Cilia and Flagella presents protocols accessible to all individuals working with eukaryotic cilia and flagella. These recipes delineate laboratory methods and reagents, as well as critical steps and pitfalls of the procedures. The volume covers the roles of cilia and flagella in cell assembly and motility, the cell cycle, cell-cell recognition and other sensory functions, as well as human diseases and disorders. Students, researchers, professors, and clinicians should find the book's combination of "classic" and innovative techniques essential to the study of cilia and flagella.Key Features* A complete guide containing more than 80 concise technical chapters friendly to both the novice and experienced researcher* Covers protocols for cilia and flagella across systems and species from Chlamydomonas and Euglena to mammals* Both classic and state-of-the-art methods readily adaptable across model systems, and designed to last the test of time, including microscopy, electrophoresis, and PCR* Relevant to clinicians interested in respiratory disease, male infertility, and other syndromes, who need to learn biochemical, molecular, and genetic approaches to studying cilia, flagella, and related structures

The Biology of Cilia and Flagella reviews advances in the study of cilia and flagella since 1928. This book focuses on four main topics—structure of cilia, factors that affect ciliary activity, movement of cilia and flagella, and coordination of beat of cilia. In these topics, this compilation specifically discusses the intracellular structures associated with ciliary bases; viscosity of the medium and ciliary activity, and energy relationships of cilia and flagella. The compounding of synchronously beating cilia, variations in shaft structure, and control of ciliary activity by the organism are also elaborated. This text likewise covers the determination of the rate of beat of cilia and root-fiber systems and coordination of ciliary beat. This publication is suitable for biologists and clinicians of other disciplines researching on the structure and physiology of cilia and flagella.

Over the past two decades experimental studies have solidified the int- pretation of the cytoskeleton as a highly dynamic network of microtubules, actin microfilaments, intermediate filaments, and myosin filaments. Rather than a network of disparate fibers, these polymers are often interconnected and display synergy, which is the combined action of two or more cytoskeletal polymers to achieve a specific cellular structure or function. Cross-commu- cation among cytoskeletal polymers is thought to be achieved through cytoskeletal polymer accessory proteins and molecular motors that bind two or more cytoskeletal polymers. Development of the modern concept of the cytoskeleton is a direct o- growth of advances in experimental tools and reagents that are available to cell and molecular biologists. Technological advances and refinements in cell imaging have made it possible to selectively image a single cytoskeletal po- mer and monitor its dynamics through the use of fluorescence probes in vitro and in vivo. Two decades ago, cytoskeletal research was limited to a few perturbation reagents that included colchicine and cytochalasin. Today, the perturbation arsenal has expanded to a highly selective group of reagents that includes Taxol, nocodazole, benomyl, latrunculin, jasplakinolide, and such endogenous proteins as gelsolin. These reagents enable the investigator to selectively perturb or destroy a cytoskeletal polymer while leaving other cytoskeletal polymers intact. Site-specific monoclonal antibodies that target a specific cytoskeletal polymer have proven to be highly selective affinity tools for cytoskeletal research.

Cilia Model Organisms And Intraflagellar Transport written by and has been published by Academic Press this book supported file pdf, txt, epub, kindle and other format this book has been release on 2009-12-01 with Science categories.

Cilia are highly conserved organelles that serve motile functions, sensory functions, or both. These organelles power cell movement, generate fluid flow in various organs, act as sensors of the extracellular environment and have been modified for various specialized tasks such as light reception and smell. Defects in these ubiquitous organelles lead to a broad array of human genetic disorders that range from polycystic kidney disease, retinal degeneration, epilepsy and infertility to developmental defects such as situs inversus and polydactyly. This volume is the third in a three-part series on cilia that focuses on the use of model organisms to gain insight into ciliary function and on the process of intraflagellar transport that is essential for the assembly and maintenance of ciliary structures. - Includes both classic and state-of-the-art methods readily adaptable across model systems, and designed to last the test of time - Covers forward and reverse genetic analysis of IFT and biochemical methods to define the role of IFT components - Methods presented cover molecular, genetic, and biochemical approaches to ciliary function in model organisms
