This invaluable book provides comprehensive coverage of contemporary serological, cellular and molecular methodologies in histocompatibility testing, and their application to human organ transplantation and transfusion. The contributors are internationally respected authorities in histocompatibility and immunogenetics, and are closely involved in the development or application of state-of-the-art technologies. The first three sections of the book are primarily intended for use as a bench manual for histocompatibility testers, immunologists and immunogeneticists; the fourth and fifth sections, on selection of donors and statistical methods, will further assist medical practitioners involved in clinical transplantation and its outcome. The final section of the book reviews the genetics and clinical relevance of minor histocompatibility antigens. Minor histocompatability antigens (MHA) are a group of cell-surface membrane proteins that are able to exclusively mediate a cellular immune response. These antigens (Ag) were initially distinguished from major histocompatibility antigens (MHC) by their weak immunogenicity in allograft transplants. As information describing their structure and function accumulated, the term MHA was misleading because these immunogens were neither weak nor minor. It became clear that non-MHC histocompatible antigens (non-MHC-HA) was a more appropriate name. Non-MHC HA are involved not only in transplantation but also in defining immunologic self and shaping the T cell reprtoire. This book investigates the biology of the non-MHC HA and their role in clinical medicine.

Immune Biology of Allogeneic Hematopoietic Stem Cell Transplantation: Models in Discovery and Translation, Second Edition once again provides clinical and scientific researchers with a deep understanding of the current research in this field and the implications for translational practice. By providing an overview of the immune biology of HSCT, an explanation of immune rejection, and detail on antigens and their role in HSCT success, this book embraces biologists and clinicians who need a broad view of the deeply complex processes involved. It then moves on to discuss the immunobiology mechanisms that influence graft-versus-host disease (GVHD), graft-versus-leukemia effect, and transplantation success. Using illustrative figures, highlighting key issues, describing recent successes, and discussing unanswered questions, this book sums up the current state of HSCT to enhance the prospects for the future. The second edition is fully revised and includes new chapters on microbiome, metabolism, kinase targets, micro-RNA and mRNA regulatory mechanisms, signaling pathways in GVHD, innate lymphoid system development, recovery and function in GVHD, genetically engineered T-cell therapies, immune system engagers for GVHD and graft-versus-tumor, and hematopoietic cell transplant for tolerance induction in solid organ grafts. Brings together perspectives from leading laboratories and clinical research groups to highlight advances from bench to the bedside Guides readers through the caveats that must be considered when drawing conclusions from studies with animal models before correlating to clinical allogeneic hematopoietic stem cell transplantation (HSCT) scenarios Categorizes the published advances in various aspects of immune biology of allogeneic HSCT to illustrate opportunities for clinical applications

A comprehensive guide to the HLA (Human Leukocyte Antigen) system for immunologists and clinicians, this book contains up-to-date information on the MHC (Major Histocompatibility Complex) and its role in the immune response and in various diseases. The book explores the biological significance and role of the HLA system in organ and haematopoietic stem cell transplantation management. This volume is an invaluable guide to the full spectrum of HLA-related science while also serving as a conceptual and technical resource for those involved in HLA-related research and in clinical or surgical practice. In addition, it will be a primary point of contact for individuals working in other areas who suddenly find that their research is drawing them into the complexities of HLA genetics.
