Hormones and Growth Factors in Development and Neoplasia Edited by Robert B. Dickson, Lombardi Cancer Research Center, Georgetown University David S. Salomon, Laboratory of Tumor Immunology and Biology, National Cancer Institute, NIH The study of hormones is critical to our understanding of developmental aberrations leading to cancer, and the discovery of polypeptide growth factors has led to profound insights into the duality of control of development and cancer by hormones at the tissue and cellular levels. In this book, leading researchers in the field present a cohesive overview of several important growth factor systems and how they interact with endocrine hormones in the context of tissue-tissue interactions; control of cellular growth, differentiation, and death; and reciprocal control of receptors and ligands at the molecular level. In its first two sections, Hormones and Growth Factors in Development and Neoplasia introduces important growth factors and hormonal systems in invertebrate and amphibian model systems, highlights early evolutionary and developmental functions for the classes of molecules later shown to be important in human cancer, establishes the roles of growth factors and hormones in mammalian development, and focuses on early embryonic events and later events leading to sexual dimorphism. The book's third section discusses in detail the control of postnatal developmental processes in male and female reproductive tracts, focusing on the prostate and mammary glands as well as the female reproductive tract, all of which are of special importance in hormonally driven cancers. Finally, the book takes a direct look at cancers and the molecular mechanisms of hormone-growth factor interactions. Among the many topics covered in this timely volume are: * Ecdysone in development of Drosophila * The progesterone receptor in mammalian development * Regulation of cell survival and apoptosis in the reproductive tracts and the breast * Hormones, growth factors, oncogenes, and prostate cancer With its novel approach, authoritative coverage, and broad scope, Hormones and Growth Factors in Development and Neoplasia is informative and relevant for researchers across a spectrum of disciplines, including cancer research, endocrinology, developmental biology, and cell biology.

Hormonal carcinogenesis is an important and controversial area of current research. In addition to accelerating existing cancers, can hormones play the role of primary carcinogens? How do genetic factors influence hormone-related cancer risk? Hormones, Genes, and Cancer addresses these questions. Over the past few decades, cancer research has focused on external environmental causes(e.g., tobacco smoke, viruses, asbestos). With the advent of new genetic sequencing techniques, we are just now beginning to understand how the body's internal environment(i.e., the hormones and growth factors that determine normal development) influences cancer etiology and prevention. From molecular insights to clinical analyses, this volume provides state-of-the-art information on the complex interactions between hormones and genes and cancer. The epidemiology and molecular endocrinology of prostate, breast, uterine, ovarian and testicular cancer are detailed in this timely treatise.

Protooncogenes and Growth Factors in Steroid Hormone Induced Growth and Differentiation reviews current information regarding the complex nature of hormone-induced cell growth and differentiation. The contributors examine the emerging consensus that protooncogenes and growth factors mediate perhaps the most crucial steps leading to cell growth and differentiation. The primary objective of this book is to unite the status of current research related to protooncogenes and growth factors from diverse physiological systems to help readers gain a comprehensive understanding of the subject. Leading researchers have contributed outstanding chapters pertaining to steroid hormone-regulated cell growth and differentiation in normal and/or neoplastic tissues. This book will appeal to basic science researchers, clinicians, industrial researchers, and graduate students.

Growth factors are elaborated to control the growth of cells in such physiological processes as wound healing, tissue regeneration and the immune response. Abnormal production of these growth factors, their receptors or intracellular med!ators of their action may lead to disease states including oncogenesis. This volume will focus on exciting developments in defining the precise molecular lesions that permit the conversion of controlled proliferative signals to neoplasia, on the possible involvement of growth factors in the development of blood vessel diseases as seen in diabetes and atherosclerosis, on the altered immune surveillance that leads to autoimmunity and on the fundamental mechanisms by which growth factors signal their target cells. We expect that the contents of this volume will help promote understanding of the role of these fundamental biological processes and their alterations in a wide variety of disease states and stimulate new investi gations in this important area of biomedical research. The critical role of growth factors in normal as well as abnormal cellular growth and function has become overwhelmingly apparent in recent years. The significant advances in reproductive biology within this rapidly expanding field were highlighted at the Serono Symposia, USA, symposium entitled Growth Factors in Reproduc tion. The conference focused on growth factors as polyfunctional regulators of growth and development in the reproductive system. The program was organized into five areas: (1) growth factors as polyfunctional regulators of growth and development, (2) growth factors and gonadal function, (3) regulation of normal and neoplastic mammary growth, (4) regulatory peptides in reproductive tract development and function, and (5) embryo-maternal signaling. This volume constitutes the contributions ofthe invited symposium speakers and is organized into sections representing the five topic areas of the sessions. Drs. Eli Y. Adashi, Robert B. Dickson, David C. Lee, James Hammond, John A. McLachlan, and R. Michael Roberts, who served as chairmen ofthese sessions, not only provided invaluable advice and assistance in organizing the symposium, but contributed significantly to its content. The enthusiasm and esprit de corps of the poster presenters and discussion participants also helped make this symposium succeed as the first of its kind. Finally, special thanks are due Drs. James Posillico and Lisa Kern ofSerono Symposia, USA, for their guidance and support in the conception and planning of the symposium and in the publication of this volume.

Recent years have seen a considerable emphasis on growth factors and the elucidation of their mode of function, which has led to the recognition that growth factors, their receptors as well as downstream elements of signalling associated with their function might be potential targets in therapeutic management of human diseases. Humanised monoclonal antibodies raised against growth factor receptors have proved to be valuable for targeted cancer treatment and in patient management. This book reviews the latest developments providing insights into the signalling processes involved in morphogenesis and pathogenesis with emphasis on using the elements of the signalling cascades as targets for therapeutic deployment. Provides a fundamental understanding of the basic functions of growth factors and their receptors, describing how they are linked in biological processes Aids the development of therapeutic treatments for cancer Focuses on the interrelationships and convergence of growth factors and their receptors in development and pathogenesis and encourages greater cooperation and integration in the areas of developmental, cancer and cancer therapeutic research
