This volume focuses on our current understanding of the molecular underpinnings of prostate cancer and their potential application for precision medicine approaches. The emergence and applications of new technologies has allowed for a rapid expansion of our understanding of the molecular basis of prostate cancer and has revealed a remarkable genetic heterogeneity that may underlie the clinically variable behavior of the disease. The book consists of five sections which provide insight about the following: (1) General principles; (2) Molecular signatures of primary prostate cancer; (3) Molecular signatures of advanced prostate cancer; (4) Key molecular pathways in prostate cancer development and progression; (5) and Precision medicine approach: Diagnosis, treatment, prognosis. Precision Molecular Pathology of Prostate Cancer is an important resource for the practicing oncologist, urologist, and pathologist, and will also be useful for researchers in the prostate cancer community.

Prostate cancer continues to be a major threat to men's health, thesecond most common cancer in men, in many countries. Therefore itis increasingly important that those faced with difficult clinicalquestions make the right decisions. Since the first volume waspublished in 2000, many debates persist but new controversies havealso emerged, reflected in the chapters of this new volume. Newauthors have been recruited to give their insight into newer areasof this controversial topic. Challenges in prostate cancer provides a series ofstate-of-the-art review articles, each of which addresses andanswers a contentious question. It includes chapters on medical andsurgical management of all stages of prostate cancer, clinicalinvestigation, epidemiology, clinical trials and basic scientificresearch and covers issues such as the genetic basis of cancer, clinical economics and influence. The authors have used their expertise to distil the availableevidence into practical advice to be used by busy urologists intheir day-to-day practice. And as well as being an overview ofcurrent practice many chapters attempt to give insights into thefuture direction of prostate cancer research and clinicalmanagement.

Signaling Pathways In Cancer Pathogenesis And Therapy written by David A. Frank and has been published by Springer Science & Business Media this book supported file pdf, txt, epub, kindle and other format this book has been release on 2011-11-12 with Medical categories.

In recent years, increasing evidence has suggested that abnormal activation of signaling pathways is a critical event in cancer pathogenesis. In particular, activation of these pathways can lead to inappropriate cellular survival, proliferation, pluripotency, invasion, metastasis, and angiogenesis. Thus, understanding the mechanisms by which signaling pathways become subverted in a cancer cell can provide insight into critical events in cancer pathogenesis. Furthermore, as our ability to target specific molecular interactions advances, we now have the ability to design small molecules, protein therapeutics, and other forms of targeted therapies. By focusing on the specific molecular abnormalities in a cancer cell, these agents hold the potential to be much more effective and much less toxic than current cytotoxic therapies.

The Dean and Betty Gallo Prostate Cancer Center (GPCC) was established with the goal of eradicating prostate cancer and improving the lives of men at risk for the disease through research, treatment, education and prevention. GPCC was founded in the memory of Dean Gallo, a beloved New Jersey Congressman who died tragically of prostate cancer diagnosed at an advanced stage. GPCC unites a team of outstanding researchers and clinicians who are committed to high-quality basic research, translation of innovative research to the clinic, exceptional patient care, and improving public education and awareness of prostate cancer. GPCC is a center of excellence of The Cancer Institute of New Jersey, which is the only NCI-designated comprehensive cancer center in the state. GPCC efforts are now integrated well as part of our Prostate Program at CINJ, in which Dr. Robert DiPaola and Dr. Cory Abate-Shen are co-leaders. The Prostate Program unites 19 investigators from 10 academic departments who have broad and complementary expertise in prostate cancer research. The overall goal and unifying theme is to elucidate basic mechanisms of prostate growth and oncogenesis, with the ultimate goal of promoting new and effective strategies for the eradication of prostate cancer. Members' wide range of research interests collectively optimize the chances of providing new insights into normal prostate biology and unraveling the molecular pathophysiology of prostate cancer. Cell culture and powerful animal models developed by program members recapitulate the various stages of prostate cancer progression, including prostatic intraepithelial neoplasia, adenocarcinoma, androgen-independence, invasion and metastases. These models promise to further strengthen an already robust program of investigator-initiated therapeutic clinical trials, including studies adopted by national cooperative groups. Efforts to translate laboratory results into clinical studies of early detection and chemoprevention are underway. The specific goals of this program are: (1) To investigate the molecular mechanisms underlying normal prostate growth and differentiation and elucidate the molecular mechanisms underlying prostate oncogenesis. (2) To build on fundamental knowledge to develop effective therapeutic approaches for the treatment of prostate cancer. (3) To improve the control of prostate cancer through early detection, chemoprevention, and outreach and education. This new disease-based program is structured to improve interdisciplinary interactions and translational results. Already, through the dynamic leadership of Drs. Cory Abate-Shen and Robert DiPaola, new investigators were attracted to the field, new collaborations engendered, and numerous investigator-initiated trials implemented. Progress in GPCC and the program overall has been outstanding. The Center has success in uniting investigators with broad and complementary expertise in prostate cancer research. The overall goal and unifying theme is to elucidate basic mechanisms of prostate growth and oncogenesis, with the ultimate goal of promoting new and effective strategies for the eradication of prostate cancer in patients and populations at risk. Members wide range of research interests collectively optimize the chances of providing new insights into normal prostate biology and unraveling the molecular pathophysiology of prostate cancer. Pharmacogenomic profiling of cancer has recently seen much activity with the accessibility of the newest generation of high-throughput platforms & technologies. Myriad mechanistic studies have been devoted to identifying dietary phytochemicals from fruit or vegetables that can help prevent cancer, with evidence suggesting an inverse correlation between intake & risk of certain cancers. To develop a comprehensive understanding of cancer pathogenesis, & potential for chemopreventive intervention with dietary factors, an integrated approach that encompasses both pharmacogenomic & mechanistic aspects is desirable. This work discusses the identification of germane molecular targets by expression profiling using dietary factors, synergistic effects in combinatorial use of dietary factors in colon cancer, regulatory studies in prostate cancer, concerted modulation of key players in inflammation & carcinogenesis, & delineation of prostate cancer biological networks. This will greatly enhance our understanding of the transcriptional circuitry in cancer & important master regulators in the cell that may potentially be exploited for preventive intervention using dietary phytomedicines.
