Cell Biology of the Major Histocompatibility Complex documents the proceedings of a symposium on ""Cell Biology of the Major Histocompatibility Complex"" held at Arden House on the Harriman Campus of Columbia University from June 8 -10, 1984. The meeting was the ninth of the P & S Biomedical Sciences Symposia. The book is organized into five parts. Part I on the structure of MHC molecules includes papers on human histocompatibility antigens; the cloning of human MHC; and organization of the genes of the H-2 complex. Part II on alternate forms of MHC molecules includes studies on the expression of a secreted form of the MHC class I antigen and alternative splicing in the H-2 multigene family. Part III deals with MHC biosynthesis, intracellular transport, and membrane expression. It includes studies on the manipulation of glycans on antigens of the MHC and dynamics of MHC molecules in lymphoid cells. Part IV on homologues of class I MHC molecules covers topics such as Tla expression in normal and malignant cells and the Qa series of antigens. Part V on the association of MHC with antigens includes studies such as histocompatibility molecules as immune response gene products and molecular nature of T-cell recognition of antigen.

The Major Histocompatibility Complex (MHC) was discovered as a con sequence of the chronic problem encountered by cancer biologists in the early years of this century: the failure to maintain tumor lines by serial passage in outbred mice. A number of observations pointed to genetic similarity being a prerequisite for successful transplantation and they were incorporated into a genetic theory of transplantation by C.C. Little. This prompted scientists like Little to initiate breeding experiments designed to test his hypothesis and produce genetically identical mice which would permit the growth of trans planted tumors. Most inbred strains of mice commonly used in immunology derive from those efforts. Transplantation of normal tissues obeyed the same rules found for malignant tissues and rejection was shown to be an immunological phenomenon. G.D. Snell showed that a single genetic locus determined rapid rejection of skin grafts. This was initially called the Major Histocompatibility Locus but was subsequently shown to include many functionally related genes and renamed the Major Histocompatibility Complex (MHC). Histocampatibility covers the genetic, immunologic, and the chemical studies on allograft rejection process. The major histocompatibility complex (MHC) is a highly polymorphic and diverse multigene locus in all jawed vertebrate species that has an integral role in adaptive/innate immune systems, transplantation, and infectious and autoimmune diseases. The MHC supra-locus in mammalian vertebrates is usually partitioned into three distinct regions, known as classes I, II, and III, which, to varying extents, can be found conserved in nonmammalian jawed vertebrates, such as bony fish, amphibians, and bird lineages. The MHC gene region is characterized particularly by the expression of class I and class II glycoproteins that bind peptides derived from intracellular or extracellular antigens to circulating T-cells. While this expressed antigenic specificity remains the predominant interest with respect to MHC function and polymorphism in a population, a broader concept has emerged that examines the MHC as a multifunctional polymorphic controller that facilitates and regulates genome diversity with a much greater array of functions and effects than just MHC-restricted antigen recognition. This volume of 19 reprints presented by various experts and collected from the Special Issue of Cells on “MHC in Health and Disease” covers a broad range of topics on the genomic diversity of the MHC regulatory system in various vertebrate species, including MHC class I, II, and III genes; innate and adaptive immunity; neurology; transplantation; haplotypes; infectious and autoimmune diseases; fecundity; conservation; allelic lineages; and evolution. Every biological system is the outcome of evolution and has a history all its own. This history dictates how the system works and why it has certain properties and not others. This is why we need to study not only the structure and function, but also the history of the system. This argument undoubtedly applies to the study of the immune system and also to the study of the major histocompatibility complex (MHC). Since 1989, researchers of various scientific disciplines who share a deep inter est in MHC evolution have held a meeting every two years to discuss their latest research developments, exchange ideas, and foster friendship. Together with my colleagues Drs. Naoyuki Takahata and Yoko Satta, I organized the Sixth Interna tional Workshop on MHC Evolution in Hayama, Japan, May 25-29, 1999. This volume is the proceedings of that conference. It covers diverse topics pertinent to MHC evolution, including the origin of the adaptive immune system, the organi zation of the MHC in humans and other model vertebrates, MHC-parasite co evolution, and the nature and origin of MHC polymorphism. I hope that this book will be of interest not only for MHC researchers and immunologists, but also for other specialists who are interested in the evolution of biological systems in gen eral.
