This book is about the escape strategies used by cancer cells to avoid the immune response of the host. The main characters of this story are the “Antigen Presenting Molecules” and the “T Lymphocytes”. The former are known as the Major Histocompatibility Complex (MHC): the H-2 and the HLA molecules. The latter are a subgroup of white cells travelling all over our body which are capable to distinguish between “self and non self”. Readers will know from the inside about the history of the HLA genetic system and will discover how T lymphocytes recognize and destroy cancer cells. One of the key important questions is: Why tumors arise, develop and metastasize? This book tries to answer this question and will explain how cancer cells become invisible to killer T lymphocytes. The induction of immune responses against tumor cells by vaccination is rapidly evolving as a therapeutic modality with new potentials for the treatment of cancer. It is based on the fact that our immune system can identify tumor cells and, once activated, is capable of developing specific immunity against the neoplastic cells. Numerous observations and intense research clearly document the major contribution of the immune system to the prevention of cancer. And there are many re ports of patients suffering from malignant melanoma or other tumors who mount a spontaneous immune response against their tumor cells that results in tumor regression. Based on the recent advances in our understanding of the compo nents of our immune system, their interactions and the regulation of immune responses, we are now able to design vaccination strategies that induce or enhance cell-mediated immunity against tumors. A ma jor advancement came with the identification and characterization of relevant tumor antigens, which are suitable target structures for anti-tu mor immune response. First clinical trials using such vaccine strategies have yielded encouraging results in patients. However, in spite of many reported cases of successful therapy of cancer by vaccination many patients still do not experience relief after such treatments. These initial clinical trials and the accompanying investigations have revealed a number of important results that indicate the direction of future re search and development in the field.

Immunotherapy began in 1774 when the Dorset farmer Benjamin Jesty inoculated his wife and two sons with the pus from the teat of a cow suffering from cow pox, using his wife's knitting needle as a vaccinating implement. It has made slow progress. Meanwhile the science of Immunology has burgeoned so much that if all immunologists read every page of the Journal of Immunology, let alone the other Immunology journals, then they would have no time left to write for it. I am pleased that some of them have found the time to write for this volume. In spite of the rapid expansion in immuno logical knowledge and the undreamt of complexity of the immune system that has been unravelled, immunologists have remained until recently erudite but therapeutically effete. Indeed anyone purporting to treat disease by immuno logical methods has been in danger of being labelled a quack or a crackpot. Happily things are changing. The nine chapters of this volume detail nine quite different approaches to manipulating the immune system for therapeutic benefit. All are experimental and they have been attended with greater or lesser degrees of success. In some cases their main effect has been to elucidate the complexity of the problem. On the other hand, there are people alive and well today as a result of these approaches who would otherwise have perished. Immunotherapy is here to stay and it can only get better.

Since the advent of hybridoma technology more than two decades ago, numerous antibodies have entered the clinical setting as potent therapeutic agents. Their repeated application in humans, however, is limited by the development of human antimouse antibodies (HAMA) in the recipient, leading to allergic re- tions against the foreign murine protein and rapid neutralization. To circumvent these limitations many new antibodies have recently been tailored through recombinant antibody technology. The initial clinical data show encouraging results, thus demonstrating the potential of these new therapeutic agents. The purpose of Recombinant Antibodies for Cancer Therapy is to present a collection of detailed protocols in recombinant antibody technology. It is pri- rily addressed to scientists working on recombinant antibodies as well as cli- cians involved with antibody-based therapies. As with other volumes of this series, we placed the main focus on providing detailed protocols describing procedures step-by-step. Moreover, each protocol supplies a troubleshooting guide containing detailed information on possible problems and hints for pot- tial solutions. Antibody technology is a subject of constant and rapid change. This volume, therefore, does not attempt to cover all possible current experimental approaches in the field. Rather, we present carefully selected protocols, written by competent authors who have successfully verified the particular method described. Given our own professional backgrounds and interest in oncology, we chose to conc- trate chiefly on therapeutic agents for cancer patients.
