Eukaryotic cells are remarkably complex structures, containing a vast repertoire of macromolecules, organelles, and other compartments that orchestrate the tasks required for life. For in-depth studies of their function and composition, reliable methods for the isolation of specific subcellular structures are often required. This laboratory manual provides step-by-step protocols for the extraction of subcellular components from animal tissues, yeasts, plants, and cultured cells. Each chapter focuses on a particular eukaryotic organelle, vesicle, membrane, or macromolecular complex. Strategies for breaking cells while maintaining the structural and functional integrity of the component of interest, enriching for that component based on its physical and biochemical characteristics, and monitoring and ensuring the success of the purification procedure are provided. The contributors describe both traditional approaches (e.g., density gradient centrifugation) and innovative techniques (e.g., the use of SPIONs) for isolating subcellular constituents. This manual is therefore an essential laboratory resource for all cell biologists seeking a comprehensive collection of dependable subcellular fractionation methods.

Ribonucleic acids are central to cellular and molecular processes and perform vital functions in both structural and functional roles. RNA molecules form the bridge between the stable genetic information contained within DNA and enzymes and proteins that carry out much of the metabolism within the cell. Many of the sites of protein synthesis, the ribosomes within the cell, are composed of these ribonucleic acids as are the tRNA molecules that deliver the amino acid building blocks to the ribosomes. Of all the RNA species, the nucleic acid intermediate, messenger RNA, is a desirable source of material to biologists, since this reflects much of, what ultimately, is translated into enzymes and proteins. In order to determine the qualitative and quantitative changes in mRNA expression, a vast number of molecular biological techniques have been developed. Key molecular methods that provide the means to initially isolate and analyze RNA molecules are the focus of this volume. In putting together this collection of protocols, we have tried to provide techniques that are most applicable and widely used. In particular, there are a number of iso- tion techniques included that have been developed, modified, or adapted to enable extraction from a variety of cell types, organisms, or subcellular organelles. Successful isolation of intact RNA is an essential starting point for any sub- quent analysis. This is why we have aimed to make this section comprehensive. The analysis of RNA is the focus of the following chapters.

Whilst significant advances have been made in whole organismal proteomics approaches, many researchers still rely on combinations of tissue selection and subcellular prefractionation methods to reduce the complexity of protein extracts from plants prior to proteomic analysis. Often this will allow identification of many lower abundance proteins of the target proteome and it may involve the selection of specific organs, cell types or the isolation of specific subcellular components. These subcellular proteomes provide insight into functions following various treatments and also contribute to the wider understanding of the entire organismal proteome by cataloguing a series of sub-proteome contents. The aim of this Research Topic is to bring together knowledge of sub cellular components in different plant species to provide a basis for accelerated research. It aims to provide a mini-review for each proposed section that summarizes the current understanding of a particular proteome, with the anticipation that every 5 - 10 years we can update these definitive publications.

Modern Methods of Plant Analysis When the handbook Modern Methods of Plant Analysis was first introduced in 1954 the considerations were 1. the dependence of scientific progress in biology on the improvement of existing and the introduction of new methods; 2. the inavailability of many new analytical methods concealed in specialized journals not normally accessible to experimental plant biologists; 3. the fact that in the methods sections of papers the description of methods is frequently so compact, or even sometimes so incomplete, that experiments are difficult to reproduce. These considerations still stand today. The series was highly successful, seven volumes appearing between 1956 and 1964. Since today there is still a demand for the old series, the publisher has decided to resume publication of Modern Methods of Plant Analysis. It is hoped that the New Series will be as acceptable to those working in plant sciences and related fields as the early volumes undoubtedly were. It is difficult to single out the major reasons for success of any publication, but we believe that the methods published in the first series were up-to-date at the time and the descriptions as applied to plant material so complete in themselves that there was little need to consult other publications.

In the post-genomic era, several plant species have been sequenced and massive genomic information is now available which contributed to expand the development of novel technical strategies for the study of additional levels of biological information of plant species. This book focuses on the “omics” approaches together with systems analysis of several different plant species, which have revealed very interesting variations on the cellular responses at the protein, transcript and metabolite levels in response to changes environmental conditions. The volume covers recent technological advances in the area of “omics” and synthesizes recent findings of the field of plant “omics” and systems biology together along with techniques that can be applied for such studies.
