"Oxidative stress" is used as the generic term describing the involve ment of reactive oxygen species in various human diseases. The scope of such a topic is becoming increasingly wide. The recent interest in radicals such as nitric oxide and the discovery of new mechanisms such as the effect of free radicals on redox sensitive proteins and genes are enlarging our understanding of the physiological role of free radicals. Oxidative stress is involved in numerous pathological. processes such as ageing, respiratory or cardiovascular diseases, cancer, neurological pathologies such as dementia or Parkinson's disease. It still remains difficult, however, to demonstrate by chemical measurement the in vivo production of free radicals and even more to realise their speciation. Therefore, the development of new tools and indicators is engrossing many researchers working in this field. Reliable indicators are abso lutely necessary not only to monitor the evolution of oxidative stress in patients but also to evaluate the efficiency of new antioxidant treat ments. The French Free radical club of Grenoble, the CERLIB has been involved for many years in the organisation of international training programs on methodology, in order to provide both theoretical and practical help to researchers from various countries. Such training sessions have been highly successful and participants value the oppor tunity to learn reliable techniques. The material presented in this book deals with basic mechanisms of free radical reactions in autoxidation processes and anitoxidant suppression of autoxidation of foods, biochemical models and biologi cal systems. Autoxidation in foods and corresponding biological effects are usually approached separately although recent mechanistic developments in the biochemistry and free radical chemistry of per oxides and their precursors tend to bring these two fields closer. Apparent ability of antioxidants in diets to reduce the inci dence of cancer has resulted in scrutiny of autoxidized products and their precursors as possibly toxic, mutagenic and carcinogenic agents. Mechanisms of any of these effects have been barely ad dressed. Yet we know now that free radicals, as esoteric as they were only a few decades ago, are being discovered in foods, biochem ical and biological systems and do play a role in the above-mentioned causalities. The purpose of the Workshop and the resulting book was to give a unifying approach towards study of beneficial and deleterious effects of autoxidation, based on rigorous scientific considerations. It is our hope that the material presented in this book will not only provide a review of the "state of the art" of autoxidation and anti oxidants, but also reflect the interaction which occurred during the Workshop between workers using model sytems, and food and biological systems.

Many studies have shown that free radical damage and lipid peroxidation increase as a function of the degree of unsaturation of the fatty acids present in the phospholipids of biological membranes. Membrane phospholipids are particularly susceptible to oxidation not only because of their highly polyunsaturated fatty acid content but also because of their association in the cell membrane with non-enzymatic and enzymatic systems capable of generating pro-oxidative-free radical species. There are two broad outcomes to lipid peroxidation: structural damage to membranes and generation of secondary products. Membrane damage derives from the generation of fragmented fatty acyl chains, lipidlipid cross-links, lipidprotein cross-links and endocyclisation to produce isoprostanes and neuroprostanes. These processes combine to produce changes in the biophysical properties of membranes that can have profound effects on the activity of membrane-bound proteins. The consequence of peroxidation of unsaturated fatty acids is severe: damage of membranes function, enzymatic inactivation, toxic effects on the cellular division, etc. This new book presents and discusses current research on oxidative stress, and lipid oxidation non-inhibited and inhibited; assessment of oxidative balance in the lipo- and hydro-philic cellular environment in biological systems; mass spectrometry detection of protein modification by cross-reaction with lipid peroxidation products; diagonal gel electrophoretic analysis of protein disulfides: principles and applications.
