Anomalous epigenetic patterns touch many areas of study including biomedical, scientific, and industrial. With perspectives from international experts, this resource offers an all-inclusive overview of epigenetics, which bridge DNA information and function by regulating gene expression without modifying the DNA sequence itself. The view “It’s all in our genes and we cannot change it” developed in the past 150 years since Gregor Mendel’s experiments with flowering pea plants. However, there is a special form of genetics, referred to as epigenetics, which does not involve any change of our genes but regulates how and when they are used. In the cell nucleus our genes are packed into chromatin, which is a complex of histone proteins and genomic DNA, representing the molecular basis of epigenetics. Our environment and lifestyle decisions influence the epigenetics of our cells and organs, i.e. epigenetics changes dynamically throughout our whole life. Thus, we have the chance to change our epigenetics in a positive as well as negative way and present the onset of diseases, such a type 2 diabetes or cancer. This textbook provides a molecular explanation how our genome is connected with environmental signals. It outlines that epigenetic programming is a learning process that results in epigenetic memory in each of the cells of our body. The central importance of epigenetics during embryogenesis and cellular differentiation as well as in the process of aging and the risk for the development of cancer are discussed. Moreover, the role of the epigenome as a molecular storage of cellular events not only in the brain but also in metabolic organs and in the immune system is described. The book represents an updated but simplified version of our textbook “Human Epigenomics” (ISBN 978-981-10-7614-8). The first five chapters explain the molecular basis of epigenetics, while the following seven chapters provide examples for the impact of epigenetics in human health and disease.

The ability of a single genome to give rise to hundreds of functionally distinct cell type programs is in itself remarkable. Pioneering studies over the past few decades have demonstrated that this plasticity is retained throughout development, a phenomenon of epigenetic programming and reprogramming that remains one of the most fascinating areas of modern biology, with major relevance to human health and disease. This book presents the basic biology involved, including key mechanistic insights into this rapidly growing field.

This book will provide an overview of basic epigenetic phenomena; interaction between epigenetic and genetic factors; and the influence of epigenetic factors on inheritance. Epigenetic states may contribute to the penetrance of genetic polymorphisms or mutations and thereby modify inheritance patterns. This may result in non‐Mendelian inheritance of genetic traits such as observed in common human disease. The relationship between epigenetics and genetics, however, has not been comprehensively summarized yet. The topic is being more and more appreciated lately due to considerable advances in genomic and epigenomic approaches to study the origins of human disease. The editors will focus not only on describing epigenetic characteristics, mechanisms and results, but also on how considerations of epigenetics can alter interpretation and analysis of risks for complex traits. This book will be a resource for those who have been working in human genetics or analysis of human genetic data and are studying the impact of epigenetics on inheritance. An overview will be given of the impacts of inter‐individual variation in epigenetic states from major changes (errors in genomic imprinting) that cause congenital developmental defects to subtle changes and their impact on complex traits. The editors will discuss the relationship between epigenetic changes and genetic changes in human disease. Several chapters will also focus on statistical analysis of epigenetics effects, either in human disease genetic studies, or in population genetics.

What lies at the heart of neuronal plasticity? Accumulating evidence points to epigenetics. This word originally indicated potentially heritable modifications in gene expression that do not involve changes in DNA sequence. Today this definition is much less strict, and epigenetic control is thought to include DNA methylation, histone modifications, histone variants, microRNA metabolic pathways and non-histone proteins modifications. Thus, while neuronal plasticity is rightly thought to be intimately associated to genomic control, it is critical to appreciate that there is much more to the genome than DNA sequence. Recent years have seen spectacular advances in the field of epigenetics. These have attracted the interest of researchers in many fields and evidence connecting epigenetic regulation to brain functions has been accumulating. Neurons daily convert a variety of external stimuli into rapid or long-lasting changes in gene expression. A variety of studies have centered on the molecular mechanisms implicated in epigenetic control and how these may operate in concert. It will be critical to unravel how specificity is achieved. Importantly, specific modifications seem to mediate both developmental processes and adult brain functions, such as synaptic plasticity and memory. Many aspects of the research in neurosciences and endocrinology during the upcoming decade will be dominated by the deciphering of epigenetic control. This book constitutes a compendium of the most updated views in the field.

This detailed volume focuses on population epigenetics, which is of increasing interest to policy makers searching for explanations for complex epidemiological observations and conceptual models on which to base interventions. With a concentration on DNA methylation, the emphasis is on the most commonly studied epigenetic phenomenon that is most amenable to further study. Written for the highly successful Methods in Molecular Biology series, chapters include introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols, and tips on troubleshooting and avoiding known pitfalls. /divAuthoritative and practical, Population Epigenetics: Methods and Protocols aims to aid population researchers in exploiting the latest insights into epigenetics to improve our understanding of the factors that influence human health and longevity.

During the twentieth century, genes were considered the controlling force of life processes, and the transfer of DNA the definitive explanation for biological heredity. Such views shaped the politics of human heredity: in the eugenic era, controlling heredity meant intervening in the distribution of "good" and "bad" genes. However, since the turn of the twenty-first century, this centrality of genes has been challenged by a number of "postgenomic" disciplines. The rise of epigenetics in particular signals a shift from notions of biological fixedness to ideas of plasticity and "impressionability" of biological material. This book investigates a long history of the beliefs about the plasticity of human biology, starting with ancient medicine, and analyses the biopolitical techniques required to govern such permeability. It looks at the emergence of the modern body of biomedicine as a necessary displacement or possibly reconfiguration of earlier plastic views. Finally, it analyses the returning of plasticity to contemporary postgenomic views and argues that postgenomic plasticity is neither a modernistic plasticity of instrumental management of the body nor a postmodernist celebration of potentialities. It is instead a plasticity that disrupts clear boundaries between openness and determination, individual and community, with important implications for notions of risk, responsibility and intervention.

Epigenetics is one of the most exciting and rapidly developing areas of modern genetics with applications in many disciplines from medicine to agriculture. The most common form of epigenetic modification is DNA methylation, which plays a key role in fundamental developmental processes such as embryogenesis and also in the response of organisms to a wide range of environmental stimuli. Indeed, epigenetics is increasing regarded as one of the major mechanisms used by animals and plants to modulate their genome and its expression to adapt to a wide range of environmental factors. This book brings together a group of experts at the cutting edge of research into DNA methylation and highlights recent advances in methodology and knowledge of underlying mechanisms of this most important of genetic processes. The reader will gain an understanding of the impact, significance and recent advances within the field of epigenetics with a focus on DNA methylation.
