosaccharomyces pombe as a Model System S. pombe functions as a suitable model system since it is easy and inexpensive to rear, has a convenient size, a short life cycle, and is genetically manipulable. As a unicellular eukaryote, the fission yeast S. pombe can exist either in a haploid or diploid state and possesses two different mating types (h+ and h-). The wild type, however, is h90, which means it can switch mating type. Figure 1.01: Left, picture of S. pombe cells At top are two dividing cells in late mitotic phase, showing the fission yeast typical septum at the point of cytoplasmic division. The lower cell is in early M phase, having its chromosomes already segregated. Figure 1.02: Right, fission yeast cell cycle Diagrammatic representation of the S. pombe cell cycles with the interchange between the two occurring in G1 phase (Figure obtained and used with permission from Trevor Pemberton, University of Sussex). [...] S. pombe can undergo two different life cycles, either the vegetative (mitotic) cycle or the sporulation (meiotic) cycle, depending on the environment it is living in. These two cycles are shown in figure 2 with the change between the two occurring in cells at the G1 stage of the mitotic cycle. Under laboratory conditions, given all nutrients required, S. pombe prefers the haploid state. This makes it a favorable organism for genetic research since it ensures that introduced mutations are not masked by another wild type allele. [...]

Every cell has developed mechanisms to respond to changes in its environment and to adapt its growth and metabolism to unfavorable conditions. The unicellular eukaryote yeast has long proven as a particularly useful model system for the analysis of cellular stress responses, and the completion of the yeast genome sequence has only added to its power This volume comprehensively reviews both the basic features of the yeast genral stress response and the specific adapations to different stress types (nutrient depletion, osmotic and heat shock as well as salt and oxidative stress). It includes the latest findings in the field and discusses the implications for the analysis of stress response mechanisms in higher eukaryotes as well.

The fungal kingdom consists of a wide variety of organisms with a diverse range of forms and functions. Fungi have been utilized for thousands of years and their importance in agriculture, medicine, food production and the environmental sciences is well known. New advances in genomic and metabolomic technologies have allowed further developments in the use of fungi in industry and medicine, increasing the need for a compilation of new applications, developments and technologies across the mycological field. Applied Mycology brings together a range of contributions, highlighting the diverse nature of current research. Chapters include discussions of fungal associations in the environment, agriculture and forestry, long established and novel applications of fungi in fermentation, the use of fungi in the pharmaceutical industry, the growing recognition of fungal infections, current interests in the use fungal enzymes in biotechnology and the new and emerging field of myconanotechnology. Demonstrating the broad coverage and importance of mycological research, this book will be of interest to researchers and students in all biological sciences.

Biochemistry And Molecular Biology written by Dirk Hoffmeister and has been published by Springer this book supported file pdf, txt, epub, kindle and other format this book has been release on 2016-02-19 with Science categories.

This new edition provides a comprehensive look at the molecular genetics and biochemical basis of fungal biology, covering important model organisms such as Aspergilli while also integrating advances made with zygomycetes and basidiomycetes. This book groups a total of 15 chapters authored by expert scholars in their respective fields into four sections. Five chapters cover various aspects of gene expression regulation. These range from regulation in organismal interactions between parasitic fungi and their host plant, heavy metal stress and global control of natural product genes to conidiation and regulation through RNA interference. Two chapters are dedicated to signal transduction, highlighting MAP-kinase-dependent signaling and heterotrimeric G-proteins. Fungal carbohydrates are the subject of the third section, which addresses both polymeric cell wall carbohydrates and trehalose as an important, low molecular weight carbohydrate. The fourth section emphasizes the metabolism of major elements (carbon, nitrogen, sulfur) and critical cellular pathways for primary and secondary products.

Fission Yeast written by Iain Hagan and has been published by this book supported file pdf, txt, epub, kindle and other format this book has been release on 2016 with Schizosaccharomyces pombe categories.

Fission yeast are unicellular, rod-shaped fungi that divide by medial fission. Studies using fission yeast were instrumental in identifying fundamental mechanisms that govern cell division, differentiation, and epigenetics, to name but a few. Their rapid growth rate, genetic malleability, and similarities to more complex eukaryotes continue to make them excellent subjects for many biochemical, molecular, and cell biological studies. This laboratory manual provides an authoritative collection of core experimental procedures that underpin modern fission yeast research. The contributors describe basic methods for culturing and genetically manipulating fission yeast, synchronization strategies for probing the cell cycle, technologies for assessing proteins, metabolites, and cell wall constituents, imaging methods to visualize subcellular structures and dynamics, and protocols for investigating chromatin and nucleic acid metabolism. Modifications to techniques commonly used in related species (e.g., budding yeast) are noted, as are useful resources for fission yeast researchers, including various databases and repositories. The well-studied fission yeast Schizosaccharomyces pombe is the focus throughout, but the emerging model S. japonicus-a larger, dimorphic species with several desirable characteristics-is also covered. This manual is an important reference for existing fission yeast laboratories and will serve as an essential start-up guide for those working with fission yeast for the first time.

Since the publication of the best-selling first edition, much has been discovered about Saccharomyces cerevisiae, the single-celled fungus commonly known as baker's yeast or brewer's yeast that is the basis for much of our understanding of the molecular and cellular biology of eukaryotes. This wealth of new research data demands our attention and r
